Induction of clusterin Expression by Neuronal Cell Death in Zebrafish

Clusterin, a protein associated with multiple functions, is expressed in a wide variety of mammalian tissues. Although clusterin is known to be involved in neurodegenerative diseases, ageing, and tumorigenesis, a detailed analysis of the consequences of gain- or loss-of- function approaches has yet to be performed to understand the underlying mechanisms of clusterin functions. Since clusterin levels change in neurological diseases, it is likely that clusterin contributes to cell death and degeneration in general. Zebrafish was investigated as a model system to study human diseases. During development, zebrafish clusterin was expressed in the notochord and nervous system. Embryonic overexpression of clusterin by mRNA microinjection did not affect axis formation, whereas its knock-down by anti-sense morpholino treatment resulted in neuronal cell death. To analyze the function of clusterin in neurodegeneration, a transgenic zebrafish was investigated, in which nitroreductase expression is regulated under the control of a neuron-specifc huC promoter which is active between the stages of early neuronal precursors and mature neurons. Nitroreductase turns metronidazole into a cytotoxic agent that induces cell death within 12 h. After metronidazole treatment, transgenic zebrafish showed neuron-specific cell death. Interestingly, we also observed a dramatic induction of clusterin expression in the brain and spinal cord in these fish, suggesting a direct or indirect role of clusterin in neuronal cell death and thus, more generally, in neurodegeneration.

研究目的：本次研究旨在比较加工方法及组成不同的双磷酸钙（biphasic calcium phosphates,BCP）的骨引导的能力和骨愈合的模式。材料和方法：10只雄性新西兰白兔，每只兔子颅顶有4个8mm环形缺损．每个缺损代表一个组别：对照组．实验组：BCP1．70％HA（羟基磷灰石）．30％β-TCP（β-三磷酸钙）：BCP2，30％HA，70％p-TCP；BCP3，20％HA．80％D-TCP。兔子在第2周（n=5）或第8周（n=5）在处死后进行组织学及组织计量学分析。结果：在双磷酸钙（BCP）组的骨增量远大于对照组（P〈005）。第2周或第8周新骨形成量无明显差异。在第2周，BCP3组的双磷酸钙的吸收作用远大于BCP1和BCP2组．然而在第8周．BCP2和BCP3的这种差别变得不明显。BCP组问新骨形成的模式和材料的吸收有明显差异。在BCP1组中．新骨在剩余颗粒中呈线性排列，在BCP2组中，可观察到丝状伪足状新骨形成;在BCP3组中．可见分散在剩余颗粒中的胶原蛋白片段及颗粒表面的裂隙状不规则新骨形成。结论：在本研究中．特定的HA、β-TCP比例对新骨形成和空间的保持无明显影响。而组间观察到的骨愈合模式的差别可归因于加工方法不同所导致的双磷酸钙（BCPs）的物理化学属性的差异。

Sesquiterpene lactones(SLs) are a group of over 500 compounds,characteristic of the Asteraceae and sporadically occurring in other plant *** are interesting from the chemical and chemotaxonomic point of view,and show anti-tumour,anti-leukaemic,anti-cardiovascular disease,reduction of inflammation,anti-microbial *** have been indicated as a property connected to appetite and digestion promotion in *** of folk medicines also implicate SLs as the active ingredient in many treatments for other ailments such as diarrhea,burns,influenza,and *** of 266 accessions of lettuce(Lactuca sativa) germplasm introduced from 43 countries conserved in RDA genebank was used this *** was extracted by methanol from freeze-dried lettuce leaves,sampled at bolting *** SLs compounds,lactucin,lactucopicrin and 8-deoxylactucin,was quantified by *** range of lactucin content showed trace 120.0 μg/g of dried leaves and average was 19.0±1.3,and distribution range trace 488.1 μg and average 16.8±2.9 in 8-deoxylactucin content,and distribution range trace1,189.0 μg and average 195.2±9.9 in lactucopicrin content,and distribution range17.4,299.9 μg and average 231.0±11.4 in total SLs *** lettuce have lowest average total SLs content,on the contrary romaine have highest SLs content and ***,wild relatives,have higher than *** accessions have lowest average total SLs content and Great Britain accessions showed highest SLs *** showed higher than bred *** two accessions have more than500μg of total SLs and seventeen accessions have less than 50μg of total *** germplasm can be used as low SLs content cultivar breeding or high SLs content cultivar breeding sources as well as research materials for medical treatment such as,anti-tumour,anti-leukaemic,and antimicrobial activities,etc.

