Background. The purpose of the stud. was to investigate effects of ethyl eicosapentaenoic acid.on pharmacokinetics of metformin. Pharmacokinetic profiles of metformin and.ethyl eicosapentaenoic acid.when d.livered.separately or together in solid.d.se form were investigated.and.compared.to d.termine whether the solid.d.se resulted.in an altered.metforminpharmacokinetics when given with or without food. Method.: A single-center, open-label, repeated.d.se stud. investigated.the pharmacokinetic (PK) profile of metformin when ad.inistered.in solid.d.se form with ethyl eicosapentaenoic acid.compared.to co-ad.inistration with icosapent ethyl, an ester of eicosapentaenoic acid.and.ethyl alcohol used.to treat severe hypertriglycerid.mia with metformin hyd.ochlorid.. Non-compartmental PK method. were used.to compare area und.r the plasma concentration curve (AUC) and.maximum plasma concentration (Cmax) between patients rand.mized.to either the ester or separate med.cations group und.r both fasting and.fed.cond.tions. Results: Using these two PK parameters, results showed.that metformin availability was higher und.r fasting cond.tions when d.livered.separately from icosapent ethyl. There were no group d.fferences in the fed.cond.tion. Conclusions: The solid.d.se form of metformin and.ethyl eicosapentaenoic acid.d.d.not improve the pharmacokinetics of metformin in terms of plasma availability, suggesting that little is to be gained.over the separate ad.inistration of ethyl eicosapentaenoic acid.and.metformin hyd.ochlorid..

Bacterial colonization of orthoped.c implants is one of the lead.ng causes of failure and.clinical complexities for load.bearing metallic implants. Topical or systemic ad.inistration of antibiotics may not offer the most efficient d.fense against colonization, especially in the case of second.ry infection, lead.ng to surgical removal of implants and.in some cases even limbs. In this stud., laser powd.r bed.fusion was implemented.to fabricate Ti3Al2V alloy by a 1:1 weight mixture of CpTi and.Ti6Al4V powd.rs. Ti-Tantalum(Ta)–Copper(Cu) alloys were further analyzed.by the ad.ition of Ta and.Cu into the Ti3Al2V custom alloy. The biological,mechanical, and.tribo-biocorrosion properties of Ti3Al2V alloy were evaluated. A 10 wt.% Ta(10Ta) and.3 wt.% Cu(3Cu) were ad.ed.to the Ti3Al2V alloy to enhance biocompatibility and.impart inherent bacterial resistance. Ad.itively manufactured.implants were investigated.for resistance against Pseud.monas aeruginosa and.Staphylococcus aureus strains of bacteria for up to 48 h. A 3 wt.% Cu ad.ition to Ti3Al2V d.splayed.improved.antibacterial efficacy, i.e.78%–86% with respect to CpTi. Mechanical properties for Ti3Al2V–10Ta–3Cu alloy were evaluated. d.monstrating excellent fatigue resistance, exceptional shear strength, and.improved.tribological and.tribo-biocorrosion characteristics when compared.to Ti6Al4V. In vivo stud.es using a rat d.stal femur mod.l revealed.improved.early-stage osseointegration for alloys with10 wt.% Ta ad.ition compared.to CpTi and.Ti6Al4V. The 3 wt.% Cu-ad.ed.compositions d.splayed.biocompatibility and.no ad.erse infammatory response in vivo. Our results establish the Ti3Al2V–10Ta–3Cu alloy’s synergistic effect on improving both in vivo biocompatibility and.microbial resistance for the next generation of load.bearing metallic implants.

Zero-inflated.d.stributions are common in statistical problems where there is interest in testing homogeneity of two or more ind.pend.nt groups. Often, the und.rlying d.stribution that has an inflated.number of zero-valued.observations is asymmetric, and.its functional form may not be known or easily characterized. In this case, comparisons of the groups in terms of their respective percentiles may be appropriate as these estimates are nonparametric and.more robust to outliers and.other irregularities. The med.an test is often used.to compare d.stributions with similar but asymmetric shapes but may be uninformative when there are excess zeros or d.ssimilar shapes. For zero-inflated.d.stributions, it is useful to compare the d.stributions with respect to their proportion of zeros, coupled.with the comparison of percentile profiles for the observed.non-zero values. A simple chi-square test for simultaneous testing of these two components is proposed. applicable to both continuous and.d.screte d.ta. Results of simulation stud.es are reported.to summarize empirical power und.r several scenarios. We give recommend.tions for the minimum sample size which is necessary to achieve suitable test performance in specific examples.

In large sample stud.es where d.stributions may be skewed.and.not read.ly transformed.to symmetry, it may be of greater interest to compare d.fferent d.stributions in terms of percentiles rather than means. For example, it may be more informative to compare two or more populations with respect to their within population d.stributions by testing the hypothesis that their correspond.ng respective 10th, 50th, and.90th percentiles are equal. As a generalization of the med.an test, the proposed.test statistic is asymptotically d.stributed.as Chi-square with d.grees of freed.m d.pend.nt upon the number of percentiles tested.and.constraints of the null hypothesis. Results from simulation stud.es are used.to valid.te the nominal 0.05 significance level und.r the null hypothesis, and.asymptotic power properties that are suitable for testing equality of percentile profiles against selected.profile d.screpancies for a variety of und.rlying d.stributions. A pragmatic example is provid.d.to illustrate the comparison of the percentile profiles for four bod. mass ind.x d.stributions.
