We aim to evaluate prostate health index as an additional risk-stratification tool in patients with Prostate Imaging Reporting and Data System score 3 lesions on multiparametric magnetic resonance *** with biochemical or clinical suspicion of having prostate cancer who underwent multiparametric magnetic resonance imaging in two tertiary centers(Queen Mary Hospital and Princess Margaret Hospital,Hong Kong,China)between January 2017 and June 2022 were ***-magnetic resonance imaging fusion biopsies were performed after prostate health index *** who only had Prostate Imaging Reporting and Data System score 3 lesions were further stratified into four prostate health index risk groups and the cancer detection rates were *** of the 747 patients,47.3%had Prostate Imaging Reporting and Data System score 3 lesions *** detection rate of clinically significant prostate cancer in this group was 15.0%.The cancer detection rates of clinically significant prostate cancer had statistically significant differences:5.3%in prostate health indexx 25.0-34.9,17.9%in prostate health index 35.0-54.9,and 52.6%in prostate health index≥55.0(Png the patients,26.9%could have avoided a biopsy with a prostate health indexxpense of a 5.3%risk of missing clinically significant prostate *** health index could be used as an additional risk stratification tool for patients with Prostate Imaging Reporting and Data System score 3 *** could be avoided in patients with low prostate health index,with a small risk of missing clinically significant prostate cancer.

Experimental Study on Impingement Processes of Fuel Sprays on Biomimetic Structured Surfaces

To improve the controllability of the wall-wetting process after the fuel spray-wall impingement in internal combustion engines,the methods of laser etching,chemical etching and surface free energy modification are used to prepare biomimetic structured surfaces with different *** impingement processes of diesel and n-butanol sprays on the walls under different conditions are experimentally *** the surface oleophilicity increases,the spreading radius of wall-impinging sprays *** about 5 s after the fuel injections,the fuel spray droplets hit the walls for the first time,and the secondary breakup and rebound *** mixture concentrations of different fuels hitting the various walls reach the peak *** a higher surface temperature,the peak value of the mixture concentration is mainly related to the heat flux to the fuel droplets in different boiling regimes from the metal *** concentration of the air–fuel mixture in the near wall region increases with increasing surface oleophilicity,increasing wall temperature and decreasing ambient *** with diesel,n-butanol presents a higher air–fuel mixture concentration in the near wall region.

Inhibition of viability of human retinal microvascular endothelial cells by vialinin A under high glucose condition

AIM:To investigate the effects of vialinin A on viability of human retinal endothelial cells(HRECs)under high glucose condition and its potential ***:The HRECs were divided into four groups:normal glucose control group(ng,5 mmol/L D-glucose),high glucose group(HG,30 mmol/L D-glucose),HG+1μmol/L vialinin A group,and HG+5μmol/L vialinin A *** cell viabilities were measured with cell counting kit-8(CCK-8)assay for proliferation,with scratch assay for migration,and tube formation,for evaluation of the impact of vialinin A on cellular ***-time PCR and Western blotting were used to determine the expression level of vascular endothelial growth factor(VEGF).RESULTS:The proliferative capacity and migration of HRECs was reduced by 5μmol/L vialinin A in high glucose environment(both Pxpression level of VEGF and PI3K in HRECs was also significantly decreased by vialinin A(Pngiogenic therapy.

TRAIL and Celastrol Combinational Treatment Suppresses Proliferation, Migration, and Invasion of Human Glioblastoma Cells via Targeting Wnt/β-catenin Signaling Pathway

Objective To investigate the mechanistic basis for the anti-proliferation and anti-invasion effect of tumor necrosis factor-related apoptosis-induced ligand(TRAIL)and celastrol combination treatment(TCCT)in glioblastoma *** Cell counting kit-8 was used to detect the effects of different concentrations of celastrol(0-16µmol/L)and TRAIL(0-500 ng/mL)on the cell viability of glioblastoma cells.U87 cells were randomly divided into 4 groups,namely control,TRAIL(TRAIL 100 ng/mL),Cel(celastrol 0.5µmol/L)and TCCT(TRAIL 100 ng/mL+celastrol 0.5µmol/L).Cell proliferation,migration,and invasion were detected by colony formation,wound healing,and Transwell assays,*** reverse transcription polymerase chain reaction and Western blotting were performed to assess the levels of epithelial-mesenchymal transition(EMT)markers(zona occludens,N-cadherin,vimentin,zinc finger E-box-binding homeobox,Slug,and β-catenin).Wnt pathway was activated by lithium chloride(LiCl,20 mol/L)and the mechanism for action of TCCT was *** Celastrol and TRAIL synergistically inhibited the proliferation,migration,invasion,and EMT of U87 cells(Pxpression of GSK-3β and down-regulated the expression of β-catenin and its associated proteins(Png c-Myc,Cyclin-D1,and matrix metalloproteinase(MMP)-*** addition,LiCl,an activator of the Wnt signaling pathway,restored the inhibitory effects of TCCT on the expression of β-catenin and its downstream genes,as well as the migration and invasion of glioblastoma cells(P<0.05 or P<0.01).Conclusions Celastrol and TRAIL can synergistically suppress glioblastoma cell migration,invasion,and EMT,potentially through inhibition of Wnt/β-catenin *** underlies a novel mechanism of action for TCCT as an effective therapy for glioblastoma.

