Machine learning identifies the risk of complications after laparoscopic radical gastrectomy for gastric cancer

ADU-S100/多柔比星原位疫苗在弥漫大B细胞淋巴瘤中的作用及机制研究

目的探讨ADU-S100/多柔比星原位疫苗在弥漫大B细胞淋巴瘤中的抗肿瘤作用及相关机制。方法选择6周龄雌性BALB/c小鼠,采用小鼠B细胞淋巴瘤A20细胞株构建小鼠B细胞淋巴瘤双侧皮下肿瘤模型,采用随机数字表法将皮下荷瘤小鼠随机分为未治疗组(不接受任何治疗)、ADU-S100原位疫苗治疗组(瘤内注射干扰素基因刺激因子激动剂ADU-S100)、多柔比星原位疫苗治疗组(瘤内注射多柔比星)和ADU-S100/多柔比星原位疫苗治疗组(瘤内注射ADU-S100+多柔比星),每组5只。将荷瘤小鼠右侧肿瘤定义为近端肿瘤,左侧肿瘤定义为远端肿瘤,仅对近端肿瘤进行瘤内药物注射治疗,远端肿瘤不处理。肿瘤接种后第23天,采用流式细胞术检测各组小鼠脾脏中CD11c+树突细胞(DC)、CD8+CD11c+DC和CD80+CD11c+DC亚群的比例。采用A20肿瘤细胞裂解物体外刺激各组小鼠的脾细胞,应用流式细胞术检测各原位疫苗治疗组CD8+T细胞中5’’-乙炔基-2’’脱氧尿嘧啶核苷阳性(EdU+)细胞和肿瘤坏死因子α阳性(TNF-α+)细胞的比例,并使用乳酸脱氢酶(LDH)细胞毒性检测试剂盒测定各组细胞毒性T淋巴细胞(CTL)的杀伤效应。在接种肿瘤后第7、12、17天通过腹腔注射抗小鼠CD8α(clone 53-6.7)抗体或同型对照抗体清除ADU-S100/多柔比星原位疫苗治疗小鼠体内的CD8+细胞。肿瘤接种后第23天,测量ADU-S100/多柔比星原位疫苗联合抗小鼠CD8α(clone 53-6.7)抗体或同型对照抗体处理组小鼠近端和远端肿瘤体积,并采用流式细胞术检测两组小鼠脾脏中CD8+T细胞和CD8+CD11c+DC的比例,免疫组织化学染色法检测两组小鼠肿瘤组织中浸润的CD8+T细胞。结果在肿瘤接种后第11、14、17、20、23天,各治疗组小鼠的近端肿瘤、远端肿瘤体积均低于未治疗组小鼠(均P<0.05)。未治疗组、ADU-S100原位疫苗治疗组、多柔比星原位疫苗治疗组和ADU-S100/多柔比星原位疫苗治疗组小鼠脾脏中募集的CD11c+DC亚群比例分别为(4.92±0.63)%、(7.54±0.84)%、(7.45±0.86)%和(11.63±0.85)%,差异有统计学意义(F=72.30,P<0.001);CD8+CD11c+DC亚群比例分别为(1.36±0.34)%、(4.02±0.43)%、(4.22±0.61)%和(6.11±0.73)%,差异有统计学意义(F=76.09,P<0.001);CD80+CD11c+DC亚群比例分别为(0.51±0.24)%、(1.69±0.23)%、(1.82±0.25)%和(4.09±0.39)%,差异有统计学意义(F=167.40,P<0.001)。各原位疫苗治疗组的CTL杀伤效应以及CD8+T细胞中EdU+细胞和TNF-α+细胞的比例均高于未治疗组(均P<0.05),ADU-S100/多柔比星原位疫苗治疗组的CTL杀伤效应以及CD8+T细胞中EdU+细胞和TNF-α+细胞的比例均高于ADU-S100原位疫苗治疗组和多柔比星原位疫苗治疗组(均P<0.05)。ADU-S100/多柔比星原位疫苗+抗小鼠CD8α抗体处理组小鼠脾脏中CD8+T细胞和CD8+CD11c+DC细胞的比例均低于ADU-S100/多柔比星原位疫苗+同型对照抗体处理组(均P<0.05),ADU-S100/多柔比星原位疫苗+抗小鼠CD8α抗体处理组小鼠近端和远端肿瘤体积均大于ADU-S100/多柔比星原位疫苗+同型对照抗体处理组(均P<0.05)。结论ADU-S100/多柔比星原位疫苗可抑制小鼠B细胞淋巴瘤双侧皮下肿瘤模型中近端肿瘤的生长,并可产生能够部分抑制远端肿瘤生长的全身性抗肿瘤免疫效应,ADU-S100/多柔比星原位疫苗可能通过CD8+CD11c+DC介导的CD8+T细胞免疫应答发挥抗肿瘤免疫效应。

