Transcriptome analysis suggests broad jejunal alterations in Linghu’s obesity-diarrhea syndrome:A pilot study

BACKGROUND Obesity is associated with a significantly increased risk for chronic diarrhea,which has been proposed as Linghu’s obesity-diarrhea syndrome(ODS);however,its molecular mechanisms are largely *** To reveal the transcriptomic changes in the jejunum involved in *** In a cohort of 6 ODS patients(JOD group),6 obese people without diarrhea(JO group),and 6 healthy controls(JC group),high-throughput sequencing and bioinformatics analyses were performed to identify jejunal mucosal mRNA expression alterations and dysfunctional biological *** another cohort of 16 ODS patients(SOD group),16 obese people without diarrhea(SO group),and 16 healthy controls(SC group),serum diamine oxidase(DAO)and D-lactate(DLA)concentrations were detected to assess changes in intestinal barrier *** The gene expression profiles of jejunal mucosa in the JO and JC groups were similar,with only 1 differentially expressed gene(DEG).The gene expression profile of the JOD group was significantly changed,with 411 DEGs compared with the JO group and 211 DEGs compared with the JC group,129 of which *** enrichment analysis of these DEGs showed that the biological processes such as digestion,absorption,and transport of nutrients(especially lipids)tended to be up-regulated in the JOD group,while the biological processes such as rRNA processing,mitochondrial translation,antimicrobial humoral response,DNA replication,and DNA repair tended to be down-regulated in the JOD *** DEGs(CDT1,NHP2,EXOSC5,EPN3,NME1,REG3A,PLA2G2A,and PRSS2)may play a key regulatory role in the pathological process of ODS,and their expression levels were significantly decreased in ODS patients(P<0.001).In the second cohort,compared with healthy controls,the levels of serum intestinal barrier function markers(DAO and D-LA)were significantly increased in all obese individuals(P<0.01),but were higher in the SOD group than in the SO group(P<0.001).CONCLUSION Compared with health

Changes in posterior corneal elevation after small incision lenticule extraction for different myopic diopters

●AIM:To study the changes and effect factors of posterior corneal surface after small incision lenticule extraction(SMILE)with different myopic diopters.●METHODS:Ninety eyes of 90 patients who underwent SMILE were included in this retrospective *** were allocated into three groups based on the preoperative spherical equivalent(SE):low myopia(SE≥-3.00 D),moderate myopia(-3.00 D>SE>-6.00 D)and high myopia(SE≤-6.00 D).Posterior corneal surfaces were measured by a Scheimpflug camera preoperatively and different postoperative times(1wk,1,3,6mo,and 1y).Posterior mean elevation(PME)at 25 predetermined points of 3 concentric circles(2-,4-,and 6-mm diameter)above the best fit sphere was analyzed.●RESULTS:All surgeries were completed uneventfully and no ectasia was found through the *** difference of myopia group was significant at the 2-mm ring at 1 and 3mo postoperatively(1mo:P=0.017;3mo:P=0.018).The effect of time onΔPME was statistically significant(2-mm ring:P=0.001;4-mm ring:P<0.001;6-mm ring:P<0.001).The effect of different corneal locations onΔPME was significant except 1wk postoperatively(1mo:P=0.000;3mo:P=0.000;6mo:P=0.001;1y:P=0.001).Posterior corneal stability was linearly correlated with SE,central corneal thickness,ablation depth,residual bed thickness,percent ablation depth and percent stromal bed thickness.●CONCLUSION:The posterior corneal surface changes dynamically after *** protrusion is observed on the posterior corneal surface in patients with different degrees of myopia within one year after *** has good stability,accuracy,safety and predictability.

