PFOA/PFOS Facilitated Intestinal Fatty Acid Absorption by Activating the PPARαPathway:Insights from Organoids Model

Perfluorooctanoic acid(PFOA)and perfluorooctanesulfonate(PFOS)continue to be extensively present in the natural environment and seriously threaten human *** intestinal tract is the primary organ of PFOA/PFOS exposure due to the consumption of contaminated food and drinking ***,it remains unclear how PFOA/PFOS affects intestinal function and overall *** aim of this study was to investigate the influence of PFOA/PFOS on the absorption of fatty acids in the intestine and the underlying mechanisms using three-dimensional(3D)intestinal *** results showed that PFOS,but not PFOA,could significantly enhance the fatty acid uptake capacity without obvious damage to the ***,PFOS markedly reduced the protein levels of ChgA in enteroendocrine cells,but with no observed impact on aldolase B+***,exposure to PFOS induced the activation of the peroxisome proliferator-activated receptor(PPAR)αpathway in intestinal organoids,with enhanced expression of PPARαtarget genes associated with fatty acid metabolism,such as Fabp1 and Cd36(fatty acid transporter genes),Acox1 and Pdk4(fatty acid oxidation genes),and Plin2 and Plin3(lipid droplet synthesis genes).These data suggest that PFOS have the potential to affect the absorption function of the intestinal epithelium through the PPARαpathway,and its effect is much stronger than that of *** findings also highlight that organoids can be used as a valuable model for conducting toxicological research on environmental chemicals.

Effect of cooling method on TiN precipitation behavior of high-titanium high-strength steel during solidification

Metallographic microscopy,scanning electron microscopy and TiN growth thermodynamic and kinetic equations were used to investigate the morphology,quantity,and size of TiN in the center of high-titanium high-strength steels under different solidification cooling *** results showed that TiN in the center of the experimental steels mainly existed in three forms:single,composite(Al2O3-TiN),and multi-particle *** began precipitating at around 1497℃(solidification fraction of 0.74).From the end of melting to solidification for 180 s,the cooling rates in the center of the experimental steels for furnace cooling,air cooling,refractory mold cooling,and cast iron mold cooling tended to stabilize at 0.17,0.93,1.65,and 2.15℃/s,*** size of TiN in the center of the experimental steel cooled using furnace cooling was mainly concentrated in the 5-15 pm *** contrast,the size of TiN in the center of the experimental steels cooled using air cooling,refractory mold cooling,and cast iron mold cooling were mainly concentrated in the 1-5 pm *** addition,their density of TiN in the center of the experimental steels is signif-icantly higher than that of the furnace-cooled experimental *** and kinetic precipitation models of TiN established predicted the growth size of TiN in a high-titanium high-strength steel when the solidification cooling rates are not below 0.93℃/s.

The load-bearing performance(LBP)of pumpable supports(PPS)is crucial for the stability of longwall pre-driven recovery room(PRR)surrounding ***,the unbalanced bearing coefficient(UBC)of the PPS(undertaking unequal load along the mining direction)has not been investigated.A mechanical model of the PRR was established,considering the main roof cantilever beam structure,to derive an assessment formula for the load,the failure criteria,and the UBC of the ***,the generation mechanisms,and influencing factors of the UBC were *** sensitivity analysis shows that the main roof hanging length(l_(2))and the spacing between the PPS(r)significantly impact the UBC.A novel design of the PPS and the coupling control technology were proposed and applied to reduce the UBC of the PPS in the adjacent longwall *** results showed no failure of the PPS at the test site,with the UBC(ζ)reduced to 1.1 consistent with the design value(1.15)basically,fully utilizing the collaborative LBP of the ***,the maximum roof-to-floor convergence of the PRR was 234 mm,effectively controlling the stability of the surrounding rock of the PRR and ensuring the mining equipment recovery.

