Study on the Relationship between AMPK/UCP2 Pathway and Mitochondrial Dysfunction in Granulosa Cells of Polycystic Ovay Syndrome

Objective:To investigate the changes of the AMP-activated protein kinase(AMPK)/uncoupling protein 2(UCP2)(AMPK/UCP2)pathway in ovarian granulosa cells with PCOS and its relationship with mitochondrial ***:PCOS mouse models and normally fed mice,ovarian granulosa cells from the two mice were extracted,and the protein expression levels of AMPKα,p-AMPKαand UCP2 were detected by western *** ROS and ATP content of granulosa cells were determined by colorimetric and chemiluminescence immunoassays to assess mitochondrial *** correlation analysis was used to determine the correlation between AMPK/UCP2 pathway-related proteins,ROS and ***:P-AMPKα/GAPDH(0.12±0.09),AMPKα/GAPDH(0.35±0.40),P-AMPKα/AMPKα(0.56±0.33)and ATP(0.36±0.04)pmol/mg in PCOS model mice were lower than those in non-POCS groups,while UCP2/GAPDH(1.18±0.28)and ROS(48810.92±4498.08)were lower than those in non-POCS *** fluorescence intensity of DCF was higher than that of the non-POCS group(Phere are abnormal changes such as decreased AMPK expression and increased UCP2 expression in ovarian granulosa cells of PCOS,and AMPK is positively and negatively correlated with mitochondrial function indexes ATP and ROS,while UCP2 is the opposite,suggesting that the imbalance in the expression and activity of AMPK/UCP2 pathway in PCOS may be one of the molecular mechanisms leading to mitochondrial *** of AMPK/UCP2 pathway activity may be a potential therapeutic target to ameliorate PCOS-related mitochondrial dysfunction.

Task allocation is a key aspect of Unmanned Aerial Vehicle(UAV)swarm collaborative *** an continuous increase of UAVs’scale and the complexity and uncertainty of tasks,existing methods have poor performance in computing efficiency,robustness,and realtime allocation,and there is a lack of theoretical analysis on the convergence and optimality of the *** paper presents a novel intelligent framework for distributed decision-making based on the evolutionary game theory to address task allocation for a UAV swarm system in uncertain scenarios.A task allocation model is designed with the local utility of an individual and the global utility of the ***,the paper analytically derives a potential function in the networked evolutionary potential game and proves that the optimal solution of the task allocation problem is a pure strategy Nash equilibrium of a finite strategy ***,a PayOff-based Time-Variant Log-linear Learning Algorithm(POTVLLA)is proposed,which includes a novel learning strategy based on payoffs for an individual and a time-dependent Boltzmann *** former aims to reduce the system’s computational burden and enhance the individual’s effectiveness,while the latter can ensure that the POTVLLA converges to the optimal Nash equilibrium with a probability of *** simulation results show that the approach is optimal,robust,scalable,and fast adaptable to environmental changes,even in some realistic situations where some UAVs or tasks are likely to be lost and increased,further validating the effectiveness and superiority of the proposed framework and algorithm.

Extracellular membrane proteins are crucial for mediating cell attachment,recognition,and signal transduction in the testicular microenvironment,particularly germline stem *** 18(CDH18),a type Ⅱ classical cadherin,is primarily expressed in the nervous and reproductive ***,we investigated the expression of CDH18in neonatal porcine prospermatogonia(ProSGs)and murine spermatogonial stem cells(SSCs).Disruption of CDH18 expression did not adversely affect cell morphology,proliferation,self-renewal,or differentiation in cultured porcine ProSGs,but enhanced cell adhesion and prolonged cell *** analysis indicated that the down-regulation of CDH18 in ProSGs significantly up-regulated genes and signaling pathways associated with cell *** further elucidate the function of CDH18 in germ cells,Cdh18 knockout mice were generated,which exhibited normal testicular morphology,histology,*** analysis showed increased expression of genes associated with adhesion,consistent with the observations in porcine *** interaction of CDH18withβ-catenin and JAK2 in both porcine ProSGs and murine SSCs suggested an inhibitory effect on the canonical Wnt and JAK-STAT signaling pathways during CDH18 ***,these findings highlight the crucial role of CDH18 in regulating cell adhesion in porcine ProSGs and mouse *** this regulatory mechanism provides significant insights into the testicular niche.

BACKGROUND In this study,we retrospectively analysed macrophage infiltration and podocyte injury in three patients with diffuse proliferative lupus nephritis(LN)who un-derwent repeated renal *** SUMMARY Clinical data of three diffuse proliferative LN patients with different pathological characteristics(case 1 was LN IV-G(A),case 2 was LN IV-G(A)+V,and case 3 was LN IV-G(A)+thrombotic microangiopathy)were *** patients underwent repeated renal biopsies 6 mo later,and renal biopsy specimens were *** infiltration was assessed by CD68 expression detected by immunohistochemical staining,and an immunofluorescence assay was used to detect podocin expression to assess podocyte *** treatment,Case 1 changed to LN III-(A),Case 2 remained as type V LN lesions,and Case 3,which changed to LN IV-S(A),had the worst *** observed reduced macro-phage infiltration after ***,two of the patients with active lesions after treatment still showed macrophage infiltration in the renal *** treatment,the three patients showed discontinuous expression of ***,the integrity of podocin was restored after treatment in Case *** It may be possible to reverse podocyte damage and decrease the infiltrating ma-crophages in LN patients through effective treatment.

Effect of ellagitannins gut microbiota metabolites ellagic acid (EA) and urolithin A-urolithin D (UroA-UroD) on human serum albumin (HSA) glycation were firstly evaluated in this research. The inhibition mechanisms were investigated by methylglyoxal (MGO) trapping and radical scavenging ability assays， docking studies and Nano LC-Orbitrap-MS/MS technology. Results indicated that the inhibition of urolithins on HSA glycation was highly positive correlated with the number of phenolic hydroxy groups. Addition of UroD and EA could effectively enhance the content of free amino group， suppress dicarbonyl compounds and advanced glycation end-products (AGEs) formation， alleviated tryptophan and protein oxidation， inhibited HSA amyloid-like aggregation. They could also trap MGO and scavenge DPPH· and ABTS·+. Molecular docking indicated that EA and UroA-UroD interact with HSA mainly through hydrogen bound and hydrophobic interaction， among which two or three hydrogen bonds were formed. The number of glycation sites were reduced from 11 to 10， 10， 7， and 10， respectively， when 90 μmol/L of EA， UroA， UroC and UroD were added. However， weak inhibition was observed on UroA and UroB. These findings can provide scientific evidence for the application of ellagitannins-rich foods in alleviating diabetic complications.
