Diagnostic Value of GDF10 for the Tumorigenesis and Immune Infiltration in Lung Squamous Cell Carcinoma

The first pandemic wave of coronavirus disease 2019(COVID-19)induced a considerable increase in several antivirals and antibiotics in surface *** common symptoms of COVID-19 are viral and bacterial infections,while comorbidities(e.g.,hypertension and diabetes)and mental shock(e.g.,insomnia and anxiety)are ***,little is known about the long-term impacts of comorbidities and mental shock on organic micropollutants(OMPs)in surface ***,we monitored 114 OMPs in surface water and wastewater treatment plants(WWTPs)in Wuhan,China,between 2019 and *** pandemic-induced OMP pollution in surface water was confirmed by significant increases in 26 OMP *** increases in four antihypertensives and one diabetic drug suggest that the treatment of comorbidities may induce OMP ***,cotinine(a metabolite of nicotine)increased 155 times to 187 ngL1,which might be associated with increased ***,the increases in zolpidem and sulpiride might be the result of worsened insomnia and ***,it is reasonable to note that mental-health protecting drugs/behavior also contributed to OMP *** the observed OMPs,telmisartan,lopinavir,and ritonavir were associated with significantly higher ecological risks because of their limited WWTP-removal rate and high *** study provides new insights into the effects of comorbidities and mental shock on OMPs in surface water during a pandemic and highlights the need to monitor the fate of related pharmaceuticals in the aquatic environment and to improve their removal efficiencies in WWTPs。

Remote estimates of suspended particulate matter in global lakes using machine learning models

Suspended particulate matter(SPM)in lakes exerts strong impact on light propagation,aquatic ecosystem productivity,which co-varies with nutrients,heavy metal and micro-pollutant in *** lakes,SPM exerts strong absorption and backscattering,ultimately affects water leaving signals that can be detected by satellite *** regression models based on specific band or hand ratios have been widely used for SPM estimate in the past with moderate *** are still rooms for model accuracy improvements,and machine learning models may solve the non-linear relationships between spectral variable and SPM in *** assembled more than 16,400 in situ measured SPM in lakes from six continents(excluding the Antarctica continent),of which 9640 samples were matched with Landsat overpasses within±7 *** machine learning algorithms and two simple regression methods(linear and partial least squares models)were used to estimate SPM in lakes and the performance were *** overcome the problem of imbalance datasets in regression,a Synthetic Minority Over-Sampling technique for regression with Gaussian Noise(SMOGN)was adopted in this *** comparison,we found that gradient boosting decision tree(GBDT),random forest(RF),and extreme gradient boosting(XGBoost)models demonstrated good spatiotemporal transferability with SMOGN processed dataset,and has potential to map SPM at different year with good quality of Landsat land surface reflectance *** all the tested modeling approaches,the GBDT model has accurate calibration(n=6428,R^(2)=0.95,MAPE=29.8%)from SPM collected in 2235 lakes across the world,and the validation(n=3214,R^(2)=0.84,MAPE=38.8%)also exhibited stable ***,the good performances were also exhibited by RF model with calibration(R^(2)=0.93)and validation(R^(2)=0.86,MAPE=24.2%)*** applied GBDT and RF models to map SPM of typical lakes,and satisfactory result was *** addition,the GBDT model was evaluated b

Construction and evaluation of a liver cancer risk prediction model based on machine learning

