Petroleum geology features and research developments of hydrocarbon accumulation in deep petroliferous basins

As petroleum exploration advances and as most of the oil-gas reservoirs in shallow layers have been explored, petroleum exploration starts to move toward deep basins, which has become an inevitable choice. In this paper, the petroleum geology features and research progress on oil-gas reservoirs in deep petroliferous basins across the world are characterized by using the latest results of worldwide deep petroleum exploration. Research has demonstrated that the deep petroleum shows ten major geological features. （1） While oil-gas reservoirs have been discovered in many different types of deep petroliferous basins, most have been discovered in low heat flux deep basins. （2） Many types of petroliferous traps are developed in deep basins, and tight oil-gas reservoirs in deep basin traps are arousing increasing attention. （3） Deep petroleum normally has more natural gas than liquid oil, and the natural gas ratio increases with the burial depth. （4） The residual organic matter in deep source rocks reduces but the hydrocarbon expulsion rate and efficiency increase with the burial depth. （5） There are many types of rocks in deep hydrocarbon reservoirs, and most are clastic rocks and carbonates. （6） The age of deep hydrocarbon reservoirs is widely different, but those recently discovered are pre- dominantly Paleogene and Upper Paleozoic. （7） The porosity and permeability of deep hydrocarbon reservoirs differ widely, but they vary in a regular way with lithology and burial depth. （8） The temperatures of deep oil-gas reservoirs are widely different, but they typically vary with the burial depth and basin geothermal gradient. （9） The pressures of deep oil-gas reservoirs differ significantly, but they typically vary with burial depth, genesis, and evolu- tion period. （10） Deep oil-gas reservoirs may exist with or without a cap, and those without a cap are typically of unconventional genesis. With continuous hydrocarbon exploration extending to deeper basins,the deepest industrial oil accumulation was discovered below 8,200 m,revealing a new exploration ***,the extent to which oil exploration can be extended,and the prediction of the depth limit of oil accumulation(DLOA),are issues that have attracted significant attention in petroleum *** it is difficult to characterize the evolution of the physical properties of the marine carbonate reservoir with burial depth,and the deepest drilling still cannot reach the ***,the DLOA cannot be predicted by directly establishing the relationship between the ratio of drilling to the dry layer and the *** this study,by establishing the relationships between the porosity and the depth and dry layer ratio of the carbonate reservoir,the relationships between the depth and dry layer ratio were obtained *** depth corresponding to a dry layer ratio of 100%is the *** on this,a quantitative prediction model for the DLOA was finally *** results indicate that the porosity of the carbonate reservoir,Lower Ordovician in Tazhong area of Tarim Basin,tends to decrease with burial depth,and manifests as an overall low porosity reservoir in deep *** critical porosity of the DLOA was 1.8%,which is the critical geological condition corresponding to a 100%dry layer ratio encountered in the *** depth of the DLOA was 9,000 *** study provides a new method for DLOA prediction that is beneficial for a deeper understanding of oil accumulation,and is of great importance for scientific guidance on deep oil drilling.

Hydrocarbon expulsion features and resource potential evaluation of source rocks are crucial for the petroleum ***-maturity marine source rocks have not exhibited a hydrocarbon expulsion mode owing to the lack of low-maturity source rocks in deep petroliferous *** considered the Ediacaran microbial dolomite in the Sichuan Basin,the largest high-maturity marine gas layer in China,to exhibit a method that quantitatively characterizes the hydrocarbon expulsion of high-maturity marine source *** experiment of fluid inclusion,rock pyrolysis,and vitrinite reflectance(Ro)of 119 microbial dolomite core samples obtained from the Dengying Formation were performed.A hydrocarbon expulsion model of high-maturity source rock was established,and its resource potential was *** results showed that the Ediacaran microbial dolomite in the Sichuan Basin is a good source rock showing vast resource *** hydrocarbon expulsion threshold is determined to be vitrinite reflectance at 0.92%.The hydrocarbon expulsion intensities in the geologic history is high with maximum of 1.6×10^(7)t/km^(2).The Ediacaran microbial dolomite expelled approximately 1.008×10^(12)t of hydrocarbons,and the recoverable resource was 1.5×10^(12)m^(3).The region can be categorized into areasⅠ,Ⅱ,Ⅲ,andⅣ,in decreasing order of hydrocarbon expulsion *** with a higher hydrocarbon expulsion intensity have a lower drilling risk and should be prioritized for exploration in the orderⅠ>Ⅱ>Ⅲ>Ⅳ.Two areas,northern and central parts of Ediacaran in the Sichuan Basin,were selected as prospects which had the drilling priority in the future gas *** production data of 55 drilled wells verified the high reliability of this *** model in this study does not require low-maturity samples and can be used for evaluating high-maturity marine source rocks,which has broad applicability in deep basins worldwide.

Tumour metastasis-associated gene profiling using one-dimensional microfluidic beads array

Great efforts have been made on the early diagnosis and molecular mechanism research of tumour metastasis in recent years. In this paper, based on the one-dimensional microfluidic beads array, a novel platform for tumour metastasis-associated genes profiling has been developed by depositing nucleic acids functional beads in the microchannel. This platform is sensitive (limit of detection: 0.02 nmol/L) and can perform mRNAs analysis without PCR. Two human colon cancer cell lines (primary and metastatic) from the same patient were used as a model, and transcriptional expression profiling of multiple tumour metastasis-associated genes in these two cell lines was successfully achieved. Fur- thermore, the results obtained on the beads array were validated by RT-PCR. This novel beads array has advantages of high sensitivity, little sample consumption, short assay time, low cost and high throughput capability. It holds the potential in early diagnosis and mechanism research of tumour me-tastasis.