辣椒(Capsicum annuum)是美洲最古老的驯化作物之一,在全世界被广泛种植。本研究对CM334(墨西哥地方品种)进行了全基因组测序和组装(覆盖度为186.6×),同时还对另外两个栽培种和一个野生种分别进行了重测序和从头(de novo)测序。结果表明辣椒基因组大小约为番茄的4倍,辣椒基因组上存在大量gypsy转座子和花椰菜病毒家族元件的积累。结合转录组综合分析认为,辣椒素合成酶基因表达模式的改变及新功能化是导致辣椒素生物合成的原因,研究还发现辣椒与番茄在乙烯合成及果实成熟调控方面存在明显不同的分子模式。辣椒参考基因组的发布为辣椒营养品质和药用价值的改善提供了重要平台。

AIM： To investigate prevalence of C/onorchis sinensis in patients with gastrointestinal symptoms, and the relation of the infection to hepatobiliary diseases in 26 hospitals in Korea. METHODS： Consecutive patients who had been admitted to the Division of Gastroenterology with gastrointestinal symptoms were enrolled from March to April 2005. Of those who had been diagnosed with clonorchiasis, epidemiology and correlation between infection and hepatobiliary diseases were surveyed by questionnaire. RESULTS： Of 3080 patients with gastrointestinal diseases, 396 （12.9%） had clonorchiasis and 1140 patients （37.2%） had a history of eating raw freshwater fish. Of those with a history of raw freshwater fish ingestion, 238 （20.9%） patients had clonorchiasis. Cholangiocarcinoma was more prevalent in C. sinensis-infected patients than nonnfected patients [34/396 （8.6%） vs 145/2684 （5.4%）, P = 0.015]. Cholangiocarcinoma and clonorchiasis showed statistically significant positive cross-relation （P = 0.008）. Choledocholithiasis, cholecystolithiasis, cholangitis, hepatocellular carcinoma, and biliary pancreatitis did not correlate with clonorchiasis. CONCLUSION： Infection rate of clonorchiasis was still high in patients with gastrointestinal diseases in Korea, and has not decreased very much during the last two decades. Cholangiocarcinoma was related to clonorchiasis, which suggested an etiological role for the parasite.

Electrostatic spraying for flne-tuning particle dimensions to enhance oral bioavailability of poorly water-soluble drugs

While spray-drying has been widely utilized to improve the bioavailability of poorly water-soluble drugs, the outcomes often exhibit suboptimal particle size distribution and large particle sizes, limiting their effectiveness. In this study, we introduce electrostatic spraying as an advanced technology tailored for poorly water-soluble drugs, enabling the fabrication of nanoparticles with flne and uniform particle size distribution. Regorafenib(1 g), as a model drug, copovidone(5 g), and sodium dodecyl sulfate(0.1 g) were dissolved in 200 ml ethanol and subjected to conventional-spray-dryer and electrostatic spray dryer. The electrostatic spray-dried nanoparticles(ESDN) showed smaller particle sizes with better uniformity compared to conventional spray-dried nanoparticles(CSDN). ESDN demonstrated signiflcantly enhanced solubility and rapid release in water. In vitro studies revealed that ESDN induced apoptosis in HCT-116 cells to a greater extent, exhibiting superior cytotoxicity compared to CSDN. Furthermore, ESDN substantially improved oral bioavailability and antitumor efflcacy compared to CSDN. These flndings suggest that ESD shows potential in developing enhanced drug delivery systems for poorly water-soluble drugs, effectively addressing the limitations associated with CSD methods.
