Regulatory role of electroacupuncture on satellite glial cell activity in the colon and dorsal root ganglion of rats with irritable bowel syndrome

OBJECTIVE: To investigate the role of satellite glial cells in irritable bowel syndrome(IBS) and the effect of electroacupuncture(EA) at the Tianshu(ST25) and Shangjuxu(ST37) combination. METHODS: A model for visceral hypersensitivity in IBS was induced through colorectal distension(CRD) stimulation. Clean-grade male Sprague-Dawley(SD) rats were randomly divided into four groups: a normal group(NG), a model group(MG), an electroacupuncture group(EA), and a glial cell inhibitor group(FCA). Bilateral EA(2/100 Hz, 1 mA, 30 min) was administered at the Tianshu(ST25) and Shangjuxu(ST37) in week 6. Abdominal withdrawal reflex(AWR) scores were used to assess the behavioral response associated with visceral hyperalgesia, while hematoxylin-eosin staining was employed to evaluate pathological changes in the colon. The protein and mRNA levels of glial fibrillary acidic protein(GFAP) in the colon and colon-related dorsal root ganglion(DRG) were analyzed using immun-ofluorescence, immunohistochemistry, Western blotting, real-time polymerase chain reaction. The impact of EA on electrophysiological properties of colon-related DRG neurons was observed through whole-cell patch clamp analysis. RESULTS: EA significantly reduced the visceral pain behavior scores in rats with IBS in response to graded(20, 40, 60, 80 mm Hg) CRD stimulation. Additionally, EA downregulated the protein and mRNA expression levels of GFAP in the colon and colon-related DRG of rats with IBS. EA also regulated the resting membrane potential, rheobase and action potential of colon-related DRG neurons in rats with IBS. CONCLUSIONS: EA can regulate the excitatory properties of colon-related DRG neurons by downregulating the protein and mRNA expression of GFAP in the colon and colon-related DRG, indicating a potential neurobiological mechanism by which EA relieves visceral hypersensitivity in rats with IBS.

Moxibustion Moxibustion(written as“jiū(灸)”in Chinese),also known as“jiūruò(灸焫)”in ancient Chinese,was described in the ancient Chinese book Shuo Wen Jie Zi as having the meaning of burning and possessing the properties of fire,while sharing the same pronunciation as“jiǔ(久).”“Moxibustion is a method of curing diseases that uses lighted moxa to press and cauterize acupoints.”Moxibustion is a traditional external treatment method that involves the application of moxa wool or other materials,either directly or indirectly(such as through burning or fumigation),to stimulate specific points or areas on body *** is used for purposes such as prevention,treatment,rehabilitation,and health *** originated in ancient times,prevailed during the Qin and Han dynasties(221 B.C.E.–220 C.E.),experienced a period of decline during the Ming and Qing dynasties(1368–1912 C.E.),and was revitalized in the contemporary era.

Influence of herb-partitioned moxibustion at Qihai (CV6) and bilateral Tianshu (ST25) and Shangjuxu (ST37) acupoints on toll-like receptors 4 signaling pathways in patients with ulcerative colitis

OBJECTIVE: To evaluate the efficacy of herb-partitioned moxibustion(HPM) at Qihai(CV6), Tianshu(ST25) and Shangjuxu(ST37) acupoints in relieving symptoms and the immune regulation of HPM on the toll-like receptors 4(TLR4) signaling pathway in ulcerative colitis(UC) ***: A randomized, single-blind study was conducted 63 patients to receive HPM or sham HPM treatment. The efficacy outcomes included scores of the Mayo, Baron, inflammatory bowel disease questionnaire(IBDQ), self-rating depression scale(SDS), self-rating anxiety scale(SAS). HE staining was used to observe the histopathological changes of the colon. The expression of inflammatory cytokines and TLR4 signaling pathway related molecules were determined by enzyme-linked immunosorbent assay and ***: Baron, SDS, SAS scores were significantly decreased in moxibustion group(P < 0.05), IBDQ score was significantly greater in the moxibustion group than in the sham moxibustion group(P < 0.05). Histopathology of mucosal biopsies showed that both two groups improved in mucosa after treatment. The expression levels of tumor necrosis factor-α, interleukin-2, interleukin-12, interferon-γ, and TLR4, lipopolysaccharide, myeloid differentiation factor 88, interleukin receptor associated kinase, tumor necrosis factor receptor associated factor 6 and nuclear factor kappa-B p65 were significantly lower in the moxibustion group than in the sham moxibustion group(P < 0.05).CONCLUSION: This study showed that HPM at Qihai(CV6), Tianshu(ST25) and Shangjuxu(ST37) acupoints is effective to relieve symptoms, anxiety, depression and improving life quality in UC patients,which may be related to the immune regulation of HPM on TLR4 signaling pathway.

