Antitumor Compound Identification from Zanthoxylum bungeanum Essential Oil Based on Composition-activity Relationship

The chemical composition of Zanthoxylum bungeanum（Z bungeanum） essential oi1（39 batches） was analyzed by gas chromatography-mass spectrometry（GC-MS） analysis（23 ingredients）, and the antitumor activity against HeLa cells was detected via the MTT[3-（4,5-dimethylthiazol-2-yl）-2,5-diphenyltetrazolium bromide] assay. Two composition-activity relationship（CAR） models, generalized regression neural network（GRNN） and support vector regression（SVR） were respectively used to calculate the mean impact value（MIV） so as to identify bioactive com- pounds. Among them 9 ingredients（peaks 4, 15, 7, 8, 13, 3, 16, 9 and 11） were selected due to their high absolute MIVs. All of them have been confirmed with potential antitumor activity by previous researches. The CAR model combined with MIV was expected to be a credible tool for the active compound identification from herbs.

In high-altitude nuclear detonations,the proportion of pulsed X-ray energy can exceed 70%,making it a specific monitoring signal for such *** pulsed X-rays can be captured using a satellite-borne X-ray detector following atmospheric *** quantitatively analyze the effects of different satellite detection altitudes,burst heights,and transmission angles on the physical processes of X-ray transport and energy fluence,we developed an atmospheric transmission algorithm for pulsed X-rays from high-altitude nuclear detonations based on scattering *** proposed method is an improvement over the traditional analytical method that only computes direct-transmission *** traditional analytical method exhibits a maximum relative error of 67.79% compared with the Monte Carlo *** improved method reduces this error to within 10% under the same conditions,even reaching 1% in certain ***,its computation time is 48,000 times faster than that of the Monte Carlo *** results have important theoretical significance and engineering application value for designing satellite-borne nuclear detonation pulsed X-ray detectors,inverting nuclear detonation source terms,and assessing ionospheric effects.

Immune checkpoint blockade(ICB)has emerged as a promising immunotherapeutic modality against cancer in the ***,only 10-30%of patients respond to ICB,primarily due to poor immunogenicity and insufficient T cell infiltration in solid ***,we presented an approach for high-performance cancer treatment using the programmed cell death protein-1 and programmed cell death ligand-1(PD-1/PD-L1)inhibitor(BMS-202)-loaded PEGylated graphene oxide(GPi).On the one hand,GPi dissociated tight junctions of vascular endothelial cells(VECs)in tumor,thus promoting the extravasation and intratumoral accumulation of liposomal doxorubicin(lipDox),which then effectively induced immunogenic cell death of tumor *** the other hand,GPi also stimulated VECs to upregulate the expression of cell-cell interaction molecules,such as intercellular cell adhesion molecule-1 and vascular cell adhesion molecule-1,which facilitated the infiltration of T cells in *** acting as a stimulator of VECs,GPi could exert responsive release of BMS-202 under the acidic tumor microenvironment and blockade PD-1/PD-L1 axis in ***,the alternating administration of GPi and lipDox effectively inhibited tumor growth in a 4T1 tumor model,providing a novel treatment mode for chemo-immunotherapy.

The injection of a polymer chain into a small circular cavity under tangential self-propelled force is studied by using Langevin dynamics *** indicate that the injection dynamics of the active polymer shows strong correlation with the polymer conformation inside the cavity depending on the polymer rigidity(k_(b)).The injection timeτvaries nonmonotonously with increasing k_(b),and reaches its minimum at k_(b)^(*).When k_(b) is small(k_(b)≪k_(b)^(*)),the polymer is nearly random coil in the cavity,and spends a long time at the final stage of the injection process due to the large repulsion between monomers inside the *** k_(b) is moderate(k_(b)∼k_(b)^(*)),the part of polymer inside the cavity forms spiral configuration under the tangential active force,and the whole polymer moves synchronously with a constant velocity during the injection process,leading to a small injection *** k_(b) is large(k_(b)≫k_(b)^(*)),the polymer is nearly straight at the initial stage of the injection process,and takes a long time to bend itself,leading to a large injection time.

