Alveolar macrophage phagocytosis-evading inhaled microgels incorporating nintedanib-PLGA nanoparticles and pirfenidone-liposomes for improved treatment of pulmonary fibrosis

Idiopathic pulmonary fibrosis(IPF)is a chronic inflammatory and fibrotic response-driven lung disease that is difficult to cure because it manifests excessive profibrotic cytokines(e.g.,TGF-β),activated myofibroblasts,and accumulated extracellular matrix(ECM).In an attempt to develop an inhalation formulation with enhanced antifibrotic efficacy,we sought to fabricate unique aerosolizable inhaled microgels(μGel)that contain nintedanib-poly(lactic-co-glycolic acid)(PLGA)nanoparticles(NPs;n-PN)and pirfenidone-liposomes(p-LP).The aero-μGel was~12μm,resisted phagocytosis by alveolar macrophages in vitro and in vivo,and protected inner-entrapped n-PN and *** n-PN/p-LP@aero-μGel caused enhanced/extended antifibrotic efficacy in a bleomycin-induced pulmonary fibrosis mouse presumably due to prolonged lung ***,the results obtained by intratracheal aerosol insufflation of our n-PN/p-LP@aero-μGel twice a week were much better than those by as many as seven doses of single or mixed applications of n-PN or *** antifibrotic/pharmacokinetic results for the n-PN/p-LP@aero-μGel included reduced fibrosis progression,restored lung physiological functions,deactivated myofibroblasts,inhibited TGF-βprogression,and suppressed ECM component production(collagen I andα-SMA)along with prolonged lung retention *** believe that our n-PN/p-LP@aero-μGel increased the local availability of both nintedanib and pirfenidone due to evasion of alveolar macrophage phagocytosis and prolonged lung retention with reduced systemic *** this approach,our inhalation formulation subsequently attenuated fibrosis progression and improved lung ***,these results hold profound implications in the therapeutic potential of our n-PN/p-LP@aero-μGel to serve as a clinically promising platform,providing significant advancements for improved treatment of many respiratory diseases including IFP.

Recently,nanovaccine-based immunotherapy has been robustly investigated due to its potential in governing the immune response and generating long-term protective ***,the presentation of a tumor peptide-major histocompatibility complex to T lymphocytes is still a challenge that needs to be addressed for eliciting potent antitumor *** 1 conventional dendritic cell(cDC1)subset is of particular interest due to its pivotal contribution in the cross-presentation of exogenous antigens to CD8+T ***,the DC-derived nanovaccine(denoted as Si9GM)selectively targets cDC1s with marginal loss of premature antigen release for effective stimulator of interferon genes(STING)-mediated antigen *** marrow dendritic cell(BMDC)-derived membranes,conjugated to cDC1-specific antibody(αCLEC9A)and binding to tumor peptide(OVA257-264),are coated onto dendrimer-like polyethylenimine(PEI)-grafted silica *** molecular weight-cargos(αCLEC9A-OVA257-264 conjugates and 2′3′-cGAMP STING agonists)are loaded in hierarchical center-radial pores that enables lysosome escape for potent antigen-cross presentation and activates interferon type I,***,Si9GM vaccination leads to the upregulation of cytotoxic T cells,a reduction in tumor regulatory T cells(Tregs),M1/M2 macrophage polarization,and immune response that synergizes with αPD-1 immune checkpoint *** nanovaccine fulfills a dual role for both direct T cell activation as an artificial antigen-presenting cell and DC subset maturation,indicating its utility in clinical therapy and precision medicine.

Inspired by erythrocytes that contain oxygen-carrying hemoglobin(Hb)and that exhibit photo-driven activity,we introduce homogenous-sized erythrocyte-like Hb microgel(μGel)systems(5-6μm)that can(i)emit heat,(ii)supply oxygen,and(iii)generate reactive oxygen species(ROS;1O2)in response to near-infrared(NIR)laser *** μGels consist of Hb,bovine serum albumin(BSA),chlorin e6(Ce6)and erbium@lutetium upconverting nanoparticles(UCNPs;~35 nm)that effectively convert 808 nm NIR light to 660 nm visible *** Hb μGels are capable of releasing oxygen to help generate sufficient reactive oxygen species(^(1)O_(2))from UCNPs/Ce6 under severely hypoxic condition upon NIR stimulation for efficient photodynamic ***,the Hb μGels emit heat and increase surface temperature due to NIR light absorption by heme(iron protoporphyrin IX)and display photothermal *** changing the Hb/UCNP/Ce6 ratio and controlling the amount of NIR laser irradiation,it is possible to formulate bespoke Hb μGels with either photothermal or photodynamic activity or both in the context of combined therapeutic *** Hb μGels effectively suppress highly hypoxic 4T1 cell spheroid growth and xenograft mice tumors in vivo.

