Influence of gastric inhibitory polypeptide on pentagastrinstimulated gastric acid secretion in patients with type 2 diabetes and healthy controls

AIM： Gastric inhibitory polypeptide is secreted from intestinal K-cells in response to nutrient ingestion and acts as an incretin hormone in human physiology. While animal experiments suggested a role for GIP as an inhibitor of gastric secretion, the GIP effects on gastric acid output in humans are still controversial. MeTHODS： Pentagastrin was administered at an infusion rate of 1 μg . kg^-1 . h^-1 over 300 min in 8 patients with type 2 diabetes （2 female, 6 male, 54± 10 years, BMI 30.5 ± 2.2 kg/m^2; no history of autonomic neuropathy） and 8 healthy subjects （2/6, 46 ± 6 years., 28.9 ± 5.3 kg/ m^2）. A hyperglycaemic clamp （140 mg/dl） was performed over 240 min. Placebo, GIP at a physiological dose （1 pmol . kg^-1 . min^-1）, and GIP at a pharmacological dose （4 mol . kg^-1 . min^-1） were administered over 60 min each. Boluses of placebo, 20 pmol GIP/kg, and 80 pmol GIP/kg were injected intravenously at the beginning of each infusion period, respectively. Gastric volume, acid and chloride output were analysed in 15-min intervals. Capillary and venous blood samples were drawn for the determination of glucose and total GIP. Statistics were carried out by repeated-measures ANOVA and one-way ANOVA. ReSULTS： Plasma glucose concentrations during the hyperglycaemic clamp experiments were not different between patients with type 2 diabetes and controls. Steady-state GIP plasma levels were 61 ±8 and 79 ± 12 pmol/I during the low-dose and 327±35 and 327± 17 pmol/I during the high-dose infusion of GIP, in healthy control subjects and in patients with type 2 diabetes, respectively （P= 0.23 and p 0.99）. Pentagastrin markedly increased gastric acid and chloride secretion （P〈 0.001）. There were no significant differences in the rates of gastric acid or chloride output between the experimental periods with placebo or any dose of GIP. The temporal patterns of gastric acid and chloride secretion were similar in patients with type 2 diabetes and healthy controls （P= 0.86 and P= 0.6

Data-driven models for battery state estimation require extensive experimental training data,which may not be available or suitable for specific tasks like open-circuit voltage(OCV)reconstruction and subsequent state of health(SOH)*** study addresses this issue by developing a transfer-learning-based OCV reconstruction model using a temporal convolutional long short-term memory(TCN-LSTM)network trained on synthetic data from an automotive nickel cobalt aluminium oxide(NCA)cell generated through a mechanistic model *** data consists of voltage curves at constant temperature,C-rates between C/30 to 1C,and a SOH-range from 70%to 100%.The model is refined via Bayesian optimization and then applied to four use cases with reduced experimental nickel manganese cobalt oxide(NMC)cell training data for higher use *** TL models’performances are compared with models trained solely on experimental data,focusing on different C-rates and voltage *** results demonstrate that the OCV reconstruction mean absolute error(MAe)within the average battery electric vehicle(BeV)home charging window(30%to 85%state of charge(SOC))is less than 22 mV for the first three use cases across all *** SOH estimated from the reconstructed OCV exhibits an mean absolute percentage error(MAPe)below 2.2%for these *** study further investigates the impact of the source domain on TL by incorporating two additional synthetic datasets,a lithium iron phosphate(LFP)cell and an entirely artificial,non-existing,cell,showing that solely the shifting and scaling of gradient changes in the charging curve suffice to transfer knowledge,even between different cell chemistries.A key limitation with respect to extrapolation capability is identified and evidenced in our fourth use case,where the absence of such comprehensive data hindered the TL process.

Although human health impacts of microplastics are not well understood,concern regarding chemical contaminants retained on or within them is *** water providers are increasingly asked about these risks,but strategies for evaluating them and the extent of treatment needed to manage them are currently *** can potentially induce health effects if the concentration of contaminants adsorbed to them exceeds predetermined drinking water guidelines(e.g.,Maximum Contaminant Levels).The risk posed by microplastics due to adsorbed contaminants is difficult to determine,but a worst-case scenario can be evaluated by using adsorption ***,a“Threshold Microplastics Concentration”(TMC)framework is developed to evaluate whether waterborne microplastic concentrations can potentially result in the intake of regulated contaminants on/in microplastics at levels of human health concern and identify treatment targets for managing associated health *** the TMC does not indicate an immediate health risk;it informs the need for detailed risk assessment or further treatment evaluation to ensure particle removal targets are ***,the TMC concept and framework provide an updateable,science-based screening tool to determine if there is a need for detailed risk assessment or treatment modification due to waterborne microplastics in supplies used for potable water production.

AIM: To evaluate the effects of percutaneous radiation on leukocyte-endothelium interaction (LeI) in experimental hepatocellular carcinoma (HCC). MeTHODS: Twelve ACI rats underwent HCC-inoculation, six of which on day 12 received low-dose external radiation and six did not. After 12 h intravital microscopy was performed. ReSULTS: LeI was significantly reduced in tumor tissue. However,irradiation of liver sinusoids and tumor tissue with 6 Gy led to a significant activation of leukocyte adhesion in the tumor with a marked increase of the proinflammatory cytokine TNF-α. CONCLUSION: The findings indicate that the immune-logical tumor-endothelial barrier can be overcome by external irradiation.
