Leptin Modulates the Differentiation of Keratinocytes via Autophagy in Psoriasis Patients with Metabolic Syndrome

Obj.ctive: Psoriasis is associated with a high prevalence of metabolic syndrome (MS), and patients with concomitant psoriasis and MS are more severely affected and less responsive to treatment. However, the molecular mechanisms behind these effects are unknown. Recent studies have shown that leptin may serve as a molecular link between psoriasis and MS, suggesting that high leptin concentrations may ex.cerbate psoriasis. However, the molecular mechanism of this effect is still unclear. We aimed to investigate the effect of leptin on autophagy in patients with psoriasis. Methods: From j.nuary 2021 to j.ne 2022 in PLA General Hospital, we enrolled 51 patients with psoriasis, including 21 patients with MS and 30 without MS, and 30 healthy controls who had undergone nevus surgery. We measured the epidermal leptin, P62, and LC3B concentrations of patients by immunohistochemistry, and measured the serum leptin concentration by enzyme-linked immunosorbent assay. We then performed correlation analyses to compare these proteins' concentrations between patients with concomitant psoriasis and MS, patients with psoriasis alone, and healthy control groups. Additionally, we performed western blotting after in vitro culture of HaCaT cells with different concentrations of leptin and measured the ex.ression levels of the autophagy markers Beclin1, LC3B, and P62; the differentiation markers K10, K16, and K17; and PI3K/AKT/mTOR signaling pathway-related proteins of HaCat cells. Nex., we transfected ATG5 into HaCaT cells to revert autophagy and used the specific PI3K inhibitor LY294002 to block PI3K/AKT/mTOR signaling. The ex.ression levels of K10, K16, and K17 of HaCat cells were again measured. One-way analysis of variance was used for the comparison of means of multiple samples, and LSD-t post hoc test was used for comparison between the 2 groups. Granger causal representation learning for groups of time series

Discovering causality from multivariate time series is an important but challenging *** ex.sting methods focus on estimating the Granger causal structures among multivariate time series,while ignoring the prior knowledge of these time series, e.g., the group of the time series. Focusing on discovering the Granger causal structures among groups of time series, we propose a Granger causal representation learning method to solve this problem. First, we use the multiset canonical correlation analysis method to learn the Granger causal representation of each group of time series. Then, we model the Granger causal relationships among the learned Granger causal representations using a recurrent neural network with temporal information. Finally, we formulate the above two stages into one unified optimization problem, which is efficiently solved using the augmented Lagrangian method. We conduct ex.ensive ex.eriments on synthetic and real-world datasets to validate the correctness and effectiveness of the proposed method.

Elite Dung Beetle Optimization Algorithm for Multi-UAV Cooperative Search in Mountainous Environments

Sri Lankan cassava mosaic virus(SLCMV)is a prominent causative agent of cassava mosaic disease in Asia and relies on the whitefly Bemisia tabaci cryptic complex.for its ***,the molecular mechanisms involved in SLCMV transmission by *** have yet to be *** this study,we identified an aminopeptidase N-like protein(BtAPN)in *** Asia II 1,an efficient vector of SLCMV,which is involved in the SLCMV transmission *** the use of glutathione S-transferase pull-down assay and LC-MS/MS analysis,we demonstrated the interaction between BtAPN and the coat protein(CP)of *** interaction was further confirmed in vitro,and we observed an induction of BtAPN gene ex.ression following SLCMV *** interfering with the function of BtAPN,the quantities of SLCMV were significantly reduced in various parts of *** Asia II 1,including the whole body,midgut,hemolymph,and primary salivary ***,we discovered that BtAPN is conserved in *** Middle East-Asia Minor 1(MEAM1)and interacts with the CP of tomato yellow leaf curl virus(TYLCV),a begomovirus known to cause severe damage to tomato *** BtAPN with antibody led to a significant reduction in the quantities of TYLCV in whitefly whole body and organs/*** results demonstrate that BtAPN plays a generic role in interacting with the CP of begomoviruses and positively regulates their acquisition by the whitefly.