BACKGROUND Liver cancer is one of the most prevalent malignant tumors worldwide,and its early detection and treatment are crucial for enhancing patient survival rates and quality of ***,the early symptoms of liver cancer are often not obvious,resulting in a late-stage diagnosis in many patients,which significantly reduces the effectiveness of *** a highly targeted,widely applicable,and practical risk prediction model for liver cancer is crucial for enhancing the early diagnosis and long-term survival rates among affected *** To develop a liver cancer risk prediction model by employing machine learning techniques,and subsequently assess its *** In this study,a total of 550 patients were enrolled,with 190 hepatocellular carcinoma(HCC)and 195 cirrhosis patients serving as the training cohort,and 83 HCC and 82 cirrhosis patients forming the validation *** regression(LR),support vector machine(SVM),random forest(RF),and least absolute shrinkage and selection operator(LASSO)regression models were developed in the training *** performance was assessed in the validation ***,this study conducted a comparative evaluation of the diagnostic efficacy between the ASAP model and the model developed in this study using receiver operating characteristic curve,calibration curve,and decision curve analysis(DCA)to determine the optimal predictive model for assessing liver cancer *** Six variables including age,white blood cell,red blood cell,platelet counts,alpha-fetoprotein and protein induced by vitamin K absence or antagonist II levels were used to develop LR,SVM,RF,and LASSO regression *** RF model exhibited superior discrimination,and the area under curve of the training and validation sets was 0.969 and 0.858,*** values significantly surpassed those of the LR(0.850 and 0.827),SVM(0.860 and 0.803),LASSO regression(0.845 and 0.831),and ASAP(0.866 and 0.813)model

Acetaminophen overdose-induced acute liver injury can be alleviated by static magnetic field

Acetaminophen(APAP),the most frequently used mild analgesic and antipyretic drug worldwide,is implicated in causing 46%of all acute liver failures in the USA and between 40%and 70%in *** predominant pharmacological intervention approved for mitigating such overdose is the antioxidant N-acetylcysteine(NAC);however,its efficacy is limited in cases of advanced liver injury or when administered at a late *** the current study,we discovered that treatment with a moderate intensity static magnetic field(SMF)notably reduced the mortality rate in mice subjected to high-dose APAP from 40%to 0%,proving effective at both the initial liver injury stage and the subsequent recovery *** the early phase of liver injury,SMF markedly reduced APAPinduced oxidative stress,free radicals,and liver damage,resulting in a reduction in multiple oxidative stress markers and an increase in the antioxidant glutathione(GSH).During the later stage of liver recovery,application of vertically downward SMF increased DNA synthesis and hepatocyte ***,the combination of NAC and SMF significantly mitigated liver damage induced by high-dose APAP and increased liver recovery,even 24 h post overdose,when the effectiveness of NAC alone substantially ***,this study provides a noninvasive non-pharmaceutical tool that offers dual benefits in the injury and repair stages following APAP *** note,this tool can work as an alternative to or in combination with NAC to prevent or minimize liver damage induced by APAP,and potentially other toxic overdoses.

Screening of Chinese giant salamander meat hydrolysates with DPP-IV inhibitory activity and systematic elucidation of their hypoglycemic functions in mouse model

Type 2 diabetes (T2D) presents a significant health challenge， underscoring the need for functional foods and nutraceutical hypoglycemic bioactive peptides for its prevention. This study investigates the potential of proteolytic hydrolysate from artificially cultivated Chinese giant salamander (CGS) meat， a rich protein source， as a preventive strategy for T2D. We produced a CGS meat hydrolysate (CGSh) and demonstrated its ability to inhibit the T2D drug target DPP-IV through in vitro assays. We identified five peptides (WRPPDH， WAPPSKD， IPDSPF， IPEMIF， and VPIAVPT) with high DPP-IV inhibitory activity in CGSh， suggesting its potential antidiabetic effects. In vivo experiments showed that CGSh effectively reduced insulin resistance in mice induced with a high-fat diet， as evidenced by a slower increase in blood glucose levels and a decreased HOMA-IR index. 16S rRNA sequencing analysis revealed that CGSh improved gut microbial homeostasis， promoting beneficial microorganisms and reducing harmful bacteria. Metabolomic analyses identified an increase in valeric acid levels and highlighted nine potential biomarker metabolites. By inhibiting metabolic pathways such as AGE-RAGE， CGSh may also prevent diabetic complications and reduce inflammation. These findings suggest that CGSh has a promising hypoglycemic effect， making it a potential functional food ingredient for T2D prevention and treatment.
