Objective: The purpose of this research was to examine the potential anticancer properties of Xiao Tan San Jie fang(XTSJF) and its potential mechanism of action against colorectal cancer. Materials and Methods: HCT116 cells were induced into HCT116 spheres in DMEM/F12 medium by treatment with epidermal growth factor + fibroblast growth factor + leukemia inhibitory factor + B27. The proliferation ability and stemness of HCT116 spheres was examined. Various concentrations of XTSJF were used to treat HCT116 spheres to observe the impact on proliferation, apoptosis, and expression of stem cell markers. Next, Wnt/β-catenin pathway-related factor proteins were detected. Results: The findings revealed that XTSJF suppressed cell growth and induced cell death in HCT116 cells in a dosage-dependent manner. Similarly, XTSJF promotes apoptosis, inhibits cell proliferation, prolongs survival, and maintains the expression of stem cells through the Wnt/-catenin/TCF4 axis. XTSJF also inhibits AKT activity and subsequently activates glycogen synthesis kinase-3β expression, inhibiting Wnt/beta-catenin pathway activity and downstream target gene transcript expression. The Wnt/β-catenin signaling pathway is inhibited by the XTSJF, leading to the suppression of colon cancer stem cell proliferation. Conclusion: Xiaotan Sanjie Prescription inhibited colon cancer stem cell growth through Wnt/β-catenin signaling pathway.

We present a systematic scheme to achieve both high birefringence and low confinement loss in index-guiding photonic crystal fibers(PCFs) ,using a structurally-simple PCF with finite number of air holes in the cladding region. By increasing the size of the outermost-ring air holes in the cladding region,highly birefringent PCFs with low confinement loss can be successfully achieved. The design strategy is based on the fact that the modal birefringence of PCFs is dominated by the inner-ring air holes in PCF,which is verified by a full-vector finite element method with anisotropic perfectly matched layers. Numerical results show that modal birefringence in the order of 10-3 and confinement loss less than 0.1 dB/km can be easily realized in the proposed PCF with only four rings of air holes in the cladding region. We expect that such fibers will be much easier to be fabvicated than those with more air holes in the cladding region.

Statistic Analysis on Quantitative Characteristics for Developing the DUS Test Guideline of Ranunculus asiaticus L.

Selection of quantitative characteristics, division of their expression ranges, and selection of example varieties are key issues on developing DUS Test Guidelines, which are more crucial for quantitative characteristics since their expressions vary in different degrees. Taking the development of DUS Test Guideline of Ranunculus asiaticus L. as an example, this paper applied statistic-based approaches for the analyses of quantitative characteristics. We selected 9 quantitative characteristics from 18 pre-selected characteristics, based on within-variety uniformity, stability between different growing cycles, and correlation among characteristics, by the analyses of coefficient of variation, paired-samples t-test and partial correlation. The expression ranges of the 9 selected quantitative characteristics were divided into different states using descriptive statistics and distribution frequency of varieties. Eight of the 9 selected quantitative characteristics were categorized as standard characteristics as they showed one peak in distribution frequency of 120 varieties in various expressions of the characteristics, whereas, plant height can be categorized as grouping characteristic since it gave two peaks, and can group the varieties into pot and cut varieties. Finally, box-plot was applied to visually select the example varieties, and varieties 7, 12, and 28 were determined as the example varieties for plant height. The methods described in this paper are effective for the selection of quantitative characteristics, division of expression ranges, and selection of example varieties in Ranunculus asiaticus L. for DUS test, and may also be interest for other plant genera.

Problems associated with organochlorine pesticide （OCP）-contaminated sites in China have received wide attention. To solve such problems, innovative ex-situ methods of site remediation are urgently needed. We investigated the feasibility of the extraction method with different organic solvents, ethanol, 1-propanol, and three fractions of petroleum ether, using a soil collected from Wujiang （W J）, China, a region with long-term contamination of dichlorodiphenyltrichloroethanes （DDTs）. We evaluated different influential factors, including organic solvent concentration, washing time, mixing speed, solutiomto-soil ratio, and washing temperature, on the removal of DDTs from the WJ soil. A set of relatively better parameters were selected for extraction with 100 mL L-1 petroleum ether （60-90 ℃）： washing time of 180 min, mixing speed of 100 r min-I, solution-to-soil ratio of 10：1, and washing temperature of 50 ℃. These selected parameters were also applied on three other seriously OCP-polluted soils. Results demonstrated their broad-spectrum effectiveness and excellent OCP extraction performance on the contaminated soils with different characteristics.
