BACKGROUND: Kang-Lai-Te (KLT) is extracted from the traditional Chinese herbal medicine Semen Coicis, which has been used in China as an effective clinical drug for over a thousand years. It contains numerous ingredients with anti-tumor effects. In our previous studies on transplanted hepatomas in rats, KLT could stop the cells in the G2+M stage of cell cycle and then reduce the number of cells entering the stage G0 and G1, but the mechanism of the anti-proliferative effect was unknown. In this experiment, we examined whether KLT inhibits HepG2 cell growth, if so, tried to explore its mechanism. METHODS: KLT at different concentrations was used for the treatment of hepatocellular carcinoma cells in vitro, respectively. The proliferation inhibitory rate was evaluated by MTT assay, induction of cell apoptosis rate and the protein levels of Fas and Fas ligand (FasL) were determined by flow cytometry (FCM), and the expression of Fas and FasL mRNA was detected by real-time fluorescent quantitative RT-PCR. RESULTS: KLT produced an obvious time and dose-dependent inhibitory effect on HepG2 cells, and marked apoptosis was detected by FCM The protein of Fas increased by 11.01%, 18.71%, 28.71% and 37.15%; the protein of FasL increased by 1.49%, 1.91%, 3.27% and 3.38% in comparison with the control (Pliary Pancreat Dis Int 2009; 8: 267-272)

Any regulatory policy is formed by two interacting forces-the interest groups who are the demanders for the policy and the government who is the policy supplier. In most researches, people pay too much attentions to the interest groups,but neglect the behaviors of the government. Directing at this weakness, this paper integrates the two forces of demander and supplier together and erects a comprehensive model, through which we find out the decision of regulatory policy is determined by the government comparison between the utilities of equality and the net gains (or loss)of the interest groups.

BACKGROUND: Transforming growth factors (TGF)-beta 1, TGF-beta R2 and Smad4 belong to the TGF family, and play important roles in carcinogenesis and the development of carcinoma, especially hepatocellular carcinoma (HCC). TGF-beta 1 is a multipotent polypeptide, which inhibits the growth of epithelial cells including hepatoma cell lines and hepatocytes by inducing apoptosis. TGF-beta R2 forms a heterodimeric complex upon binding to TGF-beta, and then generates the first step in the signal transduction pathway leading to growth inhibition in coordination with the type I receptor. Smad4 protein is an important mediator in the TGF-beta signaling pathway, and negatively regulates the growth of epithelial cells. This study aimed to detect the expression of TGF-beta 1, TGF-beta R2 and Smad4 in HCCs and their adjacent normal tissues, while assessing its relations with the clinicopathological parameters of HCC. METHODS: Forty-seven HCC specimens and their adjacent normal tissues were obtained surgically at the Affiliated Hospital of Medical College, Qingdao University. The expression of TGF-beta 1, TGF-beta R2 and Smad4 was separately detected by immunohistochemistry in all HCC specimens and their adjacent normal tissues, and its relations with the clinicopathological parameters of HCC were assessed. RESULTS: The positive expression of TGF-beta 1 was 72.34% in the HCC specimens, which was higher than that in the adjacent normal tissues (P < 0.001). The positive expression of Smad4 and TGF-beta R2 was 34.04% and 59.57% respectively in the carcinoma specimens. The expression of TGF-beta 1, TGF-beta R2 and Smad4 was significantly higher in groups with a tumor embolus of the portal vein, integrity of the amicula, and Edmondson's III-IV than that in other groups, but it was not related to tumor size (P < 0.05). CONCLUSIONS: TGF-beta 1 may play an important role in the occurrence and development of HCC. Combined detection of TGF-beta 1, TGF-beta R2 and Smad4 may be useful for the

