High-precision analysis of carbon isotopic composition for individual CO_(2)inclusions via Raman spectroscopy to reveal the multiple-stages evolution of CO_(2)-bearing fluids and melts

The carbon isotopic composition of CO_(2)inclusions trapped in minerals reflects the origin and evolution of CO_(2)-bearing fluids and melts,and records the multiple-stages carbon geodynamic cycle,as CO_(2)took part in various geological processes widely.However,the practical method for determination isotope composition of individual CO_(2)inclusion is still lacking.Developing a microanalytical technique with spatial resolution in micrometers to precisely determinate theδ^(13)C value of individual CO_(2)inclusion,will make it possible to analyze a tiny portion of a zoning mineral crystal,distinguish the differences in micro-scale,and possible to find many useful information that could not be obtained with the bulk extraction and analysis techniques.In this study,we systematically collected Raman spectra of CO_(2)standards with different d13C values(34.9‰to 3.58‰)at 32.0℃and from 7.0 MPa to 120.0 MPa,and developed a new procedure to precisely determinate theδ^(13)C value of individual CO_(2)inclusion.We investigated the relationship among the Raman peak intensity ratio,d13C value,and CO_(2)density,and established a calibration model with high accuracy(0.5‰1.5‰),sufficient for geological application to distinguish different source of CO_(2)with varyingδ^(13)CO_(2).As a demonstration,we measured theδ^(13)C values and the density of CO_(2)inclusions in the growth zones of alkali basalt-hosted corundum megacrysts from Changle,Shandong Province.We found the significant differences of density and d13C between the CO_(2)inclusions in the core of corundum and those inclusions in the outer growth zones,the d13C value decreases from core to rim with decreasing density:δ^(13)C values are from7.5‰to9.2‰for the inclusions in the core,indicating the corundum core was crystallized from mantle-derived magmas;from13.5‰to18.5‰for CO_(2)inclusions in zone 1 and from16.5‰to–22.0‰for inclusions in zone 2,indicating the outer zones of corundum grew in a lowδ^(13)C value environment,resulted from an infilling of low d13C value flu

Simulation Analysis and Verification of Temperature and Stress of Wheel-Mounted Brake Disc of a High-Speed Train

During the braking process, a large amount of heat energy is generated at the friction surfaces between the brake disc and pads and rapidly dissipates into the disc volume. In this paper, a three-dimensional thermo-mechanical coupling model of high-speed wheel-mounted brake discs containing bolted joints and contact relationships is established. The direct coupling method is used to analyze the temperature and stress of the brake discs during an emergency braking event with an initial speed of 300 km/h. A full-scale bench test is also conducted to monitor the temperatures of the friction ring and bolted joints. The simulation result shows that the surface temperature of the friction ring reaches its peak value of 414 ℃ after 102 s of braking, which agrees well with the bench test result. The maximum alternating thermal stress occurs in the bolt hole where the maximum circumferential compressive stress is-658 MPa and the maximum circumferential tensile stress is 134 MPa. During the braking process, the out-of-plane deformation of the middle part of the friction ring is larger than that of the edge, which increases the axial tensile load of the connecting bolt. This work provides support for the design of brake discs and connecting bolts.

Eucaryotic cells spatially organize their molecules to form various organelles by compartmentalization, thereby performing fundamental biological process. While some of those organelles are delimited by lipid membranes, others are membrane-less. Biomolecular condensates are widely existed as membrane-less cellular compartments in eukaryotic cells, which are usually formed by phase separation.

In 2019,the FDA’s Center for Drug Evaluation and Research(CDER)approved 48 novel drugs[https://www.***.gov/drugs/new-drugsfda-cders-new-molecular-entities-and-new-therapeutic-biologicalproducts/novel-drug-approvals-2019],1 which was fewer than the all-time record of 62 NTD approvals in 2018,2 but it was still a fruitful year.Among these approved drugs,39 were targeted drugs(Table 1),including 27 small molecules,3 antibody-drug conjugates(ADCs),1 RNA interference(RNAi)therapy,1 antisense oligonucleotide,4 monoclonal antibodies(mAbs),1 recombinant fusion protein,and 2 synthetic peptide analogs.The targets included kinases,ion channels,exons,enzymes,and receptors.Oncology,which remains the most important drug discovery area,accounted for 23%(9/39)of the targeted drug approvals.

Long non-coding RNA colon cancer-associated transcript 2:role and function in human cancers

Long non-coding RNAs(lncRNAs)are a family of non-protein-coding RNAs that span a length of over 200 nucleotides.Research reports have illustrated that lncRNAs are involved in various cellular processes and that their abnormal expression leads to the occurrence and development of various tumors.Colon cancer-associated transcript 2(CCAT2)was first reported as an oncogene in colon cancer.LncRNA CCAT2 is abnormally expressed in hepatocellular carcinoma,cholangiocarcinoma,lung cancer,breast cancer,ovarian cancer,glioma,and other tumors.In tumor tissues,abnormally overexpressed CCAT2 can affect cell proliferation,migration,epithelial-mesenchymal transition,apoptosis,and other biological behaviors through endogenous RNAs mechanisms,various signaling pathways,transcriptional regulation,and other complex mechanisms.Additionally,the overexpression of CCAT2 is also closely related to the tumor size,tumor node metastasis(TNM)stage,survival time,and other prognostic factors,suggesting that it is a potential prognostic indicator.This article reviews the biological functions of CCAT2 and its mechanisms of action in tumors from previous studies.In this review,we attempt to provide a molecular basis for future clinical applications of lncRNA CCAT2.

