The transcription factor FgNsf1 regulates fungal development,virulence and stress responses in Fusarium graminearum

Nutrient and stress factor 1(Nsf1),a transcription factor containing the classical Cys2-His2(C_(2)H_(2))zinc finger motif,is expressed under non-fermentable carbon conditions and in response to salt stress in Saccharomyces ***,the role of Nsf1 in filamentous fungi is not well *** this study,the orthologue of Nsf1 was investigated in Fusarium graminearum(named FgNsf1),a causal agent of Fusarium head blight(FHB).The functions of FgNsf1 were evaluated by constructing a FgNSF1 deletion mutant,designated asΔFgNsf1,and its functional complementation mutantΔ*** deletion experiments showed that the mycelial growth rate,asexual and sexual reproduction ofΔFgNsf1 were significantly reduced,but the pigment production ofΔFgNsf1 was remarkably increased compared with the PH-1 andΔ*** addition,the tolerance ofΔFgNsf1 to osmotic pressures,cell wall-damaging agents and oxidative stress increased *** tests to different fungicides revealed thatΔFgNsf1 exhibited increased sensitivity to carbendazim(MBC)and tebuconazole,and enhanced tolerance to fludioxonil and iprodione than PH-1 andΔ*** virulence ofΔFgNsf1 to wheat coleoptiles and flowering wheat heads were dramatically decreased,which was consistent with the decrease in the yield of deoxynivalenol(DON).All of these defects were restored by target gene *** results indicated that FgNsf1 plays a crucial role in vegetative growth,asexual and sexual reproduction,stress responses,fungicide sensitivity,and full virulence in ***.

NQO1 Mediates Lenvatinib Resistance by Regulating ROS-induced Apoptosis in Hepatocellular Carcinoma

Precisely optimizing the morphology of functional hybrid polymeric systems is crucial to improve its photophysical property and further extend their optoelectronic applications. The physic-chemical property of polymeric matrix in electrospinning (ES) processing is a key factor to dominate the condensed structure of these hybrid microstructures and further improve its functionality. Herein, we set a flexible poly(ethylene oxide) (PEO) as the matrix to obtain a series of polydiarylfluorenes (including PHDPF, PODPF and PNDPF) electrospun hybrid microfibers with a robust deep-blue emission. Significantly different from the rough morphology of their poly(N-vinylcarbazole) (PVK) ES hybrid fibers, polydiarylfluorenes/PEO ES fibers showed a smooth morphology and small size with a diameter of 1∼2 µm. Besides, there is a relatively weak phase separation under rapid solvent evaporation during the ES processing, associated with the hydrogen-bonded-assisted network of PEO in ES fibers. These relative “homogeneous” ES fibers present efficient deep-blue emission (PLQY>50%), due to weak interchain aggregation. More interestingly, low fraction of planar (β) conformation appears in the uniform PODPF/PEO ES fibers, induced by the external traction force in ES processing. Meanwhile, PNDPF/PEO ES fibers present a highest sensitivity than those of other ES fibers, associated with the smallest diameter and large surface area. Finally, compared to PODPF/PVK fibers and PODPF/PEO amorphous ES fibers, PODPF/PEO ES fibers obtained from DCE solution exhibit an excellent quenching behavior toward a saturated DNT vapor, mainly due to the synergistic effect of small size, weak separation, β-conformation formation and high deep-blue emission efficiency.

Patient-derived organoids potentiate precision medicine in advanced clear cell renal cell carcinoma

To investigate the role of patient-derived organoid(PDO)model in the precision medicine of advanced clear cell renal cell carcinoma(ccRCC),we retrospectively analyzed the clinical data of seven cases of ccRCC diagnosed by operation and pathology in Renji Hospital from September 2021 to September *** seven patients were diagnosed with advanced ccRCC with or without remote *** and radical nephrectomy was performed *** predict the response to immunotherapy and provide personalized medicine recommendation,a PDO model based on air-liquid interface system was established from the surgical resected tumor and subsequent drug screening was *** and eosin(H&E)staining and immunohistochemistry revealed that the PDO recapitulated the histological feature of parent *** staining identified that CD3^(+)T cells,SMA^(+)cancer associated fibroblasts,and CD31^(+)endothelial cells were preserved in PDO *** activated cell sorter(FACS)revealed an evidently increased ratio of CD8^(+)/CD4^(+)T cells and apoptotic tumor cells in PDO treated with toripalimab than those treated with *** results showed that toripalimab is able to rescue the excessive death of CD8^(+)T cells by critically reversing the immune exhaustion state of ccRCC in PDO *** research validated that PDO is a promising and faithful preclinical model for prediction of immunotherapy response in patients with ccRCC.

Synthesis of Mg-Al-carbonate layered double hydroxide by an atom-economic reaction

A clean method for preparing layered double hydroxides （LDHs） has been developed, featured by using the hydroxides of two different metals as starting materials by atom-economic reactions. The reactions were carried out under hydrothermal conditions in either a high pressure autoclave or a microwave digester. The compositions, structural parameters and thermal behavior of the resulting LDHs are very similar to those of materials produced by using the separate nucleation and aging steps （SNAS） method. The major advantage of the new method is that no by-product is produced, so that filtration and washing processes are unnecessary. The consequent reduction in water consumption is beneficial to the environment.