Objective： TO examine the effects of the combination of quercetin （Q）, cinnamaldehyde （C） and hirudin （H）, a Chinese medicine formula on high glucose （HG）-induced apoptosis of cultured dorsal root ganglion （DRG） neurons. Methods： DRG neurons exposed to HG （45 mmol/L） for 24 h were employed as an in vitro model of diabetic neuropathy. Cell viability, reactive oxygen species （ROS） level and apoptosis were determined. The expression of nuclear factor of Kappa B （NF- κB）, inhibitory kappa B α （IκBα ）, phosphorylated IκBα and Nf-E2 related factor 2 （Nrf2） were examined using reverse transcription-polymerase chain reaction （RT-PCR） and Western blot assay. The expression of hemeoxygenase-1 （HO-1）, interleukin-6 （IL-6）, tumor necrosis factor （TNF-α） and caspase-3 were also examined by RT-PCR and Western blot assay. Results： HG treatment markedly increased DRG neuron apoptosis via increasing intracellular ROS level and activating the NF- κB signaling pathway （P〈0.05）. Co-treatment with Q, C, H and their combination decreased HG-induced caspase-3 activation and apoptosis （P〈0.05 or P〈0.01）. The expressions of NF- κB, IL-6 and TNF-α were down- regulated, and Nrf2/HO-1 expression was up-regulated （P〈0.05 or P〈0.01）. QCH has better effect in scavenging ROS, activating Nrf-2/HO-1, and down-regulating the NF- κB pathway than other treatment group. Conclusions： DRG neurons＇ apoptosis was increased in diabetic conditions, which was reduced by QCH formula treatment. The possible reason could be activating Nrf-2/HO-1 pathway, scavenging ROS, and inhibition of NF- κB activation. The effect of QCH combination was better than each monomer or the combination of the two monomers.

BACKGROUND: Research has revealed a shift towards Th2 in many types of malignant tumor, but the state of Th1/Th2 is not clear in patients with primary hepatic cancer (PHC). This study was designed to determine the expression of Th1- versus Th2-type cytokines in primary hepatic cancer and the adjacent liver tissue in order to provide evidence for treatment of the Th1/Th2 shift. METHODS: Samples were collected from 11 patients with PHC. The gene expression of Th1/Th2 cytokines was detected by reverse transcriptase polymerase chain reaction (RT-PCR) using IFN-gamma and IL-2 as Th1-type cytokine genes, and IL-4 and IL-10 as Th2-type cytokine genes. RESULTS: Th1-type cytokines were expressed in 7/11 PHCs and 9/11 adjacent liver tissues, while Th0 type cytokines occurred in 4/11 PHCs and 2/11 adjacent liver tissues. CONCLUSION: Th1-type cytokines are expressed predominantly in primary hepatic cancer and the adjacent liver tissue.

Organic redox compounds are attractive cathode materials in aqueous zinc-ion batteries owing to their low cost,environmental friendliness,multiple-electron-transfer reactions,and resource ***,the realized energy density is constrained by the limited capacity and low ***,copper-tetracyanoquinodimethane(CuTCNQ),an organic charge-transfer complex is evaluated as a zinc-ion battery cathode owing to the good electron acceptation ability in the cyano groups that improves the voltage *** electrochemical activation,electrolyte optimization,and adoption of graphene-based separator,CuTCNQ-based aqueous zinc-ion batteries deliver much improved rate performance and cycling stability with anti-self-discharge *** structural evolution of CuTCNQ during discharge/charge are investigated by ex situ Fourier transform infra-red(FT-IR)spectra,ex situ X-ray photoelectron spectroscopy(XPS),and in situ ultraviolet visible spectroscopy(UV-vis),revealing reversible redox reactions in both cuprous cations(Cu^(+))and organic anions(TCNQ^(x-1)),thus delivering a high voltage output of 1.0 V and excellent discharge capacity of 158 mAh g^(-1).The remarkable electrochemical performance in Zn//CuTCNQ is ascribed to the strong inductive effect of cyano groups in CuTCNQ that elevated the voltage output and the graphene-modified separator that inhibited CuTCNQ dissolution and shuttle effect in aqueous electrolytes.

Inspired by the dynamic wet adhesive systems in nature,various artificial adhesive surfaces have been developed but still face different ***,the theoretical mechanics of wet adhesives has never been sufficiently ***,we develop a novel adhesive mechanism for governing wet adhesion and investigate the biological models of honeybee arolium for reproducing the natural wet adhesive ***-nano structures of honeybee arolium and arolium-prints were observed by Cryogenic scanning electron microscopy(Cryo-SEM),and the air pockets were found in the contact interface ***,the adhesive models with a three-phase composite interface(including air pockets,liquid secretion,and hexagonal frames of arolium),were formed to analyze the wet adhesion of honeybee *** results of theoretical calculations and experiments indicated an enhanced adhesive mechanism of the honeybee by liquid self-sucking effects and air-embolism *** these effects,normal and shear adhesion can be adjusted by controlling the proportion of liquid secretion and air pockets in the contact ***,the air-embolism effects contribute to the optimal coupling of smaller normal adhesion with greater shear adhesion,which is beneficial for the high stride frequency of *** works can provide a fresh perspective on the development of bio-inspired wet adhesive surfaces.
