Objective: The aim of this study was to identify the potential pharmacological mechanisms of Gualou-Xiebai-Banxia decoction(GLXBBX) against unstable angina(UA). materials and Methods: The active compounds of GLXBBX were collected from the Traditional Chinese Medicine Systems Pharmacology Database and Analysis Platform, and their targets were predicted using the Swiss Target Prediction database. The targets associated with UA were obtained from the Online Mendelian Inheritance in man, Gene Cards, and Therapeutic Target Database. Individual targets associated with UA and GLXBBX were cross-checked to identify the targets of GLXBBX involved in the treatment of UA. A protein–protein interaction network was built using the STRING online database. Cytoscape 3.7.2 software was used to screen out hub genes. Additional gene ontology and Kyoto Encyclopedia of Genes and Genomes enrichment analyses were performed using the cluster Profiler package in R. Results: A total of 28 bioactive compounds and 320 protein targets of GLXBBX associated with UA were screened out. Enrichment analysis indicated that the therapeutic effect of GLXBBX may be mediated through the PI3K/AKT, maPK, and HIF-1 signaling pathways. Molecular docking suggested that the active compounds including Vitamin E, cavidine, and baicalein can bind to their protein receptors. Conclusions: This research confirmed the multifactorial effects of GLXBBX in the treatment of UA and laid the foundation for the experimental research on GLXBBX.

目的:构建新型AFP顺式作用元件调控的基因表达载体,检测该调控元件的特异性和活性表达.方法:设计含有特定酶切位点的引物,采用PCR法从HepG2细胞中克隆AFP启动子及增强子基因亚区片段.启动子与增强子长、短片段分别与含有报告基因荧光素酶基因的载体pGL-3的多克隆位点连接,构建不同长度hAFP增强子调控的肝癌特异性Luciferase表达载体APSE-Luc/APLE-Luc.经测序,PCR及酶切鉴定各重组体.用脂质体法将表达载体转染表达或不表达AFP的肿瘤细胞系进行荧光强度及特异性表达测定.结果:成功地将AFP基因启动子、增强子克隆到报告基因载体pGL-3的多克隆位点,构建成为不同长度hAFP增强子调控的肝癌特异性Luciferase表达载体APSE-Luc/APLE-Luc,酶切鉴定和DNA序列分析无误.转染APLE-Luc质粒的细胞中Luciferase表达量明显高于转染APSE-Luc质粒的细胞,其在HepG2细胞中的表达明显高于SMMC7721细胞及Hela细胞.结论:成功构建AFP启动子与增强子联合调控载体APSE-Luc/***增强子能够特异性地增强目的基因在AFP阳性细胞中表达,并且不同的亚区活性不同,长片段的活性明显高于短片段.

Efficient and stable photocathodes with versatility are of significance in photoassisted lithium-ion batteries(PLIBs),while there is always a request on fast carrier transport in electrochemical active *** work proposes a general approach of creating bulk heterojunction to boost the carrier mobility of photocathodes by simply laser assisted embedding of plasmonic *** employed in PLIBs,it was found effective for synchronously enhanced photocharge separation and transport in light charging ***,experimental photon spectroscopy,finite difference time domain method simulation and theoretical analyses demonstrate that the improved carrier dynamics are driven by the plasmonic-induced hot electron injection from metal to TiO_(2),as well as the enhanced conductivity in TiO2 matrix due to the formation of oxygen vacancies after Schottky *** from these merits,several benchmark values in performance of TiO2-based photocathode applied in PLIBs are set,including the capacity of 276 mah g^(−1) at 0.2 A g^(−1) under illumination,photoconversion efficiency of 1.276%at 3 A g^(−1),less capacity and Columbic efficiency loss even through 200 *** results exemplify the potential of the bulk heterojunction strategy in developing highly efficient and stable photoassisted energy storage systems.

To investigate the associations between isocarbophos and isofenphos with impaired fasting glucose(IFG)and type 2 diabetes mellitus(T2DM),and to assess the mediation roles of inflammation *** were 2701 participants in the case-control study,including 896 patients with T2DM,900 patients with IFG,905 subjects with *** isocarbophos and isofenphos concentrationsweremeasured using gas chromatography and triple quadrupole tandem mass *** linear models were used to calculate the relationships between plasma isofenphos and isocarbophos levels with inflammatory factor levels and *** cell was used as mediators to estimate the mediating effects on the above *** and isofenphos were positively related with T2DM after adjusting for other *** odds ratio(95%confidence interval)(OR(95%CI))for T2DM was 1.041(1.015,1.068)and for IFG was 1.066(1.009,1.127)per unit rise in *** prevalence of T2DM increased by 6.4%for every 1 unit more of ln-isofenphos(OR(95%CI):1.064(1.041,1.087)).Additionally,a 100%rise in ln-isocarbophos was linked to 3.3%higher ln-HOma2IR and a 0.029 mmol/L higher glycosylated hemoglobin(HbA1c)(95%CI:0.007,0.051).While a 100%rise in ln-isofenphos was linked to increase in ln-HOma2 and ln-HOma2IR of 5.8%and 3.4%,***,white blood cell(WBC)and neutrophilic(NE)were found to be mediators in the relationship between isocarbophos and T2DM,and the corresponding proportions were 17.12%and 17.67%,*** and isocarbophos are associated with IFG and T2DM in the rural Chinese population,WBC and NE have a significant role in this relationship.

