Targeting TLR4 and regulating the Keap1/Nrf2 pathway with andrographolide to suppress inflammation and ferroptosis in LPS-induced acute lung injury

Acute lung injury（ALI） is a severe inflammatory condition with a high mortality rate, often precipitated by sepsis. The pathophysiology of ALI involves complex mechanisms, including inflammation, oxidative stress, and ferroptosis, a novel form of regulated cell death. This study explores the therapeutic potential of andrographolide（AG）, a bioactive compound derived from Andrographis, in mitigating Lipopolysaccharide（LPS）-induced inflammation and ferroptosis. Our research employed in vitro experiments with RAW264.7 macrophage cells and in vivo studies using a murine model of LPS-induced ALI. The results indicate that AG significantly suppresses the production of pro-inflammatory cytokines and inhibits ferroptosis in LPS-stimulated RAW264.7 cells. In vivo, AG treatment markedly reduces lung edema, decreases inflammatory cell infiltration, and mitigates ferroptosis in lung tissues of LPS-induced ALI mice. These protective effects are mediated via the modulation of the Toll-like receptor 4（TLR4）/Kelch-like ECH-associated protein 1（Keap1）/Nuclear factor erythroid 2-related factor 2（Nrf2） signaling pathway. Molecular docking simulations identified the binding sites of AG on the TLR4 protein(Kd value:-33.5 kcal·mol-1), and these interactions were further corroborated by Cellular Thermal Shift Assay（CETSA） and SPR assays. Collectively, our findings demonstrate that AG exerts potent anti-inflammatory and anti-ferroptosis effects in LPS-induced ALI by targeting TLR4 and modulating the Keap1/Nrf2 pathway. This study underscores AG's potential as a therapeutic agent for ALI and provides new insights into its underlying mechanisms of action.

Fractal Characteristics and Main Controlling Factors of High-Quality Tight Sandstone Reservoirs in the Southeastern Ordos Basin

Due to the complex conditions and strong heterogeneity of tight sandstone reservoirs,the reservoirs should be classified and the controlling factors of physical properties should be *** thin section observations,cathodoluminescence,scanning electron microscopy(SEM),X-ray diffraction(XRD),and high-pressure mercury injection(HPMI)were used to classify and optimize the *** Brooks-Corey model and stepwise regression were used to study the fractal dimension and main controlling factors of the physical properties of the high-quality *** results show that the reservoirs in the study area can be divided into four types,and the high-quality reservoir has the best physical properties and pore-throat *** the high-quality reservoir,the homogeneity of transitional pores was the best,followed by that of micropores,and the worst was *** porosity was controlled by depth and *** model with standardized coefficients is y=12.454−0.778×(Depth)+0.395×(Kaolinite).The permeability was controlled by depth,illite/montmorillonite,and siliceous cement,and the model with standardized coefficients is y=1.689−0.683×(Depth)−0.395×(Illite/Montmorillonite)−0.337×(Siliceous Cement).The pore-throat evolutionary model shows that the early-middle diagenetic period was when the reservoir physical properties were at their best,and the kaolinite intercrystalline pores and residual intergranular pores were the most important.

为探究蒸压加气混凝土砌块在建筑节能方面存在的问题,利用AIRPAK软件对蒸压加气混凝土砌块墙体的热工性能进行研究。通过对300 mm厚的B04级蒸压加气混凝土砌块墙体进行不同厚度砌筑灰缝进行建模研究,探讨不同灰缝厚度情况下蒸压加气混凝土砌块墙体传热系数变化情况以及在墙体外侧粘贴分别25 mm和50 mm石墨聚苯板保温时灰缝对墙体传热系数的影响。结果表明:当砌筑灰缝厚度为8~15 mm时,墙体传热系数与不考虑灰缝时的传热系数增加比例为15.152%~31.313%,在建筑热工计算时不能忽视灰缝对墙体传热造成的影响;随着保温层厚度的增加,灰缝对传热系数的影响逐渐减小,但不可忽略;当保温层厚度为25 mm和50 mm时,传热系数增加比例分别为6.584%~15.265%和4.231%~21.154%。