OBJECTIVE:To observe the effect of herb-partitioned moxibustion(HPM)on the miRNA expression profile of thyroid tissue in experimental autoimmune thyroiditis(EAT)***:Rats were randomly divided into normal control(NC)group,EAT model(EAT)group,HPM group and western medicine(Med)*** model rats were prepared by a combined immunization with complete and incomplete Freund’s adjuvant emulsified with porcine thyroglobulin and *** in the HPM group were treated with HPM,while rats in the Med group were treated with levothyrocine(1μg/2 m L)by *** staining was used to observe the pathological morphological changes of thyroid tissue,ELISAs was uaed to detect the serum concentrations of TGAb,TPOAb,FT3,FT4,*** then performed high-throughput mi RNA sequencing to analyse the mi RNA expression profiles in the thyroid tissues,followed by a bioinformatics ***-q PCR was used to verify the identified differentially expressed mi ***:HPM improved the thyroid tissue morphology and reduced serum TPOAb,TGAb,TSH concentration in EAT rats(P<0.05),but with no obvious effect on FT3 and FT4 *** the TSH,FT3 and FT4 concentration was significantly changed in the Med group(P<0.01 or P<0.05)compared with that of EAT *** results showed that a total of 17 mi RNAs were upregulated,and 4 were downregulated in the EAT rats,in which the expression levels of mi R-346 and mi R-331-5 p were reversed by *** target genes of the mi RNAs that regulated by HPM wereassociated with a variety of immune factors and immune ***-q PCR verification showed that the expression of mi RNA-346 and mi RNA-331-5 p was consistent with the sequencing ***:HPM could regulate the the expression of mi RNA-346 and mi RNA-331-5 p,then act on their target genes to immune and inflammation-related pathways,which may be one of the mechanisms of HPM on EAT rats.

目的:从中性粒细胞胞外诱捕网(NETs)参与克罗恩病(CD)进展的角度,阐释隔药灸减轻CD大鼠肠道炎症的效应机制。方法:将大鼠随机分为正常组、模型组、隔药灸组和美沙拉嗪组。除正常组外,其余大鼠采用2,4,6-三硝基苯磺酸制备CD模型。正常组和模型组不治疗仅做与其他组相同的固定。隔药灸组予以隔药灸气海和双侧天枢穴。美沙拉嗪组接受美沙拉嗪肠溶片灌胃治疗。比较各组大鼠一般情况,苏木精-伊红染色观察结肠组织病理学变化并评分。采用ABC酶联免疫吸附测定法检测各组大鼠血清中性粒细胞胞外诱捕网DNA(NETs-DNA)、中性粒细胞弹性蛋白酶DNA(NE-DNA)、过氧化物酶DNA(MPO-DNA)浓度,采用免疫荧光和实时定量聚合酶链反应技术观察各组大鼠结肠组织瓜氨酸化组蛋白3(citH3)、MPO和NE蛋白和mRNA的表达。结果:与正常组比较,模型组结肠黏膜溃疡深达肌层,上皮不完整,杯状细胞明显减少,可见大量炎性细胞浸润,结肠黏膜损伤指数(CMDI)评分显著增加(P<0.01),模型组血清NETs-DNA、NE-DNA、MPO-DNA浓度均增加(P<0.05),模型组结肠组织NE、citH3和MPO蛋白和mRNA表达均上调(P<0.01或P<0.05);与模型组相比,隔药灸组和美沙拉嗪组结肠黏膜上皮修复,杯状细胞明显增多,可见少量炎性细胞浸润,CMDI评分明显降低(P<0.01),隔药灸组和美沙拉嗪组血清NETs-DNA、NE-DNA、MPO-DNA浓度均下降(P<0.05),隔药灸组和美沙拉嗪组NE、citH3和MPO蛋白和mRNA表达下调(P<0.01或P<0.05)。结论:隔药灸能降低血清中性粒细胞NETs复合物的浓度,抑制结肠组织中的NETs复合物的蛋白和mRNA表达,可能是隔药灸缓解CD结肠黏膜炎症的机制之一。

Effects of zeolite imidazole frameworks on rice seedlings (Oryza sativa L.): Phytotoxicity, transformation, and bioaccumulation