Aim: To determine the expression of estrogen receptor-α (ERα) and estrogen receptor-β (ERβ) in prostate cancer (PCa) and to investigate the relationship between expression of ER and clinico-pathologic features of prostaric carcinoma. Methods: Thirty-two cases of PCa, 12 cases of normal prostate tissue (NOR) and 32 cases of benign prostate hyperplasia (BPH) were analyzed for the expression of ERα and ERβ by using semi-quantitative reverse transcription polymerase chain reaction (RT-PCR), and the products were sequenced. Results: The comparison of the normal, hyperplastic and tumor prostate tissues indicated an overexpression of ERa in tumor specimens (P<0.01) . On the contrary, the expression of ERβ was significantly reduced in tumor tissues compared with that in normal and hyperplastic specimens (P<0.01), suggesting that severe clinico-pathologic features of PCa were associated with lower ERβ expression. Spearmen analysis showed a significant negative correlation between ERβ expression and tumor stage and grade (P<0.05 ), and a significant positive correlation between ERα expression and tumor stage, grade, respectively (P<0.01) . Our analysis also showed that hormone-refractory PCa (HRPC), compare to hormone-dependent PCa (HDPC), displayed a decreased expression of ERβ (P<0.01) and an increased expression of ERα. Conclusions: The results suggest that ERα and ERβ may play an important role in the development of PCa. The loss or decrease of ERβ expression is associated with higher Gleason grade tumors and PCa with higher metastatic potential. Our data show that the loss of ERβ could be one of the key elements leading to uncontrolled growth of prostate epithelial cells.

AIM:To analyze the clinical manifestations and risk fac-tors of complications in laparoscopic low anterior resection(LAR)for rectal cancer ***:A series of 132 consecutive patients who received laparoscopic LAR for rectal cancer in our center were *** etiology,diagnosis,treatment and prevention of rectal cancer were studied among the patients with surgery-related complications using both univariate and multivariate regression ***:No conversion to open surgery was observed and 5 cases converted to hand-assisted laparoscopic *** overall morbidity rate was 20.5%.Complications occurred during the operation in 7 patients(5.3%),within 30 postoperative days in 24 patients(18.2%),and within 3 mo in 2 patients(1.5%).The most significant complications were anastomotic leak-age(9.1%)and anastomotic hemorrhage(5.3%).Sizeand location of tumor,pathological staging and preoperative nutrition were significant factors associated with LAR complications,while gender,age and pathological type showed no *** logistics regression showed that the size and location of tumor,and pathological staging were independent factors of laparoscopic *** the complications were treated during their onset of clinical manifestations by interventional or conservative ***:Anastomotic leakage is a major com-plication in laparoscopic *** complications may be associated with tumor size and site,and pathological *** therapies are of value in the management of laparoscopic LAR complications.

Aim: To determine whether pranlukast, a cysteinyl leukotriene receptor-lantagonist, exerts an anti-inflammatory effect on focal cerebral ischemia in ***: Focal cerebral ischemia in mice was induced by permanent middle cere-bral artery occlusion(MCAO). In addition to neurological deficits, infarct volume,degenerated neurons and endogenous IgG exudation, we detected accumulationof neutrophils and macrophage/microglia in the ischemic brain tissue 72 h afterMCAO. Pranlukast was ip injected 30 min before and after MCAO. Results:Pranlukast significantly attenuated neurological deficits, infarct volume, neurondegeneration and IgG exudation. Importantly, pranlukast(0.01 and 0.1 mg/kg)inhibited myeloperoxidase-positive neutrophil, but not CD11b-positive macroph-age/microglial accumulation in the ischemic cortical tissue. Conclusion: Pranlukastexerts an anti-inflammatory effect on focal cerebral ischemia in the subacute phasethat is limited to neutrophil recruitment through the disrupted blood-brain barrier.

Neutron resonance imaging(NRI)has recently emerged as an appealing technique for neutron *** complexity surpasses that of conventional transmission imaging,as it requires a high demand for both a neutron source and ***,the progression of NRI technology has been sluggish since its inception in the 1980s,particularly considering the limited studies analyzing the neutron energy range above *** white neutron source(Back-n)at the china Spallation Neutron Source(CSNS)provides favorable beam conditions for the development of the NRI technique over a wide neutron energy range from eV to ***-sensitive microchannel plates(MCP)have emerged as a cutting-edge tool in the field of neutron detection owing to their high temporal and spatial resolutions,high detection efficiency,and low *** this study,we report the development of a 10B-doped MCP detector,along with its associated electronics,data processing system,and NRI experiments at the *** heavy elements such as gold,silver,tungsten,and indium can be easily identified in the transmission images by their characteristic resonance peaks in the 1–100 eV energy range;the more difficult medium-weight elements such as iron,copper,and aluminum with resonance peaks in the 1–100 keV energy range can also be *** particular,results in the neutron energy range of dozens of keV(Aluminum)are reported here for the first time.