Rechargeable aqueous zinc-ion batteries(AZIBs)exhibit appreciable potential in the domain of electrochemical energy ***,there are serious challenges for AZIBs,for instance zinc dendrite growth,hydrogen evolution reaction(HER),and corrosion side ***,we propose a surface engineering modification strategy for coating the montmorillonite(MMT)layer onto the surface of the Zn anode to tackle these issues,thereby achieving high cycling stability for rechargeable *** results reveal that the MMT layer on the surface of the Zn anode is able to provide ordered zincophilic channels for zinc ions migration,facilitating the reaction kinetics of zinc *** functional theory(DFT)calculations and water contact angle(CA)tests prove that MMT@Zn anode exhibits superior adsorption capacity for Zn^(2+)and better hydrophobicity than the bare Zn anode,thereby achieving excellent cycling ***,the MMT@Zn||MMT@Zn symmetric cell holds the stable cycling over 5600 h at 0.5 ma cm^(-2)and 0.125 m A h cm^(-2),even exceeding 1800 h long cycling under harsh conditions of 5 m A cm^(-2)and 1.25 m A h cm^(-2).The MMT@Zn||V_(2)O_(5)full cell reaches over 3000 cycles at 2 A g^(-1)with excellent rate ***,this surface engineering modification strategy for enhancing the electrochemical performance of AZIBs represents a promising application.

蓄水坑灌体系温度对土壤氮素运移转化影响

通过研究体系温度对蓄水坑灌施条件下土壤水分及氮素运移转化的影响,明确蓄水坑灌土壤水氮时空分布特征,探究土壤水氮运移迁移转化机理,以期为水肥合理灌施提供理论基础。通过模拟构建蓄水坑灌模型,以大型控温箱精确控制土壤温度,采用克里克空间插值法分析了蓄水坑灌条件不同体系温度下的水分、硝态氮、铵态氮时空分布特征,结果显示7 h左右土壤水分、养分完成入渗进入再分布阶段,土壤水分随着时间的推移其垂向和径向迁移距离均逐渐增大,同一时刻,温度越高其横向与径向迁移距离越大,且靠近蓄水坑壁区域的土壤含水率相对越低;土壤中铵态氮含量在不同温度下随时间推移均呈现先增后减的现象,低温下第15 d时土壤养分再分布核心区出现下降趋势,中、高温第10 d时已出现下降趋势,且其迁移距离远低于水分、硝态氮的迁移距离;土壤中硝态氮含量在10℃下第10 d时出现增高现象,而20、25、35℃下第5 d时已出现增高现象,由蓄水坑周边至湿润体边缘呈现"低-高-低"的分布态势。表明再分布阶段温度升高能提高水分的再分布速率,提高脲酶活性加快尿素水解转化为铵态氮,同时促进硝化反应进程抑制铵态氮在土壤中的积累,当土壤含水量过高时,会抑制土壤中氮素的硝化作用。

血瘀证患者差异基因表达谱研究

目的:应用寡核苷酸基因芯片技术,研究血瘀证患者差异基因表达谱。方法:16例血瘀证患者经冠状动脉造影诊断后,分为冠心病血瘀证组和非冠心病血瘀证组,每组8例;并选年龄和性别相匹配的8名健康人为健康对照组。抽取静脉血,分离白细胞,抽提RNA,质控芯片(Test3芯片)对样本质量进行检测,然后与Affymetrix U133 Plus 2.0芯片进行杂交,通过扫描和软件分析,比较冠心病血瘀证组、非冠心病血瘀证组与健康对照组的基因表达谱,筛选血瘀证相关差异基因,并进一步进行基因本体(Gene Ontology,GO)和通路分析,使用实时荧光定量逆转录聚合酶链反应法对目标基因进行验证。结果:通过差异基因筛选,与血瘀证相关的差异基因共有48个,其中上调基因26个,下调基因22个;通过GO分析,其中与炎症免疫相关的基因有5个,占10.4%;通路分析结果显示,有意义的10条通路中有5个涉及炎症和免疫反应。经实时荧光定量逆转录聚合酶链反应验证了基因芯片准确可靠。结论:血瘀证基因表达谱研究显示了炎症和免疫相关基因的比例和显著性优势,说明炎症和免疫反应在一定程度上介导了血瘀证的发生发展。

以丙烯酸单体为溶剂,利用聚己内酯二元醇(PCL1000)、异佛尔酮二异氰酸酯(IPDI)、2,2-二羟甲基丁酸(DMBA)、三羟甲基丙烷(TMP)合成水性聚氨酯丙烯酸酯(WPUA),WPUA中引入十三氟辛醇(FOH),制备无VOC溶剂自交联含氟聚氨酯丙烯酸酯(WFPUA)复合乳液。加入后扩链剂乙二胺基乙磺酸钠(AAS)可提高预聚体自乳化能力,降低乳胶粒粒径,提升WFPUA稳定性。研究了表面施胶后纸张的表面形态、防水防油、耐破度、撕裂度及抗张强度等性能。结果表明:聚合物在施胶后纸张表面纤维上成膜,纸张表面空隙减少;纸张与水和二碘甲烷接触角达到136.1°和105.3°;施胶后纸张耐破度达到266 k Pa,撕裂度达到732 m N,干湿抗张强度指数达到5.1 N·m/g和53 N·m/g,可以很好地运用到特种纸的制备中。
