Endoplasmic reticulum stress and autophagy in cerebral ischemia/reperfusion injury:PERK as a potential target for intervention

Several studies have shown that activation of unfolded protein response and endoplasmic reticulum(ER)stress plays a crucial role in severe cerebral ischemia/reperfusion *** occurs within hours after cerebral ischemia,but the relationship between ER stress and autophagy remains *** this study,we established experimental models using oxygen-glucose deprivation/reoxygenation in PC12 cells and primary neurons to simulate cerebral ischemia/reperfusion *** found that prolongation of oxygen-glucose deprivation activated the ER stress pathway protein kinase-like endoplasmic reticulum kinase(PERK)/eukaryotic translation initiation factor 2 subunit alpha(e IF2α)-activating transcription factor 4(ATF4)-C/EBP homologous protein(CHOP),increased neuronal apoptosis,and induced ***,inhibition of ER stress using inhibitors or by si RNA knockdown of the PERK gene significantly attenuated excessive autophagy and neuronal apoptosis,indicating an interaction between autophagy and ER stress and suggesting PERK as an essential target for regulating *** autophagy with chloroquine exacerbated ER stress-induced apoptosis,indicating that normal levels of autophagy play a protective role in neuronal injury following cerebral ischemia/reperfusion *** from this study indicate that cerebral ischemia/reperfusion injury can trigger neuronal ER stress and promote autophagy,and suggest that PERK is a possible target for inhibiting excessive autophagy in cerebral ischemia/reperfusion injury.

Severe ground-level ozone(O_(3))pollution over major Chinese cities has become one of the most challenging problems,which have deleterious effects on human health and the sustainability of *** study explored the spatiotemporal distribution characteristics of ground-level O_(3) and its precursors based on conventional pollutant and meteorological monitoring data in Zhejiang Province from 2016 to ***,a high-performance convolutional neural network(CNN)model was established by expanding the moment and the concentration variations to general ***,the response mechanism of O_(3) to the variation with crucial influencing factors is explored by controlling variables and interpolating target *** results indicated that the annual average MDA8-90th concentrations in Zhejiang Province are higher in the northern and lower in the *** the wind direction(WD)ranges from east to southwest and the wind speed(WS)ranges between 2 and 3 m/sec,higher O_(3) concentration prone to *** different temperatures(T),the O_(3) concentration showed a trend of first increasing and subsequently decreasing with increasing NO_(2) concentration,peaks at the NO_(2) concentration around 0.02mg/m^(3).The sensitivity of NO_(2) to O_(3) formation is not easily affected by temperature,barometric pressure and dew point ***,there is a minimum IRNO_(2) at each temperature when the NO_(2) concentration is 0.03 mg/m^(3),and this minimum IRNO_(2) decreases with increasing *** study explores the response mechanism of O_(3) with the change of driving variables,which can provide a scientific foundation and methodological support for the targeted management of O_(3) pollution.

Background:Balloon catheter isolation is a promising auxiliary method for thermal ablation treatment of liver cancer. We aimed to explore the safety and effectiveness of balloon catheter isolation-assisted ultrasound-guided percutaneous microwave ablation (MWA) in treating liver cancer in difficult anatomical ***:Data of 132 patients with 145 difficult-site liver cancer treated with ultrasound-guided percutaneous MWA were retrospectively analyzed. Participants were classified into the isolation (n=40)and non-isolation (n=92) groups based on whether the patients were treated using a balloon catheter prior to ablation. The major complication rates, local tumor residuals (LTR), and tumor follow-up for local tumor progression (LTP) at 6 and 12 months post-ablation were compared between the two ***:The rates of major postoperative complications did not significantly differ between the isolation and non-isolation groups (2.5%vs. 4.3%, P=0.609). The postoperative LTR rates were significantly different between the isolation and non-isolation groups (4.8%vs. 17.5%, P=0.032). Balloon catheter isolation [odds ratio (OR)=0.225, 95%confidence interval (CI):0.085–0.595, P=0.009] and tumor diameter(OR=2.808, 95%CI:1.186–6.647, P=0.019) were identified as independent factors influencing LTR *** cumulative LTP rates at 6 and 12 months after ablation showed no significant differences between the isolation and non-isolation groups (2.6%vs. 7.9%, P=0.661;4.9%vs. 9.8%, P=0.676, respectively).Cox proportional hazards regression analysis showed that tumor diameter was an independent risk factor for cumulative LTP rate (OR=3.445, 95%CI:1.406–8.437, P=0.017).Conclusions:Balloon catheter isolation-assisted MWA was safe and effective in the treatment of difficultsite liver cancer. Additionally, tumor diameter significantly influenced LTR and LTP rates after ablation.

