Inflammatory responses,manifested in excessive oxidative stress and microglia overactivation,together with metal ion-triggered amyloid-beta(Aβ)deposition,are critical hallmarks of Alzheimer’s disease(AD).The intricate pathogenesis causes severe impairment of neurons,which,in turn,exacerbates Aβaggregation and facilitates AD ***,multifunctional melanin-like metal ion chelators and neuroinflammation regulators(named PDA@K)were constructed for targeted treatment of *** this platform,intrinsically bioactive material polydopamine nanoparticles(PDA)with potent metal ion chelating and ROS scavenging effects were decorated with the KLVFF peptide,endowing the system with the capacity of enhanced pathological blood–brain barrier(BBB)crossing and lesion site accumulation via Aβ*** vitro and in vivo experiment revealed that PDA@K had high affinity toward Aβand were able to hitch a ride on Aβto achieve increased pathological BBB *** engineered PDA@K effectively mitigated Aβaggregate and alleviated neuroinflammation.

The application of photothermal therapy(PTT)is greatly limited by the low accumulation of photothermal agents,uneven photothermal distribution,and heat endurance of cancer *** still,despite PTT enhances immunogenicity,the anti-tumor immune efficacy is still unsatisfactory due to the inefficient immunogenic cell death(ICD)induction and poor infiltration of immune *** solve the above problems of PTT,we developed hyaluronic acid(HA)modified hollow copper sulfide nanoparticles encapsulating diethyldithiocarbamate(DDTC)to construct a breast tumor targeting and near infrared(NIR)photo-responsive drug delivery system(D-HCuS@HA),which further combined with losartan to improve the accumulation and penetration in the tumor *** irradiation,D-HCuS@HA realized enhanced PTT and released cytotoxic Cu(DDTC)_(2)to eliminate heat endurance tumor cells,thereby enhancing antitumor effect and inducing effective ***,the combination with losartan could remodel the tumor microenvironment,allowing more T cells to infiltrate into the tumor,and significantly inhibiting the occurrence and development of metastatic *** vitro/vivo results revealed the great potential of D-HCuS@HA combined with losartan,which provides a new paradigm for anti-tumor and anti-metastases.

目的探讨脂联素(APN)是否可通过PPARα/HOXA10通路改善多囊卵巢综合征(PCOS)大鼠子宫内膜容受性。方法6周龄雌性SD大鼠随机分为对照组(CON,n=10)和PCOS模型组(n=40)。用来曲唑[1 mg/(kg·d)]建立PCOS大鼠模型后,再随机分为4组(n=10):模型对照组(PCOS),APN干预组(PCOS+APN)、APN联合PPARα抑制剂(GW6471)干预组(PCOS+APN+GW)及APN联合玉米油溶剂(vehicle,VEH)对照组(PCOS+APN+VEH);后3组大鼠均给与APN[10μg/(kg·d)]干预20 d。在给APN的第11天,PCOS+APN+GW组同时给与GW6471[1 mg/(kg·d)]作为阴性对照,PCOS+APN+VEH组同时给与vehicle(0.1 mL/d)作为阳性对照。检测大鼠动情周期、卵巢和子宫指数及其形态学变化、血清激素及生化指标的改变。免疫组化和Western blot检测PPARα和HOXA10蛋白的表达,电镜观察子宫内膜胞饮突的发育情况,合笼实验观察大鼠的妊娠情况。结果与CON组比较,PCOS组大鼠动情周期紊乱,多处于动情间期;平均体质量和卵巢指数增加、子宫指数下降(P<0.05),血清激素与脂质代谢异常,卵巢多囊样改变;给与APN干预后各组各项指标均得到恢复。子宫内膜组织中PPARα和HOXA10的表达:在PCOS组较CON组显著下调(P<0.05)、在PCOS+APN组中较PCOS组显著上调(P<0.05)、在PCOS+APN+GW组中较PCOS+APN组及PCOS+APN+VEH组则显著下调(P<0.05)。与CON组比较,PCOS组大鼠子宫内膜胞饮突发育欠佳;PCOS+APN组胞饮突发育较PCOS组良好;与PCOS+APN组比较,PCOS+APN+GW组胞饮突发育欠佳;与PCOS+APN+GW组比较,PCOS+APN+VEH组胞饮突发育良好。与PCOS组比较,各组大鼠妊娠率一致;但PCOS+APN+GW组大鼠胚胎数量相对于PCOS+APN组显著减少且发育迟缓。结论APN可通过上调PPARα/HOXA10通路改善PCOS大鼠子宫内膜容受性,该结果有望为改善PCOS病人妊娠结局的治疗提供实验依据。

目的:探究调脏舒痢方外用治疗慢性难愈性创面(CRW)的临床疗效。方法:选取106例CRW患者,将上述患者使用动态随机法分为两组。对照组(n=53)使用PRP凝胶治疗,观察组(n=53)使用调脏舒痢方外用进行治疗,观察两组临床疗效、创面愈合情况、疼痛程度(VAS)和治疗前后炎性指标[白细胞计数和C反应蛋白(CRP)水平]、创面修复因子[血管内皮生长因子(VEGF)、碱性成纤维细胞生长因子(bFGF)和表皮生长因子(EGF)]水平。结果:观察组总有效率高于对照组(P0.05)。结论:调脏舒痢方外用治疗CRW疗效良好,能有效改善患者创面愈合情况,减轻疼痛程度,降低炎性指标水平,提高创面修复因子水平。

