Autoimmune cholangitis arises from abnormal innate and adaptive immune responses in the liver,and T cells are critical drivers in this ***,little is known about the regulation of their functional behavior during disease *** previously reported that mice with T cell-restricted expression of a dominant negative form of transforming growth factor beta receptor type Ⅱ(dnTGFβ RII)spontaneously develop an autoimmune cholangitis that resembles human primary biliary cholangitis(PBC).Adoptive transfer of CD8^(+) but not CD4^(+) T cells into Rag1^(−/−)mice reproduced the disease,demonstrating a critical role for CD8^(+) T cells in PBC ***,we used SOMAscan technology to perform proteomic analysis of serum samples from dnTGFβRII and B6 control mice at different *** addition,we analyzed CD8 protein profiles after adoptive transfer of splenic CD8^(+) cells into Rag1^(−/−)*** use of the unique SOMAscan aptamer technology revealed critical and distinct profiles of CD8 cells,which are key to biliary *** total,254 proteins were significantly increased while 216 proteins were significantly decreased in recipient hepatic CD8^(+) cells compared to donor splenic CD8^(+) *** contrast to donor splenic CD8^(+) cells,recipient hepatic CD8^(+) cells expressed distinct profiles for proteins involved in chemokine signaling,focal adhesion,T cell receptor and natural killer cell-mediated cytotoxicity pathways.

Soybean （Glycine max （L.） Merr.） is a major crop that provides plant-origin protein and oil for humans and livestock. Al- though the soybean vegetative tissues and seeds provide a major source of high-quality protein, they suffer from low con- centration of an essential sulfur-containing amino acid, methionine, which significantly limits their nutritional quality. The level of methionine is mainly controlled by the first unique enzyme of methionine synthesis, cystathione y-synthase （CGS）. Aiming to elevate methionine level in vegetative tissues and seeds, we constitutively over-expressed a feedback-insensitive Arabidopsis CGS （AtD-CGS） in soybean cultivars, Zigongdongdou （ZD） and Jilinxiaoli 1 （JX）. The levels of soluble methionine increased remarkably in leaves of transgenic soybeans compared to wild-type plants （6.6- and 7.3-fold in two transgenic ZD lines, and 3.7-fold in one transgenic JX line）. Furthermore, the total methionine contents were significantly increased in seeds of the transgenic ZD lines （1.5- to 4.8-fold increase） and the transgenic JX lines （1.3- to 2.3-fold increase） than in the wild type. The protein contents of the transgenic soybean seeds were significantly elevated compared to the wild type, suggesting that the scarcity of methionine in soybeans may limit protein accumulation in soybean seeds. The increased protein content did not alter the profile of major storage proteins in the seeds. Generally, this study provides a promising strategy to increase the levels of methionine and protein in soybean through the breeding programs.

Abdominal drainage was previously recommended as a post-hepatectomy procedure for patients with *** report introduces a simple technique that prevents leakage of ascitic fluid after cirrhotic hepatectomy complicated by blockage of the abdominal *** 59 patients who had had cirrhotic hepatectomy complicated by leakage of ascites in the drain site after drainage removal between January 2001 and April 2011,31 underwent suture ligation(sutured group) and 28 were given urostomy bag at the abdominal drainage site(drainage group).The mean length of postoperative hospital stay in the drainage group was shorter than in the sutured group(16.11±2.61 vs 34.23±4.86 days,P=0.000).Meanwhile,the drainage group showed decreased postoperative complications,including leakage of ascites,wound infection,and collection of *** by urostomy bag can prevent prolonged leakage of ascitic fluid after the blockage of abdominal drains in patients undergoing cirrhotic hepatectomy.

OBJECTIVE:There are still a high proportion of patients with ST-segment elevation myocardial infarction (STEMI) missing out early reperfusion even in the primary percutaneous coronary intervention (PCI) era. Most of them are stable latecomers, but the optimal time to undergo delayed PCI for stable ones remains controversial.;METHODS:We investigated all STEMI patients who underwent delayed PCI (2-28 days after STEMI) during 2007-2010 in Beijing and excluded patients with hemodynamic instability. The primary outcome was major adverse cardiovascular events (MACEs).;RESULTS:This study finally enrolled 5,417 STEMI patients and assigned them into three groups according to individual delayed time (Early group, 55.9%; Medium group, 35.4%; Late group, 8.7%). During 1-year follow-up, MACEs occurred in 319 patients. The incidence of MACEs were respectively 7.1%, 5.6% and 6.7% among three groups. The Medium group had less recurrent myocardial infarction plus cardiac death (hazard ratio, 0.525; 95% confidence interval, 0.294-0.938, = 0.030) than Late group and less repeat revascularization (hazard ratio, 0.640; 95% confidence interval, 0.463-0.883, = 0.007) than Early group in pairwise comparisons. We depicted the incidence of major adverse cardiovascular event (MACE) by delayed time as a quadratic curve and found the bottom appeared at day 14.;CONCLUSIONS:The delayed PCI time varied in the real-world practice, but undergoing operations on the second week after STEMI had greater survival benefit and less adverse events for whom without early reperfusion and hemodynamic instability.