Zeolite imidazole frameworks(ZIFs),a class of the metal organic framework,have been extensively studied in environmental ***,their environmental fate and potential ecological impact on plants remain ***,we investigated the phytotoxicity,transformation,and bioaccumulation processes of two typical ZIFs(ZIF-8 and ZIF-67)in rice(Oryza sativa L.)under hydroponic ***-8 and ZIF-67 in the concentration of 50 mg/L decreased root and shoot dry weight maximally by 55.2%and 27.5%,53.5%and 37.5%,*** scanning electron microscopy(SEM)imaging combined with X-ray diffraction(XRD)patterns revealed that ZIFs on the root surface gradually collapsed and transformed into nanosheets with increasing cultivation *** fluorescein isothiocyanate(FITC)labeled ZIFs were applied to trace the uptake and translocation of ZIFs in *** results demonstrated that the transformed ZIFs were mainly distributed in the intercellular spaces of rice root,while they cannot be transported to culms and *** so,the Co and Zn contents of rice roots and shoots in the ZIFs treated groups were increased by 1145%and 1259%,145%and 259%,respectively,compared with the control *** findings suggested that the phytotoxicity of ZIFs are primarily attributed to the transformed ZIFs and to a less extent,the metal ions and their ligands,and they were internalized by rice root and increased the Co and Zn contents of *** study reported the transformation of ZIFs and their biological effectiveness in rice,highlighting the potential environmental hazards and risks of ZIFs to crop plants.

Effects of electroacupuncture and Chinese kidney-nourishing medicine on polycystic ovary syndrome in obese patients

OBJECTIVE： To explore the effect of electroacu- puncture and Chinese kidney-nourishing medicine on insulin （INS）, adiponectin （APN）, leptin （LEP）, and glucolipid metabolism of obese patients with polycystic ovary syndrome （PCOS）. METHODS： Sixty-seven obese PCOS patients were randomly divided into two groups. Thirty-three pa- tients in the acupuncture-medicine group were treated three times a week with electroacupunc- ture at the Tianshu （ST 25）, Zhongwan （CV ] 2）, Qi- hai （CV 6）, Sanyinjiao （SP 6）, Geshu （BL 17）, and Cil-iao （BL 32） acupoints. They drug, Tiankui capsule, for 3 treatment. Point-taking and also took the Chinese months as a course of treatment in the elec- troacupuncture group of 34 patients was the same as those in the acupuncture-medicine group. We observed and compared the changes in the obesi- ty-related indexes of body weight （BW）, body mass index （BMI）, and waist-hip ratio （WHR）, as well as fasting plasma glucose （FPG）, fasting insulin （FINS）, APN, and LER RESULTS： BW, BMI, WHR, and FINS decreased and insulin sensitivity index （ISI） and APN were higher in the acupuncture-medicine group than in the electroacupuncture group （P〈0.01）. There was no obvious difference in LEP between the two groups （P〉0.05）. CONCLUSION： Acupuncture combined with medi- cine is better than just electroacupuncture for obese PCOS patients by improving obesity-related indexes, insulin sensitivity, and APN level. This indi- cates that acupuncture-medicine therapy is worth clinical popularization.

Cadmium(Cd)pollution poses a serious threat to plant growth and *** have shown great application potential for alleviation of Cd toxicity to *** this study,we applied graphitic carbon nitride nanosheets(g-C_(3)N_(4)NSs)for alleviation of Cd-toxicity to soybean(Glycine max L.).The g-C_(3)N_(4)NSs supplementation significantly improved plant growth and reduced oxidative damage in the Cd-toxicated soybean seedlings through hydroponic ***,the g-C_(3)N_(4)NSs dynamically regulated the root cell wall(RCW)components by increasing pectin content and modifying its demethylation via enhancing pectin methylesterase(PME)activity,therefore greatly enhanced stronger RCW-Cd retention(up to 82.8%)and reduced Cd migration to the ***,the g-C_(3)N_(4)NSs reversed the Cd-induced chlorosis,increased photosynthetic efficiency because of enhancement in F_v/F_mration,Y(Ⅱ)and sugars *** results provide new insights into the alleviation of Cd toxicity to plants by g-C_(3)N_(4)NSs,and shed light on the application of low-cost and environmental-friendly carbon-based NMs for alleviating heavy metal toxicity to plants.

Effects of moxibustion on heat-shock protein 70 expression in the spinal cord and colonic mucosa in a rat model of ulcerative colitis

Pathological changes in the colon are closely associated with the spinal cord, and innervation of spinal cord can regulate cellular functions. Our previous studies verified that moxibustion protects and restores the colonic mucosa, but the mechanisms of action remain unknown. The present study observed the effects of moxibustion and salicylazosulfapyridine on expression of heat-shock protein 70 （HSP70） and its mRNA in the spinal cord and colonic mucosa of ulcerative colitis rats. Results demonstrated that moxibustion and salicylazosulfapyridine increased HSP70 mRNA expression in the spinal cord and colonic mucosa of ulcerative colitis rats. The decreased transcriptional activity of HSP70 in the spinal cord and colonic mucosa might participate in damage to the colonic mucosa in ulcerative colitis rats. Moxibustion exerted protective effects on colonic mucosa by up-regulating HSP70 transcriptional activity in the spinal cord and colonic mucosa.