The Chinese Bayan Obo deposit is a world-class rare earth element(REE) deposit with considerable niobium(Nb) and iron(Fe) resources. A complete genetic understanding on all metals is fundamental for establishing genetic models at Bayan Obo. With extensive research being focused on REE enrichment, the timing and controls of Nb enrichment remain unresolved at Bayan Obo, which is mainly due to the challenges in dating, i.e. multistage thermal events, fine-grained minerals with complex textures and the rare occurrence of uranium-enriched minerals with mature dating methods. Based on robust geological and petrographic frameworks, here we conducted ion probe uranium-lead(U-Pb) dating of ferrocolumbite to unravel the timing, hence the genesis of Nb mineralization. Three types of hydrothermal ferrocolumbites—key Nb-bearing minerals—are identified based on their textures and mineral assemblages. They yield U-Pb ages of 1312 ± 47 Ma(n = 99), 438 ± 7 Ma(n = 93), and 268 ± 5 Ma(n = 19), respectively. In line with deposit geology, we tentatively link the first, second and third stage Nb mineralization to Mesoproterozoic carbonatite magmatism, ubiquitous early Paleozoic hydrothermal activity, and Permian granitic magmatism, respectively. While quantifying the contribution of metal endowment from each stage requires further investigation, our new dates highlight that multi-stage mineralization is critical for Nb enrichment at Bayan Obo, which may also have implications for the enrichment mechanism of Nb in REE deposits in general.

The casing deformation prevention technology based on the optimization of cement slurry is proposed to reduce the casing deformation of shale oil and gas wells during hydraulic fracturing. In this paper, the fracture mechanism of hollow particles in cement sheath was firstly analyzed by discrete element method, and the effect of hollow particles in cement on casing deformation was investigated by laboratory experiment method. Finally, field test was carried out to verify the improvement effect of the casing deformation based on cement slurry modification. The results show that the formation displacement can be absorbed effectively by hollow particles inside the cement transferring the excessive deformation away from casing. The particles in the uncemented state provide deformation space during formation slipping. The casing with diameter of 139.7 mm could be passed through by bridge plug with the diameter of 99 mm when the mass ratio of particle/cement reaches 1:4. According to the field test feedback, the method based on optimization of cement slurry can effectively reduce the risk of casing deformation, and the recommended range of hollow microbeads content in the cement slurry is between 15% and 25%.

Objective The determination of non-inferiority margin is an important and confusing issue which directly influences the acceptability of a new medication. We reviewed the published literature, International Conference on Harmonization of Technical Requirements for Registration of Pharmaceuticals for Human Use （ICH） Guidelines and Committee for Proprietary Medicinal Products （CPMP） papers on the selection of non-inferiority margin and the corresponding statistical tests in clinical trials, in order to provide practical recommendations and suggestions for establishing reference criteria for the non-inferiority margin in China. Data sources The literature on the selection of a non-inferiority margin and statistical tests was mainly extracted from relevant English articles on non-inferior clinical trials published from 1990 to 2007. The starting point （1990） was chosen due to lack of such papers published prior to 1990. This literature was searched via PubMed, Medline and Chinese Knowledge Information （CNKI）. ICH guidelines and CPMP papers were downloaded from their official websites. The keywords ＂clinical trial＂, ＂non-inferiority＂ and ＂non-inferiority margin＂ were used. Study selection Forty-three original articles and critical reviews, ICH El0 guideline and CPMP papers were selected. Results The non-inferiority testing with treatment difference and ratio are commonly used, where the non-inferiority margin is determined with and without historical data. Traditionally, this margin is treated as a fixed value, while developed methods take the variation into account in the determination of this margin, on which the test depends is more convincing. The mixed margin consisting of a margin based on treatment difference and a margin based on treatment ratio can exactly control the type I error at the desirable level and obtain a better power.
