Influence of survivin and caspase-3 on cell apoptosis and prognosis in gastric carcinoma

AIM: To evaluate the role of survivin and caspase-3 in apoptosis of gastric carcinoma, as well as in prognosis of patients with gastric ***: Expressions of survivin and caspase-3 were investigated immunohistochemically in 80 gastric carcinoma patients without a history of chemo-radiation therapy. Tumor cell apoptosis was examined by TUNEL ***: Immunohistochemical analysis showed that survivin expression was positive in 61 of 80 patients (76%) with gastric carcinoma. In contrast, no expression of survivin in adjacent normal tissues was detected. Expression level of caspase-3 was higher in normal tissues than in *** with higher expression of survivin had worse histological grades and pathological stages. Expression of caspase-3 was significantly associated with histological stages, but not with the pathological stages. Although survivin expression in carcinoma was not inversely related to caspase-3, patients with survivin (-) and caspase-3(+) had the maximum apoptosis ***: Expression level of survivin was associated with histological grades and pathological stages of the tumor,indicating that survivin may be a poor prognosis factor for gastric carcinoma. Unlike caspase-3, survivin (an apoptosis inhibitor) can markedly inhibit the apoptosis of tumor cells.

Significance of effector protease receptor-1 expression and its relationship with proliferation and apoptotic index in patients with primary advanced gastric adenocarcinoma

Abstract: The Dai medicinal plant Winchia calophylla A. DC. (Apocynaceae) has efficacy as an anti-cough and anti-asthmatic medication. In order to investigate its relative bioactive components, we studied the chemical constituents of this plant. Using repeated column chromatography, 28 compounds, including loganin, six phenolic compounds, 17 indole alkaloids, three pyridine alkaloids, and a quinoline alkaloid, were isolated from the stem barks of W. calophylla. Loganin, paeonol, N (4)-methyl akuammicine, and cantleyine exhibited a moderate relaxation effect on isolated smooth muscles of guinea-pig tracheal spirals and lung strips and may be the bioactive components responsible for the bronchodilation produced by W. calophylla.

DeepNoise: Signal and Noise Disentanglement Based on Classifying Fluorescent Microscopy Images via Deep Learning

The high-content image-based assay is commonly leveraged for identifying the phenotypic impact of genetic perturbations in biology ***,a persistent issue remains unsolved during experiments:the interferential technical noises caused by systematic errors(e.g.,temperature,reagent concentration,and well location)are always mixed up with the real biological signals,leading to misinterpretation of any conclusion ***,we reported a mean teacher-based deep learning model(Deep Noise)that can disentangle biological signals from the experimental ***,we aimed to classify the phenotypic impact of 1108 different genetic perturbations screened from 125,510 fluorescent microscopy images,which were totally unrecognizable by the human *** validated our model by participating in the Recursion Cellular Image Classification Challenge,and Deep Noise achieved an extremely high classification score(accuracy:99.596%),ranking the 2nd place among 866 participating *** promising result indicates the successful separation of biological and technical factors,which might help decrease the cost of treatment development and expedite the drug discovery *** source code of Deep Noise is available at https://***/Scu-sen/Recursion-Cellular-Image-Classification-Challenge.

Pathological matrix stiffness promotes cardiac fibroblast differentiation through the POU2F1 signaling pathway

Cardiac fibroblast(CF)differentiation into myofibroblasts is a crucial cause of cardiac fibrosis,which increases in the extracellular matrix(ECM)*** increased stiffness further promotes CF differentiation and ***,the molecular mechanism is still *** used bioinformatics analysis to find new candidates that regulate the genes involved in stiffnessinduced CF differentiation,and found that there were binding sites for the POU-domain transcription factor,POU2F1(also known as Oct-1),in the promoters of 50 differentially expressed genes(DEGs)in CFs on the stiffer *** staining and Western blotting revealed that pathological stiffness upregulated POU2F1 expression and increased CF differentiation on polyacrylamide hydrogel substrates and in mouse myocardial infarction tissue.A chromatin immunoprecipitation assay showed that POU2F1 bound to the promoters of fibrosis repressors IL1R2,CD69,and *** expression of these fibrosis repressors was inhibited on pathological substrate *** of POU2F1 upregulated these repressors and attenuated CF differentiation on pathological substrate stiffness(35 kPa).Whereas,overexpression of POU2F1 downregulated these repressors and enhanced CF *** conclusion,pathological stiffness upregulates the transcription factor POU2F1 to promote CF differentiation by inhibiting fibrosis *** work elucidated the crosstalk between CF differentiation and the ECM and provided a potential target for cardiac fibrosis treatment.

