【目的】探究赤水乌骨鸡胸肌组织中腺苷琥珀酸裂解酶基因(ADSL)表达特征及其目的基因的干扰载体构建,为深入研究畜禽组织肌肉中肌苷酸(IMP)的表达规律及ADSL基因影响畜禽风味的机制提供理论参考。【方法】以不同月龄(3、4、5和6月龄)的赤水乌骨鸡(公鸡和母鸡各半)为试验材料,运用实时荧光定量PCR检测不同月龄及不同性别赤水乌骨鸡胸肌组织中ADSL基因的表达情况;同时根据GenBank中鸡ADSL基因mRNA序列(NM_205529.2)设计4对短发夹RNA(shRNA)干扰序列(sh1-ADSL、sh2-ADSL、sh3-ADSL和sh4-ADSL)和1对阴性对照序列(sh-NC)与pGPH1/GFP/Neo载体连接后进行测序,将构建成功的ADSL基因干扰载体转染至成肌细胞,检测并筛选出干扰效率最高的干扰载体。【结果】ADSL基因在不同月龄赤水乌骨鸡胸肌组织中的相对表达量排序为5月龄>4月龄>6月龄>3月龄,赤水乌骨母鸡3月龄母鸡的ADSL基因相对表达量极显著高于公鸡(P<0.01,下同),4月龄母鸡ADSL基因的相对表达量显著高于公鸡(P<0.05),6月龄母鸡ADSL基因的相对表达量极显著低于公鸡;ADSL基因质粒的测序结果显示干扰质粒构建成功,质粒转染后干扰组及阴性对照均可见绿色荧光,空白对照不发光,说明各重组质粒成功转染赤水乌骨鸡成肌细胞;ADSL基因沉默结果显示,sh2-ADSL沉默效果最佳,干扰效率为90.30%。【结论】赤水乌骨鸡胸肌中ADSL基因的相对表达量随着月龄增加表现为先上升后下降的变化趋势;3、4和5月龄赤水乌骨母鸡ADSL基因的相对表达量高于公鸡,随着月龄增加ADSL基因的相对表达量增加,公鸡和母鸡间的差异减小。成功分离提取赤水乌骨鸡成肌细胞,并筛选出RNA干扰效果最佳载体sh2-ADSL,为后续研究ADSL基因调控鸡肉肌苷酸及风味的分子机制提供参考。

Leaching of vanadium and chromium from converter vanadium slag intensified with surface wettability

Technology intensified with surface wettability was introduced to leach vanadium and chromium from converter vanadium slag without roasting. Parameters affecting the leaching efficiency of vanadium and chromium were investigated： sulfuric acid concentration, MnOz-to-slag mass ratio, liquid-to-solid ratio, leaching time, leaching temperature, and sodium dodecyl sulfate （SDS）-to-slag mass ratio. The leaching efficiencies of vanadium and chromium were 33.46 % and 20.02 % higher in the presence of MnO2 and SDS, respectively, compared to the control. The leaching efficiencies of vanadium and chromium were 68.93 % and 30.74 %, respectively, under the optimum conditions： sulfuric acid concentration 40 wt%, MnOz-to-slag mass ratio 10.0 wt%, liquid-to-solid ratio 5：1 mL/g; 12 h; 90 ~C; and SDS-to-slag mass ratio 0.25 wt%. The analysis of the reaction mechanism in the leaching process indicates that MnO2 combined with protons （H＋） could oxidize low-valent vanadium and chromium; SDS could change the chemical behavior and decrease the surface tension of the aqueous solution to favor MnO2 oxidization.

How Could Agricultural Land Systems Contribute to Raise Food Production Under Global Change?

To feed the increasing world population, more food needs to be produced from agricultural land systems. Solutions to produce more food with fewer resources while minimizing adverse environmental and ecological consequences require sustainable agricultural land use practices as supplementary to advanced biotechnology and agronomy. This review paper, from a land system perspective, systematically proposed and analyzed three interactive strategies that could possibly raise future food production under global change. By reviewing the current literatures, we suggest that cropland expansion is less possible amid iferce land competition, and it is likely to do less in increasing food production. Moreover, properly allocating crops in space and time is a practical way to ensure food production. Climate change, dietary shifts, and other socio-economic drivers, which would shape the demand and supply side of food systems, should be taken into consideration during the decision-making on rational land management in respect of sustainable crop choice and allocation. And ifnally, crop-speciifc agricultural intensiifcation would play a bigger role in raising future food production either by increasing the yield per unit area of individual crops or by increasing the number of crops sown on a particular area of land. Yet, only when it is done sustainably is this a much more effective strategy to maximize food production by closing yield and harvest gaps.