Characteristics and mechanism of enzyme secretion and increase in [ Ca^(2+)]_i in Saikosaponin.(I)stimulated rat pancreatic acinar cells

AIM: This investigation was to reveal the characteristics and mechanism of enzyme secretion and increase in [Ca2+]i stimulated by saikosaponin(I) (SA(I)) in rat pancreatic acini. METHODS:Pancreatic acini were prepared from male Wistar rats. Isolated acinar cells were suspended in Eagle's MEM solution. After adding drugs, the incubation was performed at 37 degrees for a set period of time. Amylase of supernatant was assayed using starch-iodide reaction. Isolated acinar single cell was incubated with Fura-2/AM at 37 degrees, then cells were washed and resuspended in fresh solution and attached to the chamber. Cytoplasm [Ca2+]i of a single cell was expressed by fluorescence ratio F340/F380 recorded in a Nikon PI Ca2+ measurement system. RESULTS: Rate course of amylase secretion stimulated by SA(I) in rat pancreatic acini appeared in bell-like shape. The peak amplitude increased depended on SA(I) concentration. The maximum rate responded to 1 x 10(-5)mol/L SA(I) was 13.1-fold of basal and the rate decreased to basal level at 30 min. CCK-8 receptor antagonist Bt(2)-cGMP markedly inhibited amylase secretion stimulated by SA(I) and the dose-effect relationship was similar to that by CCK-8. [Ca2+]i in a single acinar cell rose to the peak at 5 min after adding 5 x 10(-6)mol/L SA(I) and was 5.1-fold of basal level. In addition, there was a secondary increase after the initial peak. GDP could inhibit both the rate of amylase secretion and rising of [Ca2+]i stimulated by SA(I) in a single pancreatic acinar cell. CONCLUSION: SA(I) is highly efficient in promoting the secretion of enzymes synthesized in rat pancreatic acini and raising intracellular [Ca2+]i. Signaling transduction pathway of SA(I) involves activating special membrane receptor and increase in cytoplasm [Ca2+]i sequentially.

Preparation of monoclonal antibody against apoptosis-associated antigens of hepatoma cells by subtractive immunization

AIM: To elucidate the expression of the apoptosis-associatedmolecules in human primary hepatocellular carcinoma (HCC)cells, and prepare the monoclonal antibodies (mAb) againstthe apoptosis-associated antigens of HCC ***: Human HCC cell line HCC-9204 cells wereinduced apoptosis with 60 mL.L-1 ethanol for 6 h and theirmorphological changes were observed by transmissionelectron microscope. The cell DNA fragmentations weredetected by Terminal Deoxynucleotidyl transferase-mediateddUTP nick end labeling (TUNEL) assay, and the cell DNAcontents by flow cytometry. Ten mice were immunized withethanol-induced apoptotic HCC-9204 cells with the methodof subtractive immunization, while the other 10 mice usedas the control were immunized by the routine *** tail blood of all the mice were prepared after the lastimmunization, and the produced antibodies were determinedby the immunocytochemical ABC staining. The splenic cellsof the mice whose tail blood sera-HCC-9204 cells serumreactions were most different between the apoptotic andthe non-apoptotic were prepared and fused with the mousemyeloma cell line SP2/0 cells. The positive antibodies wereselected by ELISA assay. The fusion rates of hybridoma cellsand the producing rates of antibodies were calculated. Thefused cells that secreted candidate objective antibody werecloned continually with the of limited dilution method, andthen selected and analyzed further by theimmunocytochemical ABC staining. The chromosomes of thecloned hybridoma cells that secreted objective mAb and themAb immunoglobulin (Ig) subtype of the prepared mAb werealso determined. The molecular mass of the mAb associatedantigen was analyzed by Western blot ***: HCC-9204 cells treated with 60 mL.L-1 ethanolfor 6 h, manifested obvious apoptotic morphological changes,the majority of the cells were TUNEL-positive, and the sub-G1 apoptotic peak was evident. Predictors of patient survival following living donor liver transplantation

BACKGROUND: Living donor liver transplantation (LDLT) is considered to be the alterative choice in light of the great shortage of cadaveric donors. However, the characteristics of the patients who will benefit from LDLT have not been well identified. The aim of this study was to define the pre- and intra-operative factors that may influence patient outcome. METHODS: The data from 102 LDLT patients who had operations between 2002 and 2009 were collected and analyzed retrospectively. Data were analyzed using uni- and multi-variate analysis according to factors that are known to be associated with outcome in these patients. RESULTS: Overall, the accurate survival rate of recipients at 1, 3, and 5 years was 84%, 76%, and 70%, respectively. The independent risk factors, preoperative renal dysfunction, intraoperative red blood cell transfusions of greater than 5 units, and female to male match (donor to recipient matching), were identified by Cox regression analysis. The pre-transplant model for end-stage liver disease score and a graft to recipient weight ratio of less than 0.8% were not predictive of outcome. The overall 1-, 3-, and 5-year survival of patients with one or no risk factors and two or more risk factors were 91%, 86%, and 83% and 67%, 56%, and 47%, respectively (P<0.0001). CONCLUSIONS: In our retrospective study, preoperative renal dysfunction, intraoperative red blood cell transfusions of greater than 5 units, and female to male gender match were independent risk factors for LDLT recipient outcome. Two or more of these risk factors may contribute to poor outcome.
