OBJECTIVE:To identify the active ingredients and metabolites in rat bile after Guangtongxiao decoction(GTX)had been administered via the rectal ***:Drug-containing bile samples were collected via a catheter in the bile duct and could be used 5 h after rectal *** main active components and their metabolites in rat bile following rectal administration of GTX were identified and analyzed using ultra-high-performance liquid chromatography-quadrupole time-of-flight mass ***:Positive and negative modes were applied to analyze and identify the chemical ingredients in the bioactive fractions of *** peaks were identified by comparison with the standard compounds:berberine hydrochloride,dehydrocorydaline,tetrahydropalmatine,corydaline,magnoflorine,magnolol,obacunone and ***,60 metabolites were detected in rat bile based on mass-fragmentation behaviors,and 21 metabolites were reported for the first ***:Our findings provide a solid basis for further pharmacologic and pharmacokinetic studies of GTX.

In order to modify in-situ synthesized Mg_2Si particles in Mg_2Si/Mg-4Si composite, the modif ication effect of calcium-magnesia phosphate fertilizer on primary Mg_2Si phase in Mg_2Si/Mg-4Si composite was investigated by means of X-ray diffraction(XRD), optical microscopy(OM), scanning electron microscopy(SEM) and energy dispersive spectroscopy(EDS) analysis. The results indicate that the morphology of the primary Mg_2Si phase apparently changes from coarse dendrites to f ine dispersive polygonal particles, and the mean size is decreased from 277 μm to 17 μm. With the addition of 4.0wt.% calcium-magnesia phosphate fertilizer as a modif ier, the ultimate tensile strength and elongation of the Mg_2Si/Mg-4Si composite are increased from 78.7 MPa and 2.1% to 105.2 MPa and 2.6%, as compared to those of the base composite, which is probably attributed to the formation of the phosphorous compound and the cluster of Ca compounds that acted as the heterogeneous nucleation substrates of the primary Mg_2Si particles, resulting in a ref ined distribution of these precipitates.

Zeolites have been widely used as catalysts,ion-exchangers,and adsorbents in chemical industries,detergent industry,steel industry,glass industry,ceramic industry,medical and healthﬁeld,and environmentalﬁeld,and recently applied in energy ***-assisted synthesis is a very effective approach in promoting the crystallization of *** some cases,the target zeolite cannot be formed in the absence of seed *** homologous seed-assisted synthesis,the structure of the seed zeolite is the same to that of the target zeolite,while the structure of the seed zeolite is different to that of the target zeolite in the heterologous seed-assisted *** this review,we brieﬂy summarized the heterologous seed-assisted syntheses of zeolites and analyzed the structure-directing effect of heterologous seeds and surveyed the“common composite building units(CBUs)hypothesis”and the“common secondary building units(SBUs)hypothesis”.However,both hypotheses cannot explain all observations on the heterologous seed-assisted ***,we proposed that the formation of the target zeolite does need nuclei with the structure of target zeolite and the formation of the nuclei of the target zeolite can be promoted by either the undissolved seed crystals with the same CBUs or SBUs to the target zeolite or by the facilitated appropriate distribution of the speciﬁc building units due to the presence of the heterologous seed that does not have any common CBUs and SBUs with the target zeolite.

Current cytotoxic T lymphocyte(CTL)activating immunotherapy requires a major histocompatibility complex I(MHC-I)-mediated presentation of tumor-associated antigens,which malfunctions in around half of patients with triple-negative breast cancer(TNBC).Here,we create a LCL161-loaded macrophage membrane decorated nanoparticle(LMN)for immunotherapy of MHC-I-deficient *** on the macrophage membrane helps LMNs recognize CD47-expressing cancer cells for targeted delivery of LCL161,which induces the release of high mobility group protein 1 and proinflammatory cytokines from cancer *** released cytokines and high mobility group protein 1 activate antitumor immunity by increasing the intratumoral density of the phagocytic macrophage subtype by 15 times and elevating the intratumoral concentration of CTL lymphotoxin by 4.6 *** also block CD47-mediated phagocytosis *** inhibit the growth of MHC-I-deficient TNBC tumors,as well as those resistant to combined therapy of anti-PDL1 antibody and albumin-bound paclitaxel,and prolong the survival of animals,during which process CTLs also play important *** macrophage membrane-decorated nanoparticle presents a generalizable platform for increasing macrophagemediated antitumor immunity for effective immunotherapy of MHC-I-deficient cancers.

Aerosol liquid water content(ALWC)plays an important role in secondary aerosol *** this study,a whole year field campaign was conducted at Shanxi in north Zhejiang Province during *** estimated by ISORROPIA-Ⅱ was then investigated to explore its characteristics and relationship with secondary *** exhibited a highest value in spring(66.38μg/m^(3)),followed by winter(45.08μg/m^(3)),summer(41.64μg/m^(3)),and autumn(35.01μg/m^(3)),*** was supposed that the secondary inorganic aerosols(SIA)were facilitated under higher ALWC conditions(RH>80%),while the secondary organic species tended to form under lower ALWC *** RH(>80%)promoted the NO_(3)^(-)formation via gas-particle partitioning,while SO_(4)^(2-)was generated at a relative lower RH(>50%).The ALWC was more sensitive to NO_(3)^(-)(R=0.94)than SO_(4)^(2-)(R=0.90).Thus,the self-amplifying processes between the ALWC and SIA enhanced the particle mass *** sensitivity of ALWC and OX(NO_(2)+O_(3))to secondary organic carbon(SOC)varied in different seasons at Shanxi,more sensitive to aqueous-phase reactions(daytime R=0.84;nighttime R=0.54)than photochemical oxidation(daytime R=0.23;nighttime R=0.41)in wintertime with a high level of OX(daytime:130-140μg/m^(3);nighttime:100-140μg/m^(3)).The self-amplifying process of ALWC and SIA and the aqueous-phase formation of SOC will enhance aerosol formation,contributing to air pollution and reduction of visibility.