目的探讨UBE2I和FCGR1A表达对艾滋病(acquired immune deficiency syndrome,AIDS)合并活动性肺结核(active pulmonary tuberculosis,APTB)发病的影响,为疾病监测提供依据。方法从已验证的全基因组转录谱数据集(GSE37250)选取AIDS合并APTB患者98例、AIDS合并潜伏期肺结核感染(latent tuberculosis infection,LTBI)患者84例,筛选两组患者的前30位差异表达基因(differentially expressed gene,DEG)建立PPI交互作用网络、转录因子-差异基因(transcription factor-differentially expressed gene,TF-DEG)、DEG-miRNA和环境化学物-DEG调控网络,绘制11个关键DEGs的受试者工作特征(ROC)曲线并作Logistic回归分析。结果两组患者间存在6054个DEGs,UBE2I为PPI交互作用网络的重要核心节点,FCGR1A对AIDS合并APTB的预测指示能力最佳。单因素Logistic回归显示:UBE2I、FCGR1A的高表达是AIDS合并APTB的危险因素(P<0.05)。调控网络显示:VEGFB是TF-DEG网络的关键基因,与SEPT9、SMAD5等转录因子共同参与调节,与hsa-mir-17-5p、hsa-mir-20a-5p等miRNA存在靶向作用,并受到丙戊酸、硫酸铜等环境化学物的影响。结论VEGFB在AIDS合并APTB发病过程中发挥重要作用,UBE2I和FCGR1A的异常高表达与AIDS合并APTB的疾病进程存在关联,可通过检测UBE2I和FCGR1A的表达水平监测病情。

医学影像共享是医疗信息云共享中最重要的部分,因为医疗信息80%以上是医学影像,但信息共享面临数据安全、隐私保护和信息检索等问题。虽然已有很多密文域可逆信息隐藏(RDH-EI,Reversible Data Hiding in Encrypted Image)方案,但一般不能直接应用于DICOM医学影像上。为了满足云服务中DICOM文件的隐私保护和信息检索需求,文章提出一种基于ZUC加性同态和多层差值直方图平移的DICOM图像RDH-EI方案。所提方案不改变DICOM文件格式,不增加文件大小,且图像解密和信息提取可分离。实验结果表明,所提出的方案具有良好的灵活性和计算效率,是一种适用云共享的RDH-EI方案。

As known,the benefits of photothermal therapy(PTT)are greatly limited by the heat tolerance of cancer cells resulting from overexpressed heat shock proteins(HSPs).Then HSPs further trigger the formation of stress granules(SGs)that regulate protein expression and cell viability under various stress *** of SG formation can sensitize tumor cells to ***,we developed PEGylated pH(low)insertion peptide(PEG-pHLIP)-modified hollow copper sulfide nanoparticles(HCuS NPs)encapsulating the SG inhibitor ISRIB,with the phase-change material lauric acid(LA)as a gatekeeper,to construct a pH-driven and NIR photo-responsive controlled smart drug delivery system(IL@H-PP).The nano medicine could specifically target slightly acidic tumor *** irradiation,IL@H-PP realized PTT,and the light-controlled release of ISRIB could effectively inhibit the formation of PTT-induced SG to sensitize tumor cells to PTT,thereby increasing the antitumor effect and inducing potent immunogenic cell death(ICD).Moreover,IL@H-PP could promote the production of reactive oxygen species(ROS)by tumor-associated macrophages(TAMs),repolarizing them towards the M1 phenotype and remodeling the immunosuppressive *** vitro/vivo results revealed the potential of PTT combined with SG inhibitors,which provides a new paradigm for antitumor and antimetastases.

Effective clinical trials are necessary for understanding medical advances but early termination of trials can result in unnecessary waste of *** models can be used to predict survival probabilities in such ***,survival data from clinical trials are sparse,and DeepSurv cannot accurately capture their effective features,making the models weak in generalization and decreasing their prediction *** this paper,we propose a survival prediction model for clinical trial completion based on the combination of denoising autoencoder(DAE)and DeepSurv *** DAE is used to obtain a robust representation of features by breaking the loop of raw features after autoencoder training,and then the robust features are provided to DeepSurv as input for *** clinical trial dataset for training the model was obtained from the *** dataset.A study of clinical trial completion in pregnant women was conducted in response to the fact that many current clinical trials exclude pregnant *** experimental results showed that the denoising autoencoder and deep survival regression(DAE-DSR)model was able to extract meaningful and robust features for survival analysis;the C-index of the training and test datasets were 0.74 and 0.75 *** with the Cox proportional hazards model and DeepSurv model,the survival analysis curves obtained by using DAE-DSR model had more prominent features,and the model was more robust and performed better in actual prediction.

Locally repairable codes（LRCs）, which can recover any symbol of a codeword by reading only a small number of other symbols, have been widely used in real-world distributed storage systems, such as Microsoft Azure Storage and Ceph Storage Cluster. Since binary linear LRCs can significantly reduce coding and decoding complexity, constructions of binary LRCs are of particular interest. The aim of this paper is to construct dimensional optimal binary LRCs with disjoint local repair groups. We introduce a method to connect intersection subspaces with binary LRCs and construct dimensional optimal binary linear LRCs with locality 2b（b 3） and minimum distance d 6 by employing intersection subspaces deduced from the direct sum. This method will sufficiently increase the number of possible repair groups of dimensional optimal LRCs, thus efficiently expanding the range of the construction parameters while keeping the largest code rates compared with all known binary linear LRCs with minimum distance d 6 and locality 2b.