八角茴香中二氧化硫残留量分析及安全性评价

目的检测预包装与散装八角茴香流通、生产(分装)、餐饮环节中二氧化硫残留量情况,并对其进行安全性评价。方法在大型商场、批发零售市场、餐饮企业、网络平台等不同领域随机抽取预包装与散装八角茴香90批次、新鲜湿八角16批次,参照GB 5009.34—2022《食品安全国家标准食品中二氧化硫的测定》检测二氧化硫残留量,依据GB 2760—2014《食品安全国家标准食品添加剂使用标准》规定对八角茴香中二氧化硫残留量进行评价分析,并从食品、药品两方面对其进行健康风险评估。结果 90批次八角茴香中二氧化硫残留量超出标准限值的为13.3%,其中预包装和散装八角茴香二氧化硫残留量平均值分别为33.6 mg/kg和93.6 mg/kg,超标率分别为2.5%和22.0%。流通环节、生产(分装)环节和餐饮环节八角茴香中二氧化硫残留量平均值分别为66.3、42.4、90.6 mg/kg,超标率分别为12.5%、8.3%、21.4%。16批次新鲜湿八角中二氧化硫本底值的平均值为9.38 mg/kg。散装八角茴香中二氧化硫残留量高于预包装八角茴香,且散装八角茴香中二氧化硫残留量超标率显著高于预包装八角茴香(X^(2)=7.313,P=0.007)。流通、生产(分装)、餐饮各个环节均存在二氧化硫残留量超标现象,各环节超标率比较差异无统计学意义(X^(2)=1.092,P=0.579)。结论八角茴香作为食品使用时,各年龄段人群对散装和预包装八角茴香中的二氧化硫暴露风险系数均小于1,但作为药品使用时,低年龄段人群对部分八角茴香中的二氧化硫暴露风险系数大于1,在风险管理上需引起足够重视。在生产、消费、研发等各领域均应加强宣传监管力度,控制八角茴香中二氧化硫残留量,确保八角茴香质量安全。

Soybean root diseases are associated with numerous fungal and oomycete pathogens;however,the community dynamics and interactions of these pathogens are largely *** performed 13 loop-mediated isothermal amplification(LAMP)assays that targeted specific soybean root pathogens,and traditional isolation assays.A total of 159 samples were collected from three locations in the Huang-Huai-Hai region of China at three soybean growth stages(30,60,and 90 days after planting)in *** LAMP results,we found that pathogen communities differed slightly among locations,but changed dramatically between soybean growth *** sojae,Rhizoctonia solani,and Fusarium oxysporum were most frequently detected at the early stage,whereas Phomopsis longicolla,Fusarium equiseti,and Fusarium virguliforme were most common in the later *** samples(86%)contained two to six pathogen ***,the less detectable species tended to exist in the samples containing more detected species,and some pathogens preferentially co-occurred in diseased tissue,including ***–***–*** and ***–Calonectria ilicicola,implying potential interactions during *** LAMP detection results were confirmed by traditional isolation *** isolated strains exhibited different virulence to soybean,further implying a beneficial interaction among some pathogens.

Chemical mutagenesis and soybean mutants potential for identification of novel genes conferring resistance to soybean cyst nematode

The resistance of soybean(Glycine max(L.) Merr.) to soybean cyst nematode(SCN, Heterodera glycines Ichinohe), which is a devastating pathogen in soybean production and causes a large quantity of annual yield loss worldwide, can shift during the long-term interaction and domestication. It is vital to identify more new resistance genetic sources for identification of novel genes underlying resistance to SCN for management of this pathogen. In the present study, first, two ethane methylsulfonate-mutagenesis soybean M2 populations of PI 437654, which shows a broad resistance to almost all of SCN races, and Zhonghuang 13, which is a soybean cultivar in China conferring strong resistance to lodging, were developed. Many types of morphological phenotypes such as four-and five-leaflet leaves were observed from these two soybean M2 populations. Second, 13 mutants were identified and confirmed to exhibit alteration of resistance to SCN race 4 through the forward genetic screening of 400 mutants of the PI 437654 M2 population, the rate of mutants with alteration of SCNinfection phenotype is 3.25%. Third, these identified mutants were further verified not to show any changes in the genomic sequences of the three known SCN-resistant genes, GmSHMT08, GmSNAP18 and GmSANP11, compared to the wildtype soybean; and all of them were still resistant to SCN race 3 similar to the wild-type soybean. Taken together, we can conclude that the 13 mutants identified in the present study carry the mutations of the new gene(s) which contribute(s) to the resistance to SCN race 4 in PI 437654 and can be potentially used as the genetic soybean sources to further identify the novel SCN-resistant gene(s).
