Fast and reliable localization of high-energy transients is crucial for characterizing the burst properties and guiding the follow-up *** based on the relative counts of different detectors has been widely used for all-sky gamma-ray *** are two major methods for this count distribution localization:χ^(2)minimization method and the Bayesian *** we propose a modified Bayesian method that could take advantage of both the accuracy of the Bayesian method and the simplicity of the χ^(2)*** comprehensive simulations,we find that our Bayesian method with Poisson likelihood is generally more applicable for various bursts than the χ^(2)method,especially for weak *** further proposed a location-spectrum iteration approach based on the Bayesian inference,which could alleviate the problems caused by the spectral difference between the burst and location *** method is very suitable for scenarios with limited computation resources or timesensitive applications,such as in-flight localization software,and low-latency localization for rapidly follow-up observations.

We demonstrate radiation-pressure-driven mechanical oscillations from high optical quality factor silica microdisk resonators on chip. Mechanical quality factors of 3520 in air and 12540 in vacuum for the fundamental radial breathing modes are obtained from 73 μm-diarneter silica microdisks with mechanical frequencies of -51 MHz. The measured mechanical oscillation threshold powers for the input light are determined to be 62.5 μW in air and down to 26.6 μW in vacuum.

目的探讨香青兰总黄酮(total flavonoids of Dracocephalum Moldavica L.,TFDM)抗大鼠动脉粥样硬化(atherosclerosis,AS)及三甲胺氮氧化物(trimethylamine N-oxide,TmaO)加剧的小鼠巨噬细胞RAW264.7炎症的保护作用及其可能的作用机制。方法采用高脂饲料喂养联合腹腔注射维生素D3的方法建立SD大鼠AS模型,分为对照组、模型组、辛伐他汀组(15 mg·kg^(-1))、TFDM组(60、30、15 mg·kg^(-1)),建模同时进行给药处理,持续8周。生化检测方法检测血清总胆固醇(TC)、甘油三酯(TG)、低密度脂蛋白胆固醇(LDL-C)水平。HE染色检测主动脉组织病理变化,ELISA试剂盒检测血清中TmaO、IL-1β、IL-6及肝脏组织中TNF-α的表达,Western blot法检测主动脉中JAK、STAT、TNF-α蛋白的表达。此外,体外培养RAW264.7巨噬细胞,采用LPS+TmaO建立巨噬细胞炎症模型,TFDM(100、50、25 mg·L^(-1))进行干预。CCK-8测定细胞活力及增殖,RT-qPCR法检测细胞中TNF-α、IL-6、JAK、STAT mRNA的表达。结果TFDM可明显下调大鼠血清TC、TG、LDL-C水平及血清TmaO、IL-1β、IL-6和肝脏TNF-α的水平,减少主动脉的斑块沉积,下调主动脉中TNF-α、JAK、STAT的蛋白表达。此外,TFDM干预可明显下调TNF-α、IL-6、JAK、STAT mRNA的表达及JAK、STAT蛋白的表达。结论TFDM可降低血清中TmaO的含量,从而抑制JAK/STAT炎症信号通路,减缓炎症的发生,发挥抗AS作用。

Objective: To examine the anti-inflammatory effect of grape seed extract(GSE) in animal and cellular models and explore its mechanism of action. Methods: This study determined the inhibitory effect of GSE on macrophage inflammation and Th1 and Th17 polarization in vitro. Based on the in vitro results, the effects and mechanisms of GSE on multiple sclerosis(MS)-experimental autoimmune encephalomyelitis(EAE) mice model were further explored. The C57BL/6 mice were intragastrically administered with 50 mg/kg of GSE once a day from the 3rd day to the 27th day after immunization. The activation of microglia, the polarization of Th1 and Th17and the inflammatory factors such as tumor necrosis factor-α(TNF-α), interleukin-1β(IL-1β), IL-6, IL-12, IL-17and interferon-γ(IFN-γ) secreted by them were detected in vitro and in vivo by flow cytometry, enzyme linked immunosorbent assay(ELISA), immunofluorescence staining and Western blot, respectively. Results: GSE reduced the secretion of TNF-α, IL-1β and IL-6 in bone marrow-derived macrophages stimulated by lipopolysaccharide(Pdemyelination and inflammatory cell infiltration. Peripherally, GSE downregulated the expression of toll-like-receptor 4(TLR4) and Rho-associated kinase(ROCKⅡ, Pmatory factors(Pmatory factors(P<0.01), and prevented the activation of microglia(P<0.05).