Molecular subtyping of gastric cancer(GC)aims to comprehend its genetic ***,the efficacy of current subtyping methods is hampered by their mixed use of molecular features,a lack of strategy optimization,and the limited availability of public GC *** is a pressing need for a precise and easily adoptable subtyping approach for early DNA-based screening and *** on TCGA subtypes,we developed a novel DNA-based hierarchical classifier for gastric cancer molecular subtyping(HCG),which employs gene mutations,copy number aberrations,and methylation patterns as *** incorporating the closely related esophageal adenocarcinomas dataset,we expanded the TCGA GC dataset for the training and testing of HCG(n=453).The optimization of HCG was achieved through three hierarchical strategies using Lasso-Logistic regression,evaluated by their overall the area under receiver operating characteristic curve(auROC),accuracy,F1 score,the area under precision-recall curve(auPRC)and their capability for clinical stratification using multivariate survival ***-specific DNA alteration biomarkers were discerned through difference tests based on HCG defined *** HCG classifier demonstrated superior performance in terms of overall auROC(0.95),accuracy(0.88),F1 score(0.87)and auPRC(0.86),significantly improving the clinical stratification of patients(overall p-value=0.032).Difference tests identified 25 subtype-specific DNA alterations,including a high mutation rate in the SYNE1,ITGB4,and COL22A1 genes for the MSI subtype,and hypermethylation of ALS2CL,KIAA0406,and RPRD1B genes for the EBV *** is an accurate and robust classifier for DNA-based GC molecular subtyping with highly predictive clinical stratification *** training and test datasets,along with the analysis programs of HCG,are accessible on the GitHub website(***/LabxSCUT).

The flotation of complex solid–liquid multiphase systems involve interactions among multiple components,the core problem facing flotation ***,the combined use of multicomponent flotation reagents to improve mineral flotation has become an important issue in studies on the efficient use of refractory mineral ***,studying the flotation of complex solid–liquid systems is extremely difficult,and no systematic theory has been developed to *** addition,the physical mechanism associated with combining reagents to improve the flotation effect has not been unified,which limits the development of flotation theory and the progress of flotation *** this study,we applied theoretical thermodynamics to a solid–liquid flotation system and used changes in the entropy and Gibbs free energy of the reagents adsorbed on the mineral surface to establish thermodynamic equilibrium equations that de-scribe interactions among various material components while also introducing adsorption equilibrium constants for the flotation reagents adsorbed on the mineral *** homogenization effect on the mineral surface in pulp solution was determined using the chemical potentials of the material components of the various mineral surfaces required to maintain *** flotation effect can be improved through synergy among multicomponent flotation reagents;its physical essence is the thermodynamic law that as the number of compon-ents of flotation reagents on the mineral surface increases,the surface adsorption entropy change increases,and the Gibbs free energy change of adsorption *** to the results obtained using flotation thermodynamics theory,we established high-entropy flotation theory and a technical method in which increasing the types of flotation reagents adsorbed on the mineral surface,increasing the adsorption entropy change of the flotation reagents,decreasing the Gibbs free energy change,and improving the adsorption efficiency

The inflammatory microenvironment and neurotoxicity can hinder neuronal regeneration and functional recovery after spinal cord ***,a JAK-STAT inhibitor,exhibits effectiveness in autoimmune diseases,arthritis,and managing inflammatory cytokine *** studies have shown the neuroprotective potential of ruxolitinib in neurological trauma,the exact mechanism by which it enhances functional recovery after spinal cord injury,particularly its effect on astrocytes,remains *** address this gap,we established a mouse model of T10 spinal cord contusion and found that ruxolitinib effectively improved hindlimb motor function and reduced the area of spinal cord *** sequencing analysis showed that ruxolitinib alleviated inflammation and immune response after spinal cord injury,restored EAAT2 expression,reduced glutamate levels,and alleviated excitatory ***,ruxolitinib inhibited the phosphorylation of JAK2 and STAT3 in the injured spinal cord and decreased the phosphorylation level of nuclear factor kappa-B and the expression of inflammatory factors interleukin-1β,interleukin-6,and tumor necrosis factor-α.Additionally,in glutamate-induced excitotoxicity astrocytes,ruxolitinib restored EAAT2 expression and increased glutamate uptake by inhibiting the activation of STAT3,thereby reducing glutamate-induced neurotoxicity,calcium influx,oxidative stress,and cell apoptosis,and increasing the complexity of dendritic ***,these results indicate that ruxolitinib restores glutamate homeostasis by rescuing the expression of EAAT2 in astrocytes,reduces neurotoxicity,and effectively alleviates inflammatory and immune responses after spinal cord injury,thereby promoting functional recovery after spinal cord injury.