Combined gene therapy of endostatin and interleukin 12 with polyvinylpyrrolidone induces a potent antitumor effect on hepatoma

AIM: To study the antitumor effect of combined gene therapy of endostatin and interleukin 12 (IL-12) with polyvinylpyrrolidone (PVP) on mouse transplanted ***: Mouse endostatin eukaryotic plasmid (pSecES) with a mouse Igk signal sequence inside and mouse IL-12 eukaryotic plasmid (pmIL-22) were transfected into BHK-22cells respectively. Endostatin and IL-22 were assayed by ELISA from the supernant and used to culture endothelial cells and spleen lymphocytes individually. Proliferation of the latter was evaluated by M-I-r. H22 cells were inoculated into the leg musde of mouse, which was injected intratumorallywith pSecES/PVP, pmIL-12/PVP or pSecES+pmIL-12/PVPrepeatedly. Tumor weight, serum endostatin and serumIL-22 were assayed. Tumor infiltrating lymphocytes, tumormicrovessel density and apoptosis of tumor cells were also displayed by HE staining, CD32 staining and ***: Endostatin and IL-12 were secreted after transfection, which could inhibit the proliferation of endothelial cells or promote the proliferation of spleen *** growth was highly inhibited by 92.8% after injection of pSecES+pmIL-i2/PVP accompanied by higher serum endostatin and IL-22, more infiltrating lymphocytes, fewertumor vessels and more apoptosis cells compared with injection of pSecES/PVP, pmIL-i2/PVP or vector/***: Mouse endostatin gene and IL-12 gene can be expressed after intratumoral injection with *** of hepatoma can be inhibited synergisticly,lymphocytes can be activated to infiltrate, and tumor cells are induced to apoptosis. Hepatoma can be highly inhibited or eradiated.

The high-spin states of the odd-odd nucleus 170Ta have been studied by the 150Gd(19F,4n)170Ta reaction at the beam energy of 97 *** α=1 sequence of the semi-decoupled band has been pushed to higher-spin states and the signature inversion point was observed at 19.5ħ.The results are compared with those of the neighboring odd-odd nuclei.

信息服务的可表达性是该项目关键科学问题之一。它是准确设计和表达服务流程与内容,提供动态、精准、可伸缩的信息服务,以满足用户不确定和多样性服务需求的关键。该报告面向异构融合网络环境,提出了环境信息可表达的信息服务描述模型,并基于此模型实现了支持环境描述的OWL-CS服务描述语言。具体工作有,首先分析影响信息服务调用、组合、协同的环境因素,将环境分为自然环境和社会环境两大类,其中服务的自然环境是服务部署和运行所依托宿主的软硬件环境,而社会环境是服务生态系统中体现的服务和服务之间的关系,以及服务和用户之间的关系。其次,给出环境上下文参数及度量方法,自然环境上下文是指用于刻画服务自然环境中的实体的任何信息,而社会环境上下文是指服务在相互作用过程中所表现出来的和社会属性相关的任何信息。再次,建立服务环境上下文元模型,设计实现了支持环境描述的OWL-CS服务描述模型。目前,语义Web服务的标准本体模型OWL-S的Service类包括了Service Profile、Service Model和Service Grounding三个类。该报告在此基础上在Service类中增加一个上下文类Service Context,该类用于描述服务的环境上下文信息。这种扩展方法,也使得OWL-CS和OWL-S兼容。通过Protégé4.3软件,可以展示OWL-CS的本体结构,并编辑生成具有环境描述的基于OWL-CS的服务描述文件。最后,该报告给出一个智慧校园中的实例,进一步说明环境上下文在服务选择中的作用。为了实现不同服务之间的信息交互和协作,需要提供一种统一的方法来描述服务的功能,以进行服务的理解、发现和协同。在环境敏感的场景下,更是需要统一描述服务环境的方法。国际上已有研究人员关注到服务上下文信息,提出上下文化服务(Contextual Service)的概念。本报告在国际上较早开展此类工作,特别是在服务环境建模、服务社会环境上下文的研究上具有自己的特色。