Double-sided gastric perforation after a motorcycle accident in Korea:A case report

BACKGROUND The cellular prion protein(PrPC),traditionally associated with neurodegenerative disorders,plays an important role in cancer progression and metastasis by inhibiting *** To investigate the influence of PrPC expression in cholangiocarcinoma(CCA)on patient outcomes following surgical *** Patients who underwent curative surgical resection for either intrahepatic or hilar CCA were enrolled in this retrospective *** on the immunohistochemical staining results of the surgical specimens,patients were categorized into two groups:The low PrPC group(negative or 1+)and the high PrPC group(2+or 3+).Survival analyses,including overall survival and recurrence-free survival,were conducted using the Kaplan-Meier method and compared using the log-rank *** In total,seventy-six patients diagnosed with CCA(39 with intrahepatic and 37 with hilar CCA)underwent curative hepatectomy from January 2011 to November *** these patients,38(50%)demonstrated high PrPC expression,whereas the remaining 38(50%)showed low expression of *** a median follow-up period of 31.2 months(range:1 to 137 months),the high PrPC group had a significantly shorter median overall survival than the low PrPC group(40.4 months vs 137.9 months,respectively;P=0.041).Moreover,the high PrPC group had a significantly shorter median recurrence-free survival than the low PrPC group(13.3 months vs 23.8 months,respectively;P=0.026).CONCLUSION PrPC expression is significantly associated with early recurrence and decreased survival period in CCA patients following surgical ***,PrPC may be used as a prognostic factor in treatment planning.

Characteristic evaluation of Al_2O_3/CNTs hybrid materials for micro-electrical discharge machining

The characteristic evaluation of aluminum oxide （A1203）/carbon nanotubes （CNTs） hybrid composites for micro-electrical discharge machining （EDM） was described. Alumina matrix composites reinforced with CNTs were fabricated by a catalytic chemical vapor deposition method. A1203 composites with different CNT concentrations were synthesized. The electrical characteristic of A1203/CNTs composites was examined. These composites were machined by the EDM process according to the various EDM parameters, and the characteristics of machining were analyzed using field emission scanning electron microscope （FESEM）. The electrical conductivity has a increasing tendency as the CNTs content is increased and has a critical point at 5% A1203 （volume fraction）. In the machining accuracy, many tangles of CNT in A1203/CNTs composites cause violent spark. Thus, it causes the poor dimensional accuracy and circularity. The results show that conductivity of the materials and homogeneous distribution of CNTs in the matrix are important factors for micro-EDM of A1203/CNTs hybrid composites.

Optical Properties of Zinc-Blende InGaN/GaN Quantum Well Structures and Comparison with Experiment

Optical properties of zinc-blende InGaN/GaN Q W structures are investigated using the multiband effective-mass theory. The transition wavelength values at 300 K ranged from 440 to 570nm in the investigated range of the In composition and the well width. The theoretical wavelengths show reasonable agreement with the experimental results. The optical gain decreases with the increasing well width. This is mainly due to the reduction in the quasi-Fermi-level separation because the optical matrix element increases with the well width.

Highly dense and ultrathin gadolinia-doped ceria interlayer for enhanced performance of large-area anode-supported solid oxide fuel cell

The fabrication of a highly dense and ultrathin Gd doped ceria(GDC) interlayer on a large-area, porous electrolyte-coated planar anode support using vacuum slurry coating is studied for application in solid oxide fuel cells(SOFCs). The electrolyte-coated planar anode support body is fabricated by tape casting and pre-sintering is performed at 1100, 1150, and 1200°C. The GDC interlayer is coated onto the pre-sintered electrolyte-coated anode support using a vacuum slurry coating process with different immersion times(30–60 s). A highly dense and ultrathin GDC interlayer with a thickness of 1.48 μm is produced after co-sintering at 1400°C. The 1200°C pre-sintered electrolyte-coated anode support showed a thin and highly dense GDC interlayer which is attributed to a lower shrinkage difference between the coated GDC interlayer and pre-sintered body. The electrochemical performance of a vacuum slurry-coated GDC interlayer cell is 38 % higher compared to that of the screen-printed GDC-coated cell at 700°C due to the greatly reduced Ohmic and electrode polarization resistances. The considerable improvement in the performance is attributed to the ultrahigh GDC density which has prevented the secondary phase formation between the cathode and electrolyte. This study presents the economical, scalable, and reproducible process for the fabrication of highly robust GDC interlayer for the large-area planar anode-supported SOFCs.