炎症性肠病(inflammatory bowel disease,IBD)是一组特定的肠道疾病的统称,主要体现为慢性及复发性的肠道炎症。其全球发病率正在迅速上升,严重影响人们生活质量与生命健康,已成为当前的全球性健康问题。在过去的几十年里,大量研究表明IBD的发病率与肠道菌群失调有关。因此,以肠道益生菌为主的治疗策略成为一大热点,其中大肠杆菌Nissle 1917(Escherichia coli Nissle 1917,EcN)备受关注。本文聚焦肠道益生菌EcN在治疗IBD中的应用与机制,系统地综述了EcN的益生菌特性,概述EcN在临床实践中对IBD的治疗应用,同时从上皮屏障完整性调节、免疫调节、黏液屏障保护和肠道菌群稳态等方面阐述EcN治疗IBD的具体作用机制。此外,结合当前的研究现状,讨论了未来基于工程化EcN新疗法的发展前景,强调EcN在IBD治疗和预防中的适用性和可行性策略,并对该领域未来的研究方向进行了展望,为进一步深入研究EcN治疗IBD提供思路和参考。

采用干燥空气取代SF_(6)绝缘的中压充气柜,符合绿色环保发展方向,但空气的对流散热能力弱于SF_(6),这对充气柜的散热结构提出了更高要求。文中提出了一种采用纵向直线布置主回路的40.5 k V 2 500 A干燥空气绝缘充气柜总体结构,其具有主导电回路短、发热功率较低的特点。对该充气柜进行发热与散热分析、仿真和试验研究,发现采用常规结构气箱时,其内部空气温升达60 K左右,气箱内热量无法散到气箱外部是整体温升超标的主要因素。从增强对流散热和辐射散热方面对散热结构进行优化,重点设计了柜体侧板外表面的散热通道,优化后的方案通过了1.1×2 500 A温升试验,验证了设计和优化思路的可行性。

Identification of a major QTL for flag leaf glaucousness using a high-density SNP marker genetic map in hexaploid wheat

Cuticular wax plays an important role in protecting land plant against biotic and abiotic stresses. Cuticular wax production on plant surface is often visualized by a characteristic glaucous appearance. This study identified quantitative trait loci （QTLs） for wheat （Triticum aestivum L.） flag leaf glaucousness （FLG） using a high-density genetic linkage map developed from a recombinant inbred line （RIL） population derived from the cross HeynexLakin by single-seed descent. The map consisted of 2 068 single nucleotide polymorphism （SNP） markers and 157 simple sequence repeat （SSR） markers on all 21 wheat chromosomes and covered a genetic distance of 2 381.19 cM, with an average marker interval of 1.07 cM. Two additive QTLs for FLG were identified on chromosomes 3AL and 2DS with the increasing FLG allele contributed from Lakin. The major QTL on 3AL, ***-3AL, explained 17.5-37.8% of the phenotypic variation in different environments. ***-3AL was located in a 4.4-cM interval on chromosome 3AL that was flanked by two markers IWA1831 and IWA8374. Another QTL for FLG on 2DS, designated as ***-2DS which was identified only in Yangling in 2014 （YL14）, was flanked by IWA1939 and Xgwm261 and accounted for 11.3% of the phenotypic variation for FLG. ***- gr-3AL and ***-2DS showed Additive×Environment （AE） interactions, explaining 3.5 and 4.4% of the phenotypic variance, respectively. Our results indicated that different genes/QTLs may contribute different scores of FLG in a cultivar and that the environment may play a role in FLG.

散射是光学成像中普遍存在的现象,成像路径中存在的烟雾、水体、生物组织等散射介质导致光束发生随机散射效应,使得像面处目标信息以杂乱无章的散斑形式存在,如何应对散射介质对成像的限制是当前光学成像领域的研究热点。全息技术能够记录和重建物体全部信息,是获取和解译光场信息的有力工具之一。近年来,传统全息以及相关全息理论被推广应用至散射成像领域,取得了一系列突破性成果,文中主要介绍与归纳了散射成像领域中应用全息技术的理论原理、发展历史及最新进展,并展望其发展前景。
