Heme,an iron-containing porphyrin derivative,holds great promise in fields like medicine,food production and ***,we developed an engineered Corynebacterium glutamicum strain for efficient heme production by combining modular engineering and RBS *** whole heme biosynthetic pathway was methodically divided into 5-ALA synthetic module,uroporphyrinogen III(UPG III)synthetic module and heme synthetic module for further construction and *** heme synthetic modules were compared and the siroheme-dependent(SHD)pathway was identified to be optimal in *** for the first *** further improve heme production,the expression of genes in UPG III synthetic module and heme synthetic module was coordinated optimized through RBS engineering,***,heme oxygenase was knocked out to reduce heme *** engineered strain HS12.showed a maximum iron-containing porphyrin derivatives titer of 1592.mg/L with the extracellular secretion rate of 45.5%in fed-batch *** study constructed a *** chassis strain for efficient heme accumulation,which was beneficial for the advancement of efficient heme and other porphyrins production.

The exploitation of new green polymerization avenues for the effective synthesis of polymers by reversible-deactivation radical polymerization plays a critical role in pursuing the development of polymeric *** this work,serials of deep eutectic solvents(DES)with intermolecular-hydrogen-bonding interaction were constructed as catalysts and medium for actuating reversible complexation-mediated polymerization(RCMP)for the first time,yielding methacrylate polymers with high monomer conversion and narrow dispersion molecular weight in both water and oil *** mechanism and elementary reaction of RCMP were explored deeply,revealing that the complexation of initiator with DES to generate radicals was a ratecontrolling step and intermolecular-hydrogen-bond was primary factor to influence polymerization ***,the insights of density functional theory calculations revealed that negative electrostatic potential ensured nucleophilic *** investigation demonstrated the considerable potential of DES for RCMP,which is anticipated for other polymerization applications as a novel medium mode.

Salt caverns are extensively utilized for storing various substances such as fossil energy, hydrogen,compressed air, nuclear waste, and industrial solid waste. In China, when the salt cavern is leached through single-well water solution mining with oil as a cushion, engineering challenges arise with the leaching tubing, leading to issues like damage and instability. These problems significantly hinder the progress of cavern construction and the control of cavern shape. The primary cause of this is the flowinduced vibration instability of leaching tubing within a confined space, which results in severe bending or damage to the tubing. This study presents a model experimental investigation on the dynamic characteristics of leaching tubing using a self-developed liquid-solid coupling physical model experiment apparatus. The experiment utilizes a silicone-rubber pipe(SRP) and a polycarbonate pipe(PCP) to examine the effects of various factors on the dynamic stability of cantilevered pipes conveying fluid. These factors include external space constraint, flexural rigidity, medium outside the pipe, overhanging length, and end conditions. The experiments reveal four dynamic response phenomena: water hammer, static buckling, chaotic motion, and flutter instability. The study further demonstrates that the length of the external space constraint has a direct impact on the flutter critical flow velocity of the cantilevered pipe conveying fluid. Additionally, the flutter critical flow velocity is influenced by the end conditions and different external media.

Ocean is full of low-frequency,irregular,and widely distributed wave energy,which is suitable as the energy source for maritime Internet of Things(IoTs).Utilizing triboelectric nanogenerators(TENGs)to harvest ocean wave energy and power sensors is proven to be an effective ***,in random ocean waves,the irregular electrical energy output by general TENGs restricts the *** present,achieving regularized water wave energy harvesting relies on rather complex mechanical structure designs,which is not conducive to *** this work,we proposed a novel mechanical controlled TENG(MCTENG)with a simple controlled switch to realize the regularization *** structural parameters of the MC-TENG are optimized,and the optimal output voltage,output current,and transferred charge respectively reach 1684.2.V,85.4μA,and 389.9 nC,generating a peak power density of 38.46 W·m^(−3)·Hz^(−1).Under real water wave environment,the output of the MC-TENG is regularized and keeps stable regardless of any wave ***,the potential applications of the MC-TENG are demonstrated in powering environmental temperature,humidity,and wind speed *** work renders a simple approach to achieve effective regularized ocean wave energy harvesting,promoting the TENG industrialization toward practical application of maritime IoTs.

Objective Renal fibrosis is the ultimate pathway of various forms of acute and chronic kidney ***,the knockout of transient receptor potential channel 6(TRPC6)has shown promise in alleviating renal ***,the regulatory impact of TRPC6 on renal fibrosis remains *** In vivo,TRPC6 knockout(TRPC6−/−)mice and age-matched 12. SvEv(WT)mice underwent unilateral renal ischemia-reperfusion(uIR)injury surgery on the left renal pedicle or sham *** and serum were collected on days 7,14,2.,and 2. after *** vitro,primary tubular epithelial cells(PTECs)were isolated from TRPC6−/−and WT mice,followed by treatment with transforming growth factorβ1(TGFβ1)for 72.*** anti-fibrotic effect of TRPC6−/−and the underlying mechanisms were assessed through hematoxylin-eosin staining,Masson staining,immunostaining,qRT-PCR,and Western *** Increased TRPC6 expression was observed in uIR mice and PTECs treated with TGFβ***6−/−alleviated renal fibrosis by reducing the expression of fibrotic markers(Col-1,α-SMA,and vimentin),as well as decreasing the apoptosis and inflammation of PTECs during fibrotic progression both in vivo and in ***,we found that the phosphatidylinositol 3-kinase(PI3K)/protein kinase B(AKT)/glycogen synthase kinase 3 beta(GSK3β)signaling pathway,a pivotal player in renal fibrosis,was down-regulated following TRPC6 *** These results suggest that the ablation of TRPC6 may mitigate renal fibrosis by inhibiting the apoptosis and inflammation of PTECs through down-regulation of the PI3K/AKT/GSK3β*** TRPC6 could be a novel therapeutic strategy for preventing chronic kidney disease.

Methanol is one of the characteristic gases that distinguish between healthy individuals and lung cancer(LC)lesions in exhaled human *** concentration in exhaled breath is generally below one part per million(1×10^(-6)).The second-phase composite is widely regarded as one of the methods to improve the gas-sensing perfor-mance of metal oxide semiconductor(MOS)*** this study,LaF_(3)-Co_(3)O_(4) was synthesized by a simple hydrothermal method to enhance its low-concentration methanol gas-sensing performance.5 at%LaF_(3)-Co_(3)O_(4) nanorods exhibited excellent methanol detection performance,including a wider linear detection concentration range(0.2.10^(-6)~5×10^(-6)),a response value exceeding 4.0 for 1×10^(-6)methanol at 2.5°C and 75%relative humidity(RH),long-term stability(maximum deviation within 15%over 2.weeks),and excellent *** mechanism of performance enhancement was studied using various techniques,and density functional *** special spinel structure of Co_(3)O_(4),the high ionic migration of F-in LaF_(3),the larger specific surface area of 5 at%LaF_(3)-Co_(3)O_(4) nanorods,and the generated crystal defects all explain the excellentmethanol gas-sensing *** work provides a novel route to prepare MOS composite materials for low-concentration methanol gas detection.
