The first pandemic wave of coronavirus disease 2019(COVID-19)induced a considerable increase in several antivirals and antibiotics in surface *** common symptoms of COVID-19 are viral and bacterial infections,while comorbidities(e.g.,hypertension and diabetes)and mental shock(e.g.,insomnia and anxiety)are ***,little is known about the long-term impacts of comorbidities and mental shock on organic micropollutants(OMPs)in surface ***,we monitored 114 OMPs in surface water and wastewater treatment plants(WWTPs)in Wuhan,china,between 2019 and *** pandemic-induced OMP pollution in surface water was confirmed by significant increases in 26 OMP *** increases in four antihypertensives and one diabetic drug suggest that the treatment of comorbidities may induce OMP ***,cotinine(a metabolite of nicotine)increased 155 times to 187 ngL1,which might be associated with increased ***,the increases in zolpidem and sulpiride might be the result of worsened insomnia and ***,it is reasonable to note that mental-health protecting drugs/behavior also contributed to OMP *** the observed OMPs,telmisartan,lopinavir,and ritonavir were associated with significantly higher ecological risks because of their limited WWTP-removal rate and high *** study provides new insights into the effects of comorbidities and mental shock on OMPs in surface water during a pandemic and highlights the need to monitor the fate of related pharmaceuticals in the aquatic environment and to improve their removal efficiencies in WWTPs。

●AIM:To evaluate the role of semaphorin 7A(Sema7A)and its associated regulatory mechanisms in modulating the barrier function of cultured human corneal epithelial cells(HCEs).●METHODS:Barrier models of HCEs were treated with recombinant human Sema7A at concentrations of 0,125,250,or 500 ng/mL for 24,48,or 72h in *** electrical resistance(TEER)as well as Dextran-fluorescein isothiocyanate(FITC)permeability assays were conducted to assess barrier *** quantify tight junctions(TJs)such as occludin and zonula occludens-1(ZO-1)at the mRNA level,reverse transcriptionpolymerase chain reaction(RT-PCR)analysis was *** was used to examine the activity of the nuclear factor-kappa B(NF-κB)signaling pathway and the production of TJs *** analyses were employed to localize the ***-linked immunosorbent assay(EliSA)and RT-PCR were utilized to observe changes in interleukin(IL)-1β*** investigate the role of NF-κB signaling activation and IL^(-1)βin Sema7A’s anti-barrier mechanism,we employed 0.1μmol/L IκB kinase 2(IKK2)inhibitor IV or 500 ng/mL IL^(-1)receptor(IL-1R)antagonist.●RESULTS:Treatment with Sema7A resulted in decreased TEER and increased permeability of Dextran-FITC in HCEs through down-regulating mRNA and protein levels of TJs in a time-and dose-dependent manner,as well as altering the localization of ***,Sema7A stimulated the activation of inhibitor of kappa B alpha(IκBα)and expression of IL-1β.The anti-barrier function of Sema7A was significantly suppressed by treatment with IKK2 inhibitor IV or IL-1R antagonists.●CONCLUSION:Sema7A disrupts barrier function through its influence on NF-κB-mediated expression of TJ proteins,as well as the expression of IL-1β.These findings suggest that Sema7A could be a potential therapeutic target for the diseases in corneal epithelium.

Aflatoxin B1(AFB1) is a carcinogenic toxin naturally produced in most food crops that severely threaten human health， and effective methods are urgent to improve the detection accuracy. Herein an indirect competitive immunosorbent approach was elaborately developed based on high-affinity immunoglobulin g (Igg) coupled CuO-anchored Fe3O4nanozymes for precise and ultrasensitive detection of AFB1in food crops including peanut， corn and wheat. The high-affinity nanozymes were fabricated by the assembly of inner core Fe3O4nanoparticles (Fe3O4NPs) and mesoporous silica capping layer， CuO further situated within large aperture of the coating layer via in-situ growth， and then conjugated with ligand rabbit anti-mouse Igg， which can specifically bind with AFB1. The results showed the hybrid high-affinity nanozymes displayed enhanced peroxidase (POD)-mimic activities and catalytic performances， achieving a linear range of 0.06-61.93 ng/mL and a detection limit of 0.0037 ng/mL， 30 times better than that of the conventional EliSA method. The constructed nanozymes were successfully applied to the detection of AFB1in food products with an average spiked recovery of 96.53% and relative standard deviations (RSDs) less than 2.8%. Therefore， the accurate hybrid nanozymes may serve for AFB1detection in various foods in future.

Choline acetyltransferase(ChAT)-positive neurons in neural stem cell(NSC)niches can evoke adult neurogenesis(AN)and restore impaired brain function after injury,such as acute ischemic stroke(AIS).However,the relevant mechanism by which ChAT+neurons develop in NSC niches is poorly *** RNA-seq analysis revealed that dimethylarginine dimethylaminohydrolase 1(DDAH1),a hydrolase for asymmetric ng,ng-dimethylarginine(ADMA),regulated genes responsible for the synthesis and transportation of acetylcholine(ACh)(Chat,Slc5a7 and Slc18a3)after stroke *** dual-luciferase reporter assay further suggested that DDAH1 controlled the activity of ChAT,possibly through hypoxia-inducible factor 1α(HIF-1α).KC7F2,an inhibitor of HIF-1α,abolished DDAH1-induced ChAT expression and suppressed *** expected,DDAH1 was clinically elevated in the blood of AIS patients and was positively correlated with AIS *** comparing the results among Ddah1 general knockout(KO)mice,transgenic(Tg)mice and wild-type(WT)mice,we discovered that DDAH1 upregulated the proliferation and neural differentiation of NSCs in the subgranular zone(SgZ)under ischemic *** a result,DDAH1 may promote cognitive and motor function recovery against stroke impairment,while these neuroprotective effects are dramatically suppressed by NSC conditional knockout of Ddah1 in mice.

Based on the continuum-based discrete element method (CDEM), this paper presents a computational' model to simulate fractured materials. The relationship of macroscopic failure of material and internal rupture strain strength is exploited. First, the CDEM treats the 8-node solid isoparametric element in FEM as a fundamental system composed of 8 particles and interconnecting springs. A complete process from continuum to discontinuum under loading can be simulated by CDEM. Second, a spring changes its properties if this spring is intersected by a fracture: 1, the shear and tension forces are equal to zero on fracture plane when the spring in normal direction of the fracture plane is in tension. 2, When the spring is in compression, the normal force in the fracture plane is the projection of spring force in normal direction, and the shear force in the fracture plane is the one in tangential direction. When the normal and shear forces satisfy the Mohr-Coulomb failure criterion, the spring fails and the spring stiffness should be revalued. Third, the stress to strain curve for fractured samples are tested under tension and compression. It is found that the material performs linear elasticity when fracture density is small and external load is low. However, nonlinearity occurs with the larger load. Furthermore, the 'hardening' and 'softening' property of fractured material is simulated under displacement loading test.