Diffusion mechanism of immiscible Fe-Mg system induced by high-density defects at the steel/Mg composite interface

Due to positive mixing heat between Fe and Mg,it is difficult to diffuse for Fe-Mg at the interface of steel/Mg laminated composites,resulting in the inability to achieve high-strength metallurgical bonding.In this paper,20#steel/Mg laminated composites were prepared by large deformation rolling and subse-quent diffusion heat treatment process.The interfacial bonding strength was improved by constructing high-density crystal defects at the interface to promote element diffusion.The mechanisms of interface morphology evolution and element diffusion were analyzed by finite element simulation and theoretical calculation.The results show after diffusion heat treatment,the bond strength of the large deformation rolled interface was increased from 14 to 30 MPa.Fe-Mg transition layer with about 80 nm thickness as well as high-density vacancies,dislocations and grain boundaries were formed in the large deforma-tion rolled interface region.During diffusion heat treatment,Mg elements diffused into grain interior and grain boundary regions of 20#steel under the effect of heat-force coupling,and the thickness of Fe-Mg transition layer increased to 150 nm,forming an Fe-based supersaturated solid solution.The in-terface with high-density defects constituted a non-equilibrium interface.The 20#steel internal energy in the non-equilibrium interface is able to overcome positive mixing heat of immiscible Fe-Mg system and provide the driving force for Mg elements diffusion.Promoting elemental diffusion by constructing high-density defects can be a new concept to achieve metallurgical bonding at the interface of immiscible metal laminated composites.

Spatial–temporal characteristics of haze and vertical distribution of aerosols over the Yangtze River Delta of China

Variation of haze events occurred in the Yangtze River Delta（YRD） of China, the characteristics of meteorological elements and the vertical distribution of aerosols during haze episodes were analyzed by utilizing data of ground observation, radiosonde and CALIPSO. The results illustrate that the frequency of haze events between 1981 and 2010 peaked in winter but bottomed out in summer and decreased from north to south in the YRD region, reaching at the lowest point in ＂low frequency center＂ - Shanghai. When haze happened, the most seriously affected area was 2-4 km above the ground and the concentrated range of total backscattering coefficient（TBC） that decreased with altitude was 0.8 × 10^（-3）-2.5 × 10^（-3） km^（-1）·sr^（-1）. Particulate depolarization ratio（PDR） was less than 40%in a large part and 93% aerosols over the YRD area were regular particles, while the irregular ones concentrated on 2 km above the surface and the irregularity rose up but the diversity diminished when altitude increased. Color ratio（CR） was lower than 1.2 mostly at all altitudes and distributed asymmetrically above the ground. Nearly 80% aerosols under10 km were fine particles（CR 〈1.0） clustered at 2-4 km. Large particles（CR 〉 1.2） aggregated in lower troposphere massively yet relatively smaller ones gathered in middle and upper troposphere. In the YRD region, aerosols with more powerful capabilities were wider and less regular than the ones of Northwestern China.

Designing gradient solid electrolyte interphase for stable lithium metal batteries

The practical application of lithium metal batteries(LMBs)has been impeded by the unstable electrolyte interphase and uncontrollable Li dendrites growth.The structures and components of solid electrolyte interphase(SEI)are extremely important to affect the electrochemical performance of LMBs,but it is hard to regulate them due to the complicated reaction mechanisms.Therein,a gradient SEI layer was designed by adding bisfluoroacetamide into electrolyte,which could guide the uniform deposition of Li ions and suppress the growth of Li dendrites.In addition,this additive also could improve the stability of cathode,resulting in a stable LMBs.

Self-assembling carrier-free nanodrugs are attractive agents because they accumulate at tumor by an enhanced permeability and retention(EPR) effect without introduction of inactive substances,and some nanodrugs can alter the immune environment. We synthesized a peptidyl arginine deiminase 4(PAD4) molecular inhibitor, ZD-E-1 M. It could self-assembled into nanodrug ZD-E-1. Using confocal laser scanning microscopy, we observed its cellular colocalization, PAD4 activity and neutrophil extracellular traps(NETs) formation. The populations of immune cells and expression of immune-related proteins were determined by single-cell mass cytometry. ZD-E-1 formed nanoflowers in an acidic environment, whereas it formed nanospheres at pH 7.4. Accumulation of ZD-E-1 at tumor was pHresponsive because of its pH-dependent differences in the size and shape. It could enter the nucleus and bind to PAD4 to prolong the intracellular retention time. In mice, ZD-E-1 inhibited tumor growth and metastasis by inhibiting PAD4 activity and NETs formation. Besides, ZD-E-1 could regulate the ratio of immune cells in LLC tumor-bearing mice. Immunosuppressive proteins like LAG3 were suppressed,while IFN-γ and TNF-a as stimulators of tumor immune response were upregulated. Overall, ZD-E-1 is a self-assembling carrier-free nanodrug that responds to pH, inhibits PAD4 activity, blocks neutrophil extracellular traps formation, and improves the tumor immune microenvironment.