Polychlorinated naphthalenes(PCNs)are detrimental to human health and the *** the commercial production of PCNs banned,unintentional releases have emerged as a significant environmental ***,relevant information is still *** this study,provincial emissions for eight PCNs homologues from 37 sources in the Chinese mainland during the period of 1960-2019 were estimated based on a source-specific and time-varying emission factor *** results showed that the total PCNs emissions in 2019 reached 757.0 kg with Hebei ranked at the top among all the provinces and iron&steel industry as the biggest ***-chlorinated PCNs comprised 90%of emissions by mass,while highly chlorinated PCNs dominated in terms of toxicity,highlighting divergent priorities for mitigating emissions and safeguarding human *** emissions showed an overall upward trend from 1960 to 2019 driven by emission increase from iron&steel industry in terms of source,and from North China and East China in terms of geographic ***-capita emissions followed an inverted U-shaped environmental Kuznets curvewhile emission intensities decreased with increasing per-capita Gross Domestic Product(GDP)following a nearly linear pattern when log-transformed.

Localization has become an indispensable function of modern cellular communication systems [1]. In a cloud radio access network(C-RAN), the remote radio head(RRH) is the actual signal transmitter, whose location can assist in wireless network layout optimization and wireless resource management. The measurement of the RRH location can only be done manually on-site due to the lack of global positioning systems(GPS) at the RRH side, which is thus not a cost-effective solution for the widely used C-RAN with a large number of RRH units.

We explore the decay properties of the isovector and isoscalar 1-+light hybrids,π1and η1,in N _f=2 lattice QCD at a pion mass mπ≈417 *** McNeile and Michael method is adopted to extract the effective couplings for individual decay modes,which are used to estimate the partial decay widths ofπ1（1600） and η1（1855） by assuming SU（3） *** partial decay widths ofπ1（1600） are predicted to be (Γb1π,Γf1（1285）π,Γρπ,ΓK*K)=(325±75,O（10）,52±7,8.6±1.3) Me V,and the total width is estimated to be 396±90 MeV,considering only statistical *** η1（1855） and the 4.4σsignal observed by BESIII (labeled as η1（2200）) are taken as the two mass eigenstates of the isoscalar 1-+light hybrids in SU（3）,then the dominant decay channel（s）of η1（1855）(η1（2200）) is K1（1270）K (K1（1270）K and K1（1400）K) through the 1+（-）0-（+）*** vector-vector decay modes are also significant for the two η*** the mixing angle α≈22.7°obtained from lattice QCD for the two η1states,the total widths are estimated to beΓη1（1855）=282（85） MeV andΓη1（2200）=455（143） Me *** former is compatible with the experimental width of η1（1855）.Although many systematic uncertainties are not well controlled,these results are qualitatively informative for the experimental search for light hybrids.

Previous studies have demonstrated that cis9， trans11， cis15-CLNA (c9， t11， c15-CLNA)， a metabolite of Bifidobacterium breve CCFM683， possesses anti-inflammatory properties in the intestine. However， its safety profile remains unexplored. This study establishes the safe dose range for c9， t11， c15-CLNA and investigate the potential damage pathways. The results indicate that the maximum safe dose for mice is lower than 625 mg/kg， primarily due to hepatocyte damage. In cellular experiments revealed that an overdose of c9， t11， c15-CLNA activates peroxisome proliferator-activated receptor gamma (PPAR-γ)， leading to hepatic lipid accumulation. The addition of the PPAR-γ inhibitor partially mitigated this effect. In summary， this study established that the maximum tolerated dose of c9， t11， c15-CLNA in healthy adult C57BL/6N mice is 625 mg/kg. For human application， the estimated safe dose is below 50.62 mg/kg. Additionally， c9， t11， c15-CLNA was found to activate the PPARγ protein in the liver， leading to lipid accumulation within hepatic tissue.

It has been reported that the PI3K/AKT signaling pathway plays a key role in the pathogenesis of ischemic *** a result,the development of drugs targeting the PI3K/AKT signaling pathway has attracted increasing attention from *** article reviews the pathological mechanisms and advancements in research related to the signaling pathways in ischemic stroke,with a focus on the PI3K/AKT signaling *** key findings include the following:(1)The complex pathological mechanisms of ischemic stroke can be categorized into five major types:excitatory amino acid toxicity,Ca^(2+)overload,inflammatory response,oxidative stress,and apoptosis.(2)The PI3K/AKT-mediated signaling pathway is closely associated with the occurrence and progression of ischemic stroke,which primarily involves the NF-κB,NRF2,BCL-2,mTOR,and endothelial NOS signaling pathways.(3)Natural products,including flavonoids,quinones,alkaloids,phenylpropanoids,phenols,terpenoids,and iridoids,show great potential as candidate substances for the development of innovative anti-stroke medications.(4)Recently,novel therapeutic techniques,such as electroacupuncture and mesenchymal stem cell therapy,have demonstrated the potential to improve stroke outcomes by activating the PI3K/AKT signaling pathway,providing new possibilities for the treatment and rehabilitation of patients with ischemic *** investigations should focus on the direct regulatory mechanisms of drugs targeting the PI3K/AKT signaling pathway and their clinical translation to develop innovative treatment strategies for ischemic stroke.
