Double fertilization is a key process of sexual reproduction in higher plants. The role of calcium in the activation of female sex cells through fertilization has recently received a great deal of attention. The establishment of a Ca^2＋-imaging technique for living, single, female sex cells is a difficult but necessary prerequisite for evaluating the role of Ca^2＋ in the transduction of external stimuli, including the fusion with the sperm cell, to internal cellular processes. The present study describes the use of Fluo-3 for reporting the Ca^2＋ signal in isolated, single, female sex cells, egg cells and central cells, of tobacco plants. A suitable loading protocol was optimized by loading the cells at pH 5.6 with 2 μM Fluo-3 for 30 min at 30 ℃. Under these conditions, several key factors related to in vitro fertilization were also investigated in order to test their possible effects on the [Ca^2＋]cyt of the female sex cells. The results indicated that the bovine serum albumin-fusion system was superior to the polyethlene glycol-fusion system for detecting calcium fluctuations in female sex cells during fertilization. The central cell was fertilized with the sperm cell in bovine serum albumin; however, no evident calcium dynamic was detected, implying that a transient calcium rise might be a specific signal for egg cell fertilization.

We report both experimentally and numerically that ultra-broadband asymmetric acoustic transmission is realized by a brass plate and a right triangle reflector immersed in water. This exotic phenomenon arises from the asymmetric excitation of the leaky asymmetric zero-order Lamb mode in the brass plate induced by the incident angle of external bulk waves. The results show that the bandwidth of the asymmetric acoustic transmission could reach 2000 k Hz, and the positive transmitted wave is only a single acoustic beam. The device has the advantages of ultra-broadband, single transmitted beam,and simpler structure, which has great potential applications in ultrasonic devices.

BACKGROUND Insomnia is the most common sleep *** disrupts the patient’s life and work,increases the risk of various health issues,and often requires long-term *** financial burden and inconvenience of treatments discourage patients from complying with them,leading to chronic *** To investigate the long-term home-practice effects of mindful breathing combined with a sleep-inducing exercise as adjunctive insomnia *** A quasi-experimental design was used in the present work,in which the patients with insomnia were included and grouped based on hospital admission:40 patients admitted between January and April 2020 were assigned to the control group,and 40 patients admitted between May and August 2020 were assigned to the treatment *** control group received routine pharmacological and physical therapies,while the treatment group received instruction in mindful breathing and a sleep-inducing exercise in addition to the routine *** Pittsburgh Sleep Quality Index(PSQI),Generalized Anxiety Disorder 7-item(GAD-7)scale,and Insomnia Severity Index(ISI)were utilized to assess sleepquality improvement in the patient groups before the intervention and at 1 wk,1 mo,and 3 mo *** The PSQI,GAD-7,and ISI scores before the intervention and at 1 wk postintervention were not significantly different between the ***,compared with the control group,the treatment group exhibited significant improvements in sleep quality,daytime functioning,negative emotions,sleep latency,sleep duration,sleep efficiency,anxiety level,and insomnia severity at 1 and 3 mo postintervention(P<0.05).The results showed that mindful breathing combined with the sleep-inducing exercise significantly improved the long-term effectiveness of insomnia *** 3 mo,the PSQI scores for the treatment vs the control group were as follows:Sleep quality 0.98±0.48 vs 1.60±0.63,sleep latency 1.98±0.53 vs 2.80±0.41,sleep duration 1.53±0.6

BACKGROUND Cancer is a growing threat to human *** to the double torment of cancer and cancer treatment,patients with advanced cancer generally have a low quality of *** present,there is a lack of systematic spiritual care plans for patients with advanced cancer as well as systematic guidance plans on the specific clinical application of spiritual care for advanced cancer *** hypothesized that our care plan would be effective in improving the spiritual and mental health and quality of life of patients with advanced *** To construct a spiritual care plan suitable for Chinese patients with advanced cancer through literature *** From February to December 2018,through purpose sampling,we selected 100 advanced cancer patients from the Oncology Department and Hospice Ward of a tertiary hospital in Liaoning Province who met the study *** were randomly divided into experimental and control groups,with 50 cases in each *** control group received the current routine care,while the experimental group received the advanced cancer spiritual care intervention in addition to routine *** After the intervention,the overall spiritual health score for the experimental group was higher than that of the control group(4.68±1.36 vs 3.63±1.71).The difference between the groups was statistically significant(P<0.05).The proportion of anxiety-free patients in the experimental group was 95.45%,which was significantly higher than the 60.98%in the control ***,the proportion of non-depressed patients in the experimental group was 97.73%,which was significantly higher than the 85.37%in the control group(P<0.05).The overall quality of life score for the experimental group was significantly higher than that of the control group(5.36±1.16 vs 4.39±1.36,P<0.05).CONCLUSION Our spiritual care plan for patients with advanced cancer could improve their spiritual health and quality of life and reduce negative mental health sympt