Effects of Ta content on microstructure and oxidation behavior of AlCoCrFeNiYTa_(x)high entropy alloy

The oxidation behavior of the Al CoCrFeNiYTa_(x)high entropy alloy(HEA)doped with different Ta contents at 1100℃is *** this study,different reasons for the spallation of the oxide scales of HEAs without/with excessive Ta addition after 1000 h of oxidation are discussed,and the most suitable Ta doping content for this HEA is explored,in which the HEA exhibits both an exceptionally low oxidation rate,and the formation of oxide scales composed of intact a-Al_(2)O_(3).Meanwhile,it is found that the addition of Ta can effectively inhibit the phase transition of β phase,which makes the surface of the oxide scale *** combining our experimental results with Wagner–Hauffe theory,the influence of Ta drag effect on oxide scale growth is explained,which provides some theoretical guidance for the subsequent design of AlCoCrFeNi HEAs,and can lead to the development of thermal barrier coatings at higher service temperature.

Approaching the theoretical capacity of TiO_(2) anode in a photorechargeable lithium-ion battery

New generation of lithium-ion batteries(LIBs)integrating solar energy conversion and storage is emerging,as they could solve the fluctuation problem in the utilization of solar ***-rechargeable lithium-ion batteries(PR-LIBs)are ideal devices for such target,in which solar energy is converted into electricity and stored in *** order to achieve the high performance of PR-LIB,it is crucial to develop dual-function electrode materials that can synergistically capture solar energy and store ***,we present photo-rechargeable lithium-ion batteries using defective black TiO_(2) as photoanode prepared by lithium *** photoanode exhibits excellent photo response in full solar spectrum with a capacity enhancement of 46.4%under illumination,corresponding to the energy conversion efficiency of 4.4%at the current density of 1 A·g^(-1).When illumination is applied at 20 mA·g^(-1),the battery capacity increases from~230 in dark to~349 mAh·g^(-1) at the first cycle,and then stabilizes at 310 mAh·g^(-1),approaching the theoretical value of 335 mAh·g^(-1) of TiO_(2) electrode *** finding provides thoughts for breaking the capacity limitations in TiO_(2) and paves the way for powering LIBs by solar illumination.

Sulfur doped iron-nitrogen-hard carbon nanosheets as efficient and robust noble metal-free catalysts for oxygen reduction reaction in PEMFC

Transition metal-nitrogen-carbon(M-N-C)as a promising substitute for the conventional noble metalbased catalyst still suffers from low activity and durability for oxygen reduction reaction(ORR)in proton exchange membrane fuel cells(PEMFCs).To tackle the issue,herein,a new type of sulfur-doped ironnitrogen-hard carbon(S-Fe-N-HC)nanosheets with high activity and durability in acid media were developed by using a newly synthesized precursor of amide-based polymer with Fe ions based on copolymerizing two monomers of 2,5-thiophene dicarboxylic acid(TDA)as S source and 1,8-diaminonaphthalene(DAN)as N source via an amination *** as-synthesized S-Fe-N-HC features highly dispersed atomic Fe Nxmoieties embedded into rich thiophene-S doped hard carbon nanosheets filled with highly twisted graphite-like microcrystals,which is distinguished from the majority of M-N-C with soft or graphitic carbon *** unique characteristics endow S-Fe-N-HC with high ORR activity and outstanding durability in 0.5 M H_(2)SO_(4).Its initial half-wave potential is 0.80 V and the corresponding loss is only 21 m V after 30,000 ***,its practical PEMFC performance is a maximum power output of 628.0 mW cm^(-2)and a slight power density loss is 83.0 m W cm^(-2)after 200-cycle practical ***,theoretical calculation shows that the activity of Fe Nxmoieties on ORR can be further enhanced by sulfur doping at meta-site near FeN_(4)*** results evidently demonstrate that the dual effect of hard carbon substrate and S doping derived from the precursor platform of amid-polymers can effectively enhance the activity and durability of Fe-N-C catalysts,providing a new guidance for developing advanced M-N-C catalysts for ORR.