Efficacy and safety of xileisan combined with mesalazine for ulcerative colitis:A meta-analysis and trial sequential analysis

BACKGROUND The benefits and risks of xileisan(xLS)in the treatment of ulcerative colitis(UC)remain *** The present study aimed to evaluate the efficacy and safety of the combination of xLS and mesalazine when treating *** We searched eight databases for clinical trials evaluating the combination of xLS and mesalazine in the treatment of UC,up to January ***-analysis and trial sequential analysis(TSA)were performed using Revman 5.3 and TSA 0.***.5.10 beta,*** The present study included 13 clinical studies involving 990 patients,of which 501 patients received xLS combined with mesalazine while 489 patients received mesalazine *** meta-analysis showed that,in terms of efficacy,the combination of xLS and mesalazine significantly improved the clinical efficacy rate by 22%[risk ratio(RR)=1.22;95%CI:1.15–1.28;Png the duration of abdominal pain by 2.25 days[mean difference(MD)=-2.25;95%CI:-3.35 to-1.14;Png/mL(MD=-16.25;95%CI:-20.48 to-12.01;Png/mL(MD=-14.14;95%CI:-24.89 to-3.39;P=0.01).The TSA indicated conclusiveness in the meta-analysis of the efficacy *** terms of safety,the meta-analysis revealed that the combination of xLS and mesalazine did not increase the occurrence of total and gastrointestinal adverse events,abdominal distension,and erythema(P>0.05).The TSA showed non conclusive findings in the meta-analysis of the safety ***’s test showed no publication bias(P=0.734).CONCLUSION Treatment with xLS alleviated the clinical symptoms,intestinal mucosal injury,and inflammatory response in patients with UC,while demonstrating good safety.

煤炭工业园区多环芳烃多介质污染特征和风险

以煤炭为中心的工业活动过程中会向环境中排放大量多环芳烃(PAHs),污染土壤、水体和农作物,对人体健康造成潜在威胁。该文以山西省长治市某典型煤炭工业园区为研究区,分别在园区中心及其上下风向2、5、8km处采集饮用水、室内外灰尘和谷物样品,采用色谱-质谱仪(GS-MS)测定样品中PAHs的浓度,分析PAHs的多介质污染特征并进行健康风险评价。结果表明:研究区7个样点饮用水中∑PAHs的检出范围为233.57~337.05ng/L,符合《生活饮用水卫生标准》(GB5749-2022)中多环芳烃(总量)在2×10^(3)ng/L的限值,随着距园区中心距离的增加,上风向和下风向饮用水中∑PAHs都呈现先上升后下降的趋势;小米和面粉中∑PAHs的检出范围分别为113.70~2318.57 ng/g和108.42~512.30 ng/g,且随着距离增加,上风向和下风向谷物(小米和面粉)中∑PAHs都呈现“N”趋势;室内灰尘中∑PAHs的浓度高于室外灰尘,随着距离增加,室外灰尘中∑PAHs在上风向和下风向都呈先下降后上升的趋势。健康风险评估显示,研究区饮用水中PAHs的致癌风险指数处于10^(-6)~10^(-4)范围内,具有潜在致癌风险,且成人的致癌风险高于儿童;灰尘中PAHs对人体具有强致癌风险,且室内外灰尘对儿童、成人的致癌风险均呈现出室内>室外、成人>儿童的特征;小米和面粉对人体的致癌风险均属于高致癌风险。

A novel CARD containing splice-isoform of CIITA regulates nitric oxide synthesis in dendritic cells

MHC class II expression is controlled mainly at transcriptional level by class II transactivator(CIITA),which is a non-DNA binding coactivator and serves as a master control factor for MHC class II genes ***,we describe the function of a novel splice-isoform of CIITA,DC-expressed caspase inhibitory isoform of CIITA(or DC-CASPIC),and we show that the expression of DCCASPIC in DC is upregulated upon lipopolysaccharides(LPS)***-CASPIC localizes to mitochondria,and protein-protein interaction study demonstrates that DC-CASPIC interacts with caspases and inhibits its activity in ***,DC-CASPIC suppresses caspases-induced degradation of nitric oxide synthase-2(NOS2)and subsequently promotes the synthesis of nitric oxide(NO).NO is an essential regulatory molecule that modulates the capability of DC in stimulating T cell proliferation/activation in vitro;hence,overexpression of DC-CASPIC in DC enhances this ***,our findings reveal that DC-CASPIC is a key molecule that regulates caspases activity and NO synthesis in DC.

血清β-半乳糖凝集素-3在非创伤股骨头坏死中的意义

[目的]本研究旨在探讨血清β-半乳糖凝集素-3(lectin galactoside-binding soluble 3,LGALS3)水平在非创伤性股骨头坏死(non-traumatic osteonecrosis of the femoral head,NONFH)诊断中的临床价值。[方法]2023年4月—2023年9月纳入NONFH患者84例为坏死组,选取同期检者健康人78例为对照组。采用酶联免疫吸附法检测两组血清中LGALS3的水平,并收集相关临床资料,包括性别、年龄、体重指数(body mass index,BMI)、吸烟史、NONFH病因、受累侧数、疼痛视觉模拟评分(visual analogue scale,VAS)、Harris评分、坏死程度及ARCO分期。对两组LGALS3浓度及其与各临床参数的关系进行比较分析。[结果]坏死组血清中LGALS3浓度显著高于正常人组[(9.8±7.7)ng/ml vs(4.2±4.1)ng/ml,Png/ml vs(5.3±3.3)ng/ml,Png/ml vs(3.1±1.2)ng/ml,Png/ml vs(4.1±1.3)ng/ml vs(11.0±5.8)ng/ml vs(16.8±8.2)ng/ml,P<0.001]。NONFH患者的LGALS3浓度与VAS评分呈显著正相关(r=0.843,P<0.001),与Harris评分呈显著负相关(r=-0.710,P<0.001),与ARCO分期呈显著正相关(r=0.822,P<0.001)。ROC分析表明,LGALS3浓度预测是否NONFH的曲线下面积(area under curve,AUC)为0.769。[结论]血清中的LGALS3水平可反映NONFH病情的严重程度,并可能作为该病早期诊断的潜在生物标志物。
