Ameliorated stress related proteins are associated with improved cardiac function by sarcoplasmic reticulum calcium ATPase gene transfer in heart failure

Background Previous studies showed that overexpression of sarco-endoplasmic retieulum calcium ATPase （SERCA2a） in a variety of heart failure （HF） models was associated with greatly enhanced cardiac performance. However, it still undefined the effect of SERCA2a overexpression on the systemic inflammatory response and neuro-hormonal factors. Methods A rapid right ventricular pacing model of experimental HF was used in beagles. Then the animals underwent recombinant adeno-associated vires 1 （rAAV1） mediated gene trans- fection by direct intra-myocardium injection. HF animals were randomized to receive the SERCA2a gene, enhanced green fluorescent protein （control） gene, or equivalent phosphate buffered saline. Thirty days after gene delivery, the cardiac function was evaluated by echocardiographic testing. The protein level of SERCA2a was measured by western blotting. The proteomic analysis of left ventricular （LV） sample was determined using two-dimensional （2-D） gel el^ctrophoresis and MALDI-TOF-MS. The serum levels of the systemic inflammatory and neuro-hormonal factors were assayed using radioimmunoassay kits. Results The cardiac function improved after SERCA- 2a gene transfer due to the significantly increased SERCA2a protein level. Beagles treated with SERCA2a had significantly decreased serum levels of the inflammatory markers （interleukin-6 and tumor necrosis factor-a） and neuro-hormonal factors （brain natriuretic peptide, endothelin-1 and angiotensin II） compared with HF animals. The myocardial proteomic analysis showed that haptoglobin heavy chain, heat shock protein （alpha-crystallin-related, B6） were down-regulated, and galectin-1 was up-regulated in SERCA2a group compared with HF group, companied by up-regulated contractile proteins and NADH dehydrogenase. On the Monge–Kantorovich Mass Transfer Problem in Higher Dimensions

This paper mainly investigates the approximation of a global maximizer of the Monge–Kantorovich mass transfer problem in higher dimensions through the approach of nonlinear partial differential equations with Dirichlet *** an approximation mechanism,the primal maximization problem can be transformed into a sequence of minimization *** applying the systematic canonical duality theory,one is able to derive a sequence of analytic solutions for the minimization *** the final analysis,the convergence of the sequence to an analytical global maximizer of the primal Monge–Kantorovich problem will be demonstrated.

The development of stereodivergent synthetic methods to access all four stereoisomers of biologically importantα-fluoroγ-butyrolactones containing vicinal stereocenters is of great importance and poses a formidable challenge owing to ring strain and steric ***,a novel asymmetric[3+2]annulation ofα-fluoroα-azaaryl acetates with vinylethylene carbonate was successfully developed through Cu/Ir-catalyzed cascade allylic alkylation/lactonization,affording a variety of enantioenrichedα-fluoroγ-butyrolactones bearing vicinal stereogenic centers with high reaction efficiency and excellent levels of both stereoselectivity and regioselectivity(up to 98%yield,generally>20:1 dr and>99%ee).Notably,all four stereoisomers of these pharmaceutically valuable molecules could be accessed individually via simple permutations of two enantiomeric *** addition,other azaaryl acetates bearingα-methyl,α-chlorine orα-phenyl group were tolerated well in this *** mechanistic investigations were conducted to explore the process of this bimetallic catalysis based on the results of reaction intermediates,isotopic labelling experiments,and kinetic studies.

Correlation between coagulation function, tumor stage and metastasis in patients with renal cell carcinoma： a retrospective study

Background The coagulation function in carcinoma patients is abnormal, but in renal cell carcinoma the extent and relationships of coagulation function remain unclear. This study retrospectively investigated the relationships between coagulation function, clinical stage and metastasis in patients with renal cell *** A total of 350 consecutive patients admitted to our Urology Department from 2004 to 2010 were diagnosed with renal cell carcinoma by histolopathologic examination and were included in this study. A total of 231 cases of renal benign tumors were considered as the control group. Fibrinogen, prothrombin time, activated partial thromboplastin time and international normalized ratio were evaluated in all subjects. Tumor size, clinical stage, lymph node metastasis, and distant metastasis were evaluated using radiologic imaging, intraoperative findings, and histological *** The preoperative plasma fibrinogen levels of patients with renal cell carcinoma （（383.9±146.7） mg/dl） were significantly higher than those of the control group （（316.7±62.0） mg/dl）（P ＜0.01）. We divided the renal cell carcinoma group into stages la, lb, Ⅱ, Ⅲ, and Ⅳ. The fibrinogen values were （315.6±64.6） mg/dl, （358.3±91.1） mg/dl, （465.6±164.7）mg/dl, （500.0±202.1） mg/dl, and （585.8±179.7） mg/dl, respectively. There were no significant differences in fibrinogen values between stage la and control groups. However, results of other stages showed significant differences when compared to control group values （P ＜0.01）. Using the cutoff value of 440 mg/dl, which defines hyperfibrinogenemia,plasma fibrinogen levels had a positive predictive value of 39.8% and a negative predictive value of 93.3% for predicting distant metastasis, with a sensitivity of 64.7% and specificity of 83.3%.Concluslons Preoperative plasma fibrinogen levels are elevated in patients with renal cell carcinoma with distant metastasis or lymph node metastasis. Stochastic resonance is a phenomenon in which adding random noise to a system enhances the detection or transmission of a weak signal. It occurs when the noise interacts with the system's nonlinearity, thus improving signal-to-noise ratio and increasing sensitivity. Although parametric resonances in an atomic spinor Bose-Einstein condensate have been investigated, the question of whether one can observe stochastic resonance in such a system persists. In this study we propose a scheme for generating stochastic resonance in a cavity-spinor Bose-Einstein condensate coupling system. We demonstrate stochastic resonance through numerical calculations using the mean-field theory and truncated Wigner approximation methods. Furthermore, the characteristics of the system's response to noise and periodic signals are studied in detail. This study unravels a new scheme for observing stochastic resonance via linking atomic many-body physics with cavity quantum electrodynamics.