Brazilin Actuates Ferroptosis in Breast Cancer Cells via p53/SLC7A11/GPX4 Signaling Pathway

Objective:To investigate the mechanism of induction of ferroptosis by brazilin in breast cancer ***:Breast cancer 4T1 cells were divided into 6 groups:control,brazilin 1/2 half maximal inhibitory concentration(IC50),IC50,2×IC50,erastin（10 μg/mL） and capecitabine（10 μg/mL） *** effect of brazilin on the proliferation of 4T1 cells was detected by cell counting kit-8 assay,and the treatment dose of brazilin was *** effect of brazilin on the mitochondrial morphology of 4T1 cells,and the mitochondrial damage was evaluated under electron *** levels of Fe2+,reactive oxygen species（ROS）,malondialdehyde（MDA）,glutathione（GSH） and glutathione peroxidase 4（GPX4） were estimated using various detection *** invasion and migration abilities of 4T1 cells were detected by scratch assay and transwell *** expressions levels of tumor protein p53, solute carrier family 7 member 11（SLC7A11）,GPX4 and acyl-CoA synthetase long-chain family member 4（ACSL4） proteins were quantified by Western blot ***:Compared to the control group,the 10(1/2 IC50),20(IC50) and 40(2×IC50) μg/mL brazilin,erastin,and capecitabine groups showed a significant decrease in the cell survival rate,invasion and migration abilities,GSH,SLC7A11 and GPX4 protein expression levels,and mitochondrial volume and ridge（P<0.05）,and a significant increase in the mitochondria membrane density,Fe2+,ROS and MDA levels,and p53 and ACSL4protein expression levels（P<0.05）.Conclusions:Brazilin actuated ferroptosis in breast cancer cells,and the underlying mechanism is mainly associated with the p53/SLC7A11/GPX4 signaling pathway.

The infection caused by porcine epidemic diarrhea virus(PEDV)is associated with high mortality in piglets *** factors involved in the efficient replication of PEDV,however,remain largely *** recent proteomic study in the virus-host interaction network revealed a significant increase in the accumulation of CALML5(EF-hand protein calmodulin-like 5)following PEDV infection.A further study unveiled a biphasic increase of CALML5 in 2 and 12 h after viral *** trends were observed in the intestines of piglets in the early and late stages of the PEDV ***,CALML5 depletion reduced PEDV mRNA and protein levels,leading to a one-order-of-magnitude decrease in virus *** the early stage of PEDV infection,CALML5 affected the endosomal trafficking pathway by regulating the expression of endosomal sorting complex related cellular ***5 depletion also suppressed IFN-βand IL-6 production in the PEDV-infected cells,thereby indicating its involvement in negatively regulating the innate immune *** study reveals the biological function of CALML5 in the virology field and offers new insights into the PEDV-host cell interaction.

Conventionally,the spatially structured light beams produced by metasurfaces primarily highlight the polarization modulation of the beams propagating along the optical axis or the beams'spatial transmission *** particular,along the optical axis,the polarization state is either constant or varies continuously in each output ***,we develop innovative spatially structured light beams with continually changing polarization along any arbitrary spatial transmission *** tri-layer metallic metasurfaces,the geometric characteristics of each layer structure can be adjusted to modulate the phase and polarization state of the incident terahertz(THz)*** beam will converge to the predefined trajectory along several paths to generate a Bessel-like beam with longitudinal polarization *** demonstrate the versatility of the approach by designing two THz-band structured light beams with varying polarization states along the spatial helical transmission *** linear polarization changes and linear polarization to right circular polarization(RCP)and back to linear polarization changes are realized *** experimental results are basically consistent with the simulated *** proposal for arbitrary trajectory structured light beams with longitudinally varying polarization offers a practical method for continuously regulating the characteristics of spatial structured light beams with non-axial *** technique has potential uses in optical encryption,particle manipulation,and biomedical imaging.