Highly Thermoconductive,Strong Graphene‑Based Composite Films by Eliminating Nanosheets Wrinkles

Graphene-based thermally conductive composites have been proposed as effective thermal management materials for cooling high-power electronic ***,when flexible graphene nanosheets are assembled into macroscopic thermally conductive composites,capillary forces induce shrinkage of graphene nanosheets to form wrinkles during solution-based spontaneous drying,which greatly reduces the thermal conductivity of the ***,graphene nanosheets/aramid nanofiber(GNS/ANF)composite films with high thermal conductivity were prepared by in-plane stretching of GNS/ANF composite hydrogel networks with hydrogen bonds andπ-π*** in-plane mechanical stretching eliminates graphene nanosheets wrinkles by suppressing inward shrinkage due to capillary forces during drying and achieves a high in-plane orientation of graphene nanosheets,thereby creating a fast in-plane heat transfer *** composite films(GNS/ANF-60 wt%)with eliminated graphene nanosheets wrinkles showed a significant increase in thermal conductivity(146 W m^(−1)K^(−1))and tensile strength(207 MPa).The combination of these excellent properties enables the GNS/ANF composite films to be effectively used for cooling flexible LED chips and smartphones,showing promising applications in the thermal management of high-power electronic devices.

Nonlinear proinflammatory effect of short-term PM_(2.5)exposure:A potential role of lipopolysaccharide

The association between PM_(2.5)(particulate matter≤2.5μm)short-term exposure and its health effect is non-linear from the epidemiological *** this nonlinearity is suggested to be related with the PM_(2.5)heterogeneity,however,the underlying biological mechanism is still ***,a total of 38 PM_(2.5)filters were collected continuously for three weeks in winter Beijing,with the ambient PM_(2.5)varying between 10 and 270μg/m^(3).Human monocytes-derived macrophages(THP-1)were treated with PM_(2.5)water-soluble elutes at10μg/mL to investigate the PM_(2.5)short-term exposure effect from a proinflammatory *** proinflammatory cytokine tumor necrosis factor(TNF)induced by the PM_(2.5)elutes at equal concentrations were unequal,showing the heterogeneity of PM_(2.5)proinflammatory *** the various chemical and biological components,lipopolysaccharide(LPS)showed a strong positive association with the TNF ***,some outliers were observed among the TNF-LPS ***,for PM_(2.5)from relatively clean air episodes,the higher LPS amount corresponded to relatively low TNF *** this phenomenon was also observed in the promotion tests by treating macrophages with PM_(2.5)elutes dosed with additional trace *** expression analysis indicated the involvement of oxidative-stress related genes in the LPS signaling ***,a potential oxidative-stress-mediated suppression on the PM_(2.5)-borne LPS proinflammatory effect was proposed to be accounted for the ***,the results showed the differential role of LPS in the heterogeneity of PM_(2.5)proinflammatory effects from a component-based *** experimental studies are needed to elucidate the signaling pathway of LPS attached on PM_(2.5)from different air quality episodes.

Multiple myeloma(MM)remains a challenging hematologic malignancy despite advancements in chimeric antigen receptor T-cell(CAR-T)*** targets of CAR-T cells used in MM immunotherapy have limitations,with a subset of patients experiencing antigen loss resulting in ***,novel targets for enhancing CAR-T cell therapy in MM remain *** receptor-like 5(FCRL5)is a protein marker with considerably upregulated expression in MM and has emerged as a promising target for CAR-T cell therapeutic interventions,offering an alternative treatment for *** further explore this option,we designed FCRL5-directed CAR-T cells and assessed their cytotoxicity in vitro using a co-culture system and in vivo using MM cell-derived xenograft models,specifically focusing on MM with gain of chromosome *** the challenges in CAR-T therapies arising from limited T cell persistence,our approach incorporates interleukin-15(IL-15),which enhances the functionality of central memory T(TCM)cells,into the design of FCRL5-directed CAR-T cells,to improve cytotoxicity and reduce T-cell dysfunction,thereby promoting greater CAR-T cell survival and *** in vitro and xenograft models displayed that FCRL5 CAR-T cells incorporating IL-15 exhibited potent antitumor efficacy,effectively inhibiting the proliferation of MM cells and leading to remarkable tumor *** results highlight the capacity of FCRL5-specific CAR-T cells with the integration of IL-15 to improve the therapeutic potency,suggesting a potential novel immunotherapeutic strategy for MM treatment.