Proteo-transcriptomic profiles reveal key regulatory pathways and functions of LDHA in the ovulation of domestic chickens(Gallus gallus)

Background In poultry, the smooth transition of follicles from the preovulatory-to-postovulatory phase impacts egg production in hens and can benefit the poultry industry. However, the regulatory mechanism underlying follicular ovulation in avians is a complex biological process that remains *** Critical biochemical events involved in ovulation in domestic chickens(Gallus gallus) were evaluated by transcriptomics, proteomics, and in vitro assays. Comparative transcriptome analyses of the largest preovulatory follicle(F1) and postovulatory follicle(POF1) in continuous laying(CL) and intermittent laying(IL) chickens indicated the greatest difference between CL_F1 and IL_F1, with 950 differentially expressed genes(DEGs), and the smallest difference between CL_POF1 and IL_POF1, with 14 DEGs. Additionally, data-independent acquisition proteomics revealed 252 differentially abundant proteins between CL_F1 and IL_F1. Perivitelline membrane synthesis, steroid biosynthesis, lysosomes, and oxidative phosphorylation were identified as pivotal pathways contributing to ovulation regulation. In particular, the regulation of zona pellucida sperm-binding protein 3, plasminogen activator, cathepsin A, and lactate dehydrogenase A(LDHA) was shown to be essential for ovulation. Furthermore, the inhibition of LDHA decreased cell viability and promoted apoptosis of ovarian follicles in *** This study reveals several important biochemical events involved in the process of ovulation, as well as crucial role of LDHA. These findings improve our understanding of ovulation and its regulatory mechanisms in avian species.

单桩式基础作为海上风电应用最为广泛的支撑型式,其周围普遍存在不同程度冲蚀。目前,海上风电基础冲刷后的结构动力响应研究较少考虑叶片、机舱、轮毂等细节特征,而是将上部结构简化为质点或静止状态,缺乏基础-结构-荷载一体化数值仿真方法,难以反映冲刷后风机结构振动规律。为此,依托菩提岛海上风电场,通过多波束扫测探明了单桩基础最大冲刷深度,基于AFL(等效嵌固长度)法考虑桩基柔性特征,计算不同冲刷深度下桩基等效嵌固长度,建立风、浪共同作用下风机仿真模型,揭示不同冲刷深度下单桩式海上风电动力响应规律。结果表明,现有桩基冲刷深度难以引起结构共振,且风机的整体振幅较小。塔架和桩基均在F-A(前后)向出现最大振幅,而S-S(侧)向和YAW(垂)向的位移幅值相对较小,故需要对F-A向振动进行重点监测。随着桩基局部冲刷深度增加,风机结构固有频率单调递减,当冲刷深度达到12 m,结构整体刚度不足,给出了海上风电单桩基础冲刷安全阈值。提出的单桩式海上风电动力响应分析方法可为工程中不同冲刷深度引起风机振动安全性评价提供依据。

Engineering myocardium has shown great clinal potential for repairing permanent myocardial ***,the lack of perfusing blood vessels and difficulties in preparing a thick-engineered myocardium result in its limited clinical *** prepared a mixed gel containing fibrin(5 mg/ml)and collagen I(0.2 mg/ml)and verified that human umbilical vein endothelial cells(HUVECs)and human-induced pluripotent stem cell-derived cardiomyocytes(hiPSC-CMs)could fom microvascular lumens and myocardial cell clusters by harnessing the low-hardness and hyperelastic characteristics of ***-CMs and HUVECs in the mixed gel formed self-organized cell clusters,which were then cultured in different media using a three-phase *** successfully constructed vascularized engineered myocardial tissue had a spherical structure and final diameter of *** tissue exhibited autonomous beats that occured at a frequency similar to a normal human heart *** internal microvascular lumen could be maintained for 6 weeks and showed good results during preliminary surface re-vascularization in vitro and vascular remodeling in *** summary,we propose a simple method for constructing vascularized engineered myocardial tissue,through phased cultivation that does not rely on high-end manufacturing equipment and cutting-edge preparation *** constructed tissue has potential value for clinical use after preliminary evaluation.