Solution chemistry of carbonate minerals and its effects on the flotation of hematite with sodium oleate

The effects of carbonate minerals（dolomite and siderite） on the flotation of hematite using sodium oleate as a collector were investigated through flotation tests, supplemented by dissolution measurements, solution chemistry calculations, zeta-potential measurements, Fourier transform infrared（FTIR） spectroscopic studies, and X-ray photoelectron spectroscopy（XPS） analyses. The results of flotation tests show that the presence of siderite or dolomite reduced the recovery of hematite and that the inhibiting effects of dolomite were stronger. Dissolution measurements, solution chemistry calculations, and flotation tests confirmed that both the cations（Ca^（2＋） and Mg^（2＋）） and CO_3^（2＋）ions dissolved from dolomite depressed hematite flotation, whereas only the 23CO-ions dissolved from siderite were responsible for hematite depression. The zeta-potential, FTIR spectroscopic, and XPS analyses indicated that Ca^（2＋）, Mg^（2＋）, and CO_3^（2-）（HCO_3^-） could adsorb onto the hematite surface, thereby hindering the adsorption of sodium oleate, which was the main reason for the inhibiting effects of carbonate minerals on hematite flotation.

Differential Responses of Nitrifier and Denitrifier to Dicyandiamide in Short- and Long-Term Intensive Vegetable Cultivation Soils

Nitrification inhibitors, such as dicyandiamide （DCD）, have been shown to decrease leaching from urea- and ammoniumbased fertilizers in agricultural soils. The effect of nitrification inhibitors on nitrifier and denitrifier in short- and long-term intensive vegetable cultivation soils was poorly understood. In this study, the pot trial was conducted to investigate the differential responses of nitrifier （amoA-containing bacteria） and denitrifier （nirK-containing bacteria） to DCD in short-（soil S） and long-term （soil L） intensive vegetable cultivation soils. Quantitative polymerase chain reaction （qPCR） and terminal restriction fragment length polymorphism （T-RFLP） were employed to detect the abundance and composition of amoA- and nirK-containing communities. The results indicated that application of DCD led to a consistently higher NH4＋-N concentration during the whole incubation in soil L, while it was quickly decreased in soil S after 21 days. Furthermore, DCD induced more severe decrease of the abundance of amoA-containing bacteria in soil L than in soil S. However, the abundance of the nirK- containing community was not significantly affected by DCD in both soils. Long-term vegetable cultivation resulted in a super-dominant amoA-containing bacteria group and less divergence in soil L compared with soil S, and DCD did not cause obvious shifts of the composition of ammonia-oxidising bacteria （AOB）. On the contrary, both amoA- and nirK-containing bacterial compositions were influenced by DCD in soil S. The results suggested that long-term intensive vegetable cultivation with heavy nitrogen fertilization resulted in significant shifts of AOB community, and this community was sensitive to DCD, but denitrifiers were not clearly affected by DCD.

Rice Homeobox Transcription Factor HOX1a Positively Regulates Gibberellin Responses by Directly Suppressing EL1

Homeobox transcription factors are involved in various aspects of plant development, including maintenance of the biosynthesis and signaling pathways of different hormones. However, few direct targets of homeobox proteins have been identified. We here show that overexpression of rice homeobox gene HOXla resulted in enhanced gibberellin （GA） response, indicating a positive effect of HOXla in GA signaling. HOXla is induced by GA and encodes a homeobox transcription factor with transcription repression activity. In addition, HOXla suppresses the transcription of early flowering1 （EL1）, a negative regulator of GA signaling, and further electrophoretic mobility shift assay and chromatin immunoprecipitation analysis revealed that HOXla directly bound to the promoter region of EL1 to suppress its expression and stimulate GA signaling. These results demonstrate that HOXla functions as a positive regulator of GA signaling by suppressing EL1, providing informative hints on the study of GA signaling.

Bacterial and archaeal communities in the surface sediment from the northern slope of the South China Sea

Microbial diversity of sediments from the northern slope of the South China Sea was studied by constructing bacterial and archaeal 16S rRNA gene clone libraries. Fourteen bacterial phylogenetic groups were detected, including Gammaproteobacteria, Deltaproteobacteria, Planctomycetes, Alphaproteobacteria, Firmicutes, Bacteroidetes, Actinobacteria, Chloroflexi, Acidobacteria, Betaproteobacteria, Nitrospirae, candidate divisions OP8 and OP 11, and an unknown group. Gammaproteobacteria was the predominant group in bacterial libraries with the percentage ranging from 31.8% to 63.2%. However, archaeal libraries had relatively lower diversity, with most clones belonging to marine archaeal group I uncultured Crenarehaeota. In addition, two novel euryarchaeal clones were detected not to match any culture-dependent or -independent isolates. Compared with other gas hydrate-rich ecosystems and different areas of the South China Sea, a distinct microbial community was revealed in this study.

Anilido-imine Zinc Complexes： Efficient Catalysts for Ring-opening Polymerization ofL-Lactide

A series of anilido-imine zinc complexes ortho-C6H4（CH：NAr）（NAr）ZnEt[Ar=2,6-Me2C6H3（2a）; Ar=2,6-Et2C6H3（2b）] was synthesized, characterized and used as catalysts for the ring-opening polymerization of L-lactide in the presence of benzyl alcohol. The effects of the structures of catalysts and reaction conditions on the reactivity were investigated. The polymerization of L-lactide initiated by each of these complexes was found to take place with a controlled manner.