High-efficiency preparation of multifunctional conjugated electrospun graphene doped PVDF/CF yarns for energy harvesting and human movement monitoring in TENG textile

Portable power is an effective solution to realize self-powered sensors for wearable devices,promoting future sustainable ***-based triboelectric nanogenerators(M-TENGs)have emerged as a promising technology for harvesting biomechanical energy from human motion owing to their advantages,such as simple structure,lightweight design,and efficient energy ***,the poor durability,low adaptability,and un-washability of two-dimensional membrane materials have largely hindered their application in wearable *** this study,we propose a sheath-core polyvinylidene fluoride(PVDF)/graphene(G)-carbon fiber(CF)yarn fabricated via conjugate electrospinning,comprising a commercial CF core and an electrospun graphene-doped PVDF sheath,which improves the fatigue resistance of electrospun nanofiber films under prolonged friction and keeps a high degree of *** resulting electronic textile,woven with the large-scale electrospun PVDF/G-CF yarn,demonstrates a remarkable power density of 25.5 mW·m^(-2).The tight distribution of PVDF/G nanofibers on the textile surface ensures excellent softness,washability,and ***,the electrospun PVDF/G-CF textile exhibits significant potential in pressure sensing,self-powered operation,and motion detection,making it highly suitable for wearable electronics applications.

Sustainable Intensification and Large-scale Operation of Cultivated Land Use at the Farmers’ Scale:A Case Study of Shandong Province,China

Sustainable intensification of cultivated land use(SICLU) and large-scale operations(LSO) are widely acknowledged strategies for enhancing agricultural ***,the existing literature has faced challenges in precisely defining SICLU and constructing comprehensive indicators,which has hindered the exploration of factors influencing LSO within the SICLU *** address this gap,we integrated self-efficacy theory into the design of an index framework for evaluating *** subsequently employed econometric models to analyze the significant factors that impact *** findings reveal that SICLU can be divided into four key dimensions:intensive management,efficient output,resource conservation,and ecological environment ***,it is crucial to incorporate belief-based cognitive factors into the index system,as farmers’ understanding of fertilizer and pesticide application significantly influences their willingness to engage in ***,we identify grain market turnover as the most influential factor in promoting LSO,with single-factor contribution rates reaching 70.9% for cultivated land transfer willingness and 62.5% for the total planting ***,unlike irrigation and agricultural machinery inputs,increased labor inputs correspond to larger planting areas for *** trend may be attributed to reduced labor availability because of rural labor migration,whereas the reduction in irrigation and agricultural input is contingent on innovations in production practices and the transfer of cultivated land management ***,SICLU dynamically influences LSO,with each index related to SICLU having an optimal range that fosters *** insights offer valuable guidance for policymakers,emphasizing farmers as their central focus,with the adjustment of input and output factors as a means to achieve LSO as the ultimate *** conclusion,we propose research avenues for further enriching the SICLU framework to ensur

Supersaturation-driven self-assembly formation of ceftriaxone sodium spherulites:From amorphous form to spherulites

Drug particles with spherical morphology possess amazing advantages in terms of particle flowability,mechanical properties,drug solubility,and *** growth mechanism of drug spherulite is of great importance for the preparation and regulation of ***,ceftriaxone sodium spherulites were fabricated by the antisolvent crystallization method using dropwise addition of ceftriaxone sodium solution to *** observation of the whole crystallization process combined with electron microscopy technique revealed the spherical growth process from amorphous form to *** the supersaturation of the crystallization system was adjusted,the ceftriaxone sodium crystals transformed from amorphous form to *** the process of antisolvent crystallization with acetone as antisolvent,when the theoretical supersaturation degree S was higher than 2.62,the crystallization system tended to appear amorphous form;when S was between 2.57 and 2.62,the amorphous form transformed into clustered spherulites;when S was less than 2.57,the surface of spherulites will be covered with flaky crystal,which transformed into urchin-like *** the understanding of the spherical growth mechanism,the ceftriaxone sodium spherulites prepared in this research with modified supersaturation control had a low residue of antisolvent acetone,and the flowability was significantly improved.
