Single-cell transcriptome analysis reveals the regulatory effects of artesunate on splenic immune cells in polymicrobial sepsis

Sepsis is characterized by a severe and life-threatening host immune response to polymicrobial infection accompanied by organ *** on the therapeutic effect and mechanism of immunomodulatory drugs on the sepsis-induced hyperinflammatory or immunosuppression states of various immune cells remain *** study aimed to investigate the protective effects and underlying mechanism of artesunate(ART)on the splenic microenvironment of cecal ligation and puncture-induced sepsis model mice using single-cell RNA sequencing(scRNA-seq)and experimental *** scRNA-seq analysis revealed that ART inhibited the activation of pro-inflammatory macrophages recruited during *** could restore neutrophils’chemotaxis and immune function in the septic *** inhibited the activation of T regulatory cells but promoted the cytotoxic function of natural killer cells during *** also promoted the differentiation and activity of splenic B cells in mice with *** results indicated that ART could alleviate the inflammatory and/or immunosuppressive states of various immune cells involved in sepsis to balance the immune homeostasis within the ***,this study provided a comprehensive investigation of the regulatory effect of ART on the splenic microenvironment in sepsis,thus contributing to the application of ART as adjunctive therapy for the clinical treatment of sepsis.

Fatigue cracking criterion of high-strength steels induced by inclusions under high-cycle fatigue

Fatigue properties of high-strength steels become more and more sensitive to inclusions with enhancing the ultimate tensile strength (UTS) because the inclusions often cause a relatively low fatigue strength and a large scatter of fatigue lives. In this work, four S–N curves and more than 200 fatigue fracture morphologies were comprehensively investigated with a special focus on the size and type of inclusions at the fatigue cracking origin in GCr15 steel with a wide strength range by different heat treatments after high-cycle fatigue (HCF). It is found that the percentage of fatigue failure induced by the inclusion including Al2 O3 and TiN gradually increases with increasing the UTS, while the percentage of failure at sample surfaces decreases conversely and the fatigue strength first increases and then decreases. Besides, it is interestingly noted that the inclusion sizes at the cracking origin for TiN are smaller than that for Al2 O3 because the stress concentration factor for TiN is larger than that for Al2 O3 based on the finite element simulation. For the first time, a new fatigue cracking criterion including the isometric inclusion size line in the strength-toughness coordinate system with specific physical meaning was established to reveal the relationship among the UTS, fracture toughness, and the critical inclusion size considering different types of inclusions based on the fracture mechanics. And the critical inclusion size of Al2 O3 is about 1.33 times of TiN. The fatigue cracking criterion could be used to judge whether fatigue fracture occurred at inclusions or not and provides a theoretical basis for controlling the scale of different inclusion types for high-strength steels. Our work may offer a new perspective on the critical inclusion size in terms of the inclusion types, which is of scientific interest and has great merit to industrial metallurgical control for anti-fatigue design.

胰體尾+脾切除一直是治療胰體尾病變的經典術式,由於脾臟同胰腺關係緊密,操作時易損傷,因此保留脾臟的胰體尾切除難度大。本院收治1例胰體尾腫瘤,行保留脾臟的胰體尾切除術。現報導如下。患者男性,33歲,因“左上腹痛4小時”入院,腹部CT示胰腺體尾部見一類圓形稍低密度灶,邊緣欠清,範圍約1.6cm×1.6cm,性質待定,腺瘤?查腫瘤指標:CEA、AFP、CA-199等均在正常範圍之間。診斷為胰體尾腫瘤,於2014年5月14日在全麻下行保留脾臟的胰體尾切除。

Owing to their inexpensive and environmentally friendly properties,iron-based catalysts have been actively investigated for new organic *** this account,we summarized our recent results on iron-catalyzed cross-coupling reactions and homo-coupling *** iron-based catalysts,we constructed diverse carbon-carbon bonds,i.e.,C(sp^2)-C(sp^3),C(sp^3)-C(sp^3),C(sp^3)-C(sp^2)and C(sp^2)-C(sp^2)*** order to demonstrate the usefulness of our iron protocol,we also carried out these reactions on gram-scale reactions,leading to good yields.

34例細菌性肝膿腫臨床分析

目的總結細菌性肝膿腫臨床特點及治療。方法回顧性分析2011年1月~2014年4月本院收治34例細菌性肝膿腫患者的臨床表現、實驗室、影像學、檢查資料以及治療方法。結果34例患者均治癒,無死亡病例,住院7~50天,平均住院時間20.76天。94%患者有發熱,58%病人出現腹部症狀;實驗室檢查85%患者出現白細胞升高,94%中性粒細胞左移,近100%C反應蛋白異常;影像超聲波診斷率93%,腹部CT診斷率達到97.0%;致病菌培養結果中:80.76%為肺炎克雷佰氏桿菌,19.23%大腸桿菌;28(82%)例患者成功進行影像學引導經皮膚介入肝穿刺置管引流術,僅1例需外科手術引流治療。結論細菌性肝膿腫與糖尿病有關,肺炎克雷佰氏桿菌主要常見病原菌,有效抗生素應用和影像學引導經皮膚肝介入穿刺置管引流術主要治療方法。

Improved Symmetry Property of High Order Weighted Essentially Non-Oscillatory Finite Difference Schemes for Hyperbolic Conservation Laws

This study aims to investigate the rapid loss of numerical symmetry for problems with symmetrical initial conditions and boundary conditions when solved by the seventh and higher order nonlinear characteristic-wise weighted essentially non-oscillatory(WENO)finite difference *** the one-dimensional double rarefaction wave problem and the Sedov blast-wave problems,and the twodimensional Rayleigh-Taylor instability(RTI)problem as examples,we illustrate numerically that the sensitive interaction of the round-off error due to the numerical unstable explicit form of the local lower order smoothness indicators in the nonlinear weights definition,which are often given and used in the literature,and the nonlinearity of the WENO scheme are responsible for the rapid growth of asymmetry of an otherwise symmetric *** equivalent but compact and numerical stable compact form of the local lower order smoothness indicators is suggested for delaying the onset of and reducing the magnitude of the symmetry *** benefits of using the compact form of the local lower order smoothness indicators should also be applicable to non-symmetrical strongly non-linear problems in terms of improved numerical stability,reduced rounding errors and increased computational efficiency.

目的探討乳腺癌前哨淋巴結活檢(sentinel 1ymph node biopsy,SLNB)的方法和臨床應用價值。方法2015年8月-2016年8月本科共收治41例乳腺癌患者,綵取美蘭染色SLNB,進行術中冰凍診斷,並與術後SLN連續切片及免疫組化檢查作比較。結果41例中,40例成功行SLNB,成功率97.5%,術中冰凍切片發現20例SL N癌轉移,20例陰性者術後行連續切片及常規免疫組化檢查發現2例微轉移。結論美蘭染色SLNB成功率較高.安全.價廉,前哨淋巴結狀況對乳腺癌腋窩淋巴結轉移有重要預示價值,SLN連續切片聯合免疫組化檢查,可增加SLN微轉移的檢出率,減少假陰性率。