Cross-Reference Benchmarks for Translating the Genetic Soil Classification of China into the Chinese Soil Taxonomy

Soil classification is the foundation for exchange and extension of research findings in soil science and for modern management of soil resources. This study explained database and research methodology to create a cross-reference system for translating the Genetic Soil Classification of China (GSCC) into the Chinese Soil Taxonomy (CST). With the help of the CST keys, each of the 2 540 soil species in GSCC has been interpreted to its corresponding soil order, suborder, great group, and sub-group in CST. According to the methodology adopted, the assigned soil species have been linked one another to their corresponding polygons in the 1:1000000 digital soil map of China. Referencibility of each soil species between the GSCC and CST systems was determined statistically on the basis of distribution area of each soil species at a high taxon level of the two systems. The soils were then sorted according to their maximum referencibility and classified into three categories for discussion. There were 19 soil great groups in GSCC with maximum referencibility > 90% and 22 great groups between 60%-90%. These soil great groups could serve as cross-reference benchmarks. There were 19 great groups in GSCC with maximum referencibility < 60%, which could be used as cross-reference benchmarks until new and better results were available. For these soils, if the translation was made at a lower soil taxon level or on a regional basis, it would improve their referencibility enabling them to serve as new cross-reference benchmarks.

We report the realization of broadband reflected acoustic focusing lenses based on thermoacoustic phased arrays of Bessel-like beams, in which the units of phase manipulation are composed of three rigid insulated boundaries and a thermal insulation film in air with different temperatures. Based on these units, we realize a reflected focusing lens which can focus reflected acoustic energy on a line, and its fractional bandwidth can reach about 0.29. In addition, we discuss the influences of the base angle of Bessel-like beam, the number of basic unit, and the variation of unit temperature on focusing performances in details. Furthermore, the reflected focusing lens for the cylindrical acoustic wave based on the Bessel-like beam is also demonstrated. The proposed focusing lens has the advantages of a broad working bandwidth, large focus size,and high robustness, which may provide possibilities for the design and application of acoustic lenses.

Classification of network traffic using port-based or payload-based analysis is becoming increasingly difficult when many applications use dynamic port numbers, masquerading techniques, and encryption to avoid detection. In this article, an approach is presented for online traffic classification relying on the observation of the first n packets of a transmission control protocol （TCP） connection. Its key idea is to utilize the properties of the observed first ten packets of a TCP connection and Bayesian network method to build a classifier. This classifier can classify TCP flows dynamically as packets pass through it by deciding whether a TCP flow belongs to a given application. The experimental results show that the proposed approach performs well in online Internet traffic classification and that it is superior to naive Bayesian method.

AIM: To investigate the effect of astilbic acid (3β6β-dihydroxyolean-12-en-27-oic acidAA) on human colorectal carcinoma COLO 205 cell proliferation and apoptosis. METHODS: Proliferation of COLO 205 cells was measued by MTT assay. Content of DNA in COLO 205 cell was measued by modified diphenylamine assay. AA-induced morphological changes was observed with fluorescence microscope and transmission electron microscope . DNA fragmentation was visualized by agarose gel electrophoresis. Apoptosis rate and cell cycle distribution were determined by flow cytometric analysis. Expressions of Bcl-2 and Bax proteins were visioned by immunohistochemical analysis. The change of relative mitochondral transmembrane potential (MTP) in COLO 205 cell was analyzed with FCM after rhodamine 123 staining. RESULTS: The IC50 (96 h) of AA for inhibiting COLO 205 cell proliferation was 61.56±0.34 umol/L. A novel in vitro system for gamete fusion in maize

Various systems by using electric pulse, calcium, or polyethylene glycol have been developed in the past decade for the in vitro fusion of plant gametes. These in vitro systems provide a new way to study the fertilization mechanisms of plants. In this study, we developed a bovine serum albumin (BSA)-mediated fusion system for the in vitro fusion of maize gametes. The in vitro fusion of the isolated single egg cell and sperm cell of maize was observed microscopically in the BSA solution and the fertilized egg cell showed normal cell wall regeneration and nuclear division. The effects of the BSA concentration, pH value and calcium level on the efficiency of the maize gamete fusion were also assessed. BSA concentration and pH value did significantly affect the efficiency of the gamete fusion. Calcium was not necessary for the gamete fusion when BSA was present. The optimal solution for the gamete fusion contained 0.1% BSA, pH 6.0. The fusion frequency was as high as 96.7% in that optimal solution. This new in vitro fertilization system offers an alterna- tive tool for the in vitro study of fertilization mechanisms with much simpler manipulating procedure than PEG system, and it will be especially useful for the in vitro study of the calcium dynamics during plant fertilization.
