Gene expression must be precisely regulated in cells and functional nucleic acids are the most widely utilized tools for gene *** of how these nucleic acid tools work in the cellular environment can precisely manipulate gene expression through a non-invasive *** we report a methodology on multiplex photocontrol of functional nucleic acids to achieve totally temporal and orthogonal regulation of gene expression in living *** select two functional nucleic acid systems as examples,DNAzyme and CRISPR/Cas9,and demonstrate the power of light control for precise gene manipulation by rational design of chemically modified oligonucleotides through introduction of two photocleavable *** the previous modulation of functional nucleic acids by simply activating or deactivating,we successfully achieve versatile controlling patterns using light as the governing *** design represents a generalized pathway towards the photo-controllable functional nucleic acids,which greatly enriches the toolbox for optogenetic studies.

Deformation energy of tectonic coal under hydrostatic conditions:A new calculation model based on critical state theory

Human neural stem cell-derived extracellular vesicles protect against ischemic stroke by activating the PI3K/AKT/mTOR pathway

Chattering-Free Fault-Tolerant Cluster Control and Fault Direction Identification for HIL UAV Swarm With Pre-Specified Performance

In this paper, the problem of pre-specified performance fault-tolerant cluster consensus control and fault direction identification is solved for the human-in-the-loop(HIL) swarm unmanned aerial vehicles(UAVs) in the presence of possible nonidentical and unknown direction faults(NUDFs) in the yaw *** control strategy begins with the design of a pre-specified performance event-triggered observer for each individual *** observers estimate the outputs of the human controlled UAVs, and simultaneously achieve the distributed design of actual control signals as well as cluster consensus of the observer *** is worth mentioning that these observers require neither the high-order derivatives of the human controlled UAVs' output nor a priori knowledge of the initial conditions. The fault-tolerant controller realizes the pre-specified performance output regulation through error transformation and the Nussbaum function. It should be pointed out that there are no chattering caused by the jump of the Nussbaum function when a reverse fault occurs. In addition, to provide a basis for further solving the problem of physical malfunctions, a fault direction identification algorithm is proposed to accurately identify whether a reverse fault has occurred. Simulation results verify the effectiveness of the proposed control and fault direction identification strategies when the reverse faults occur.

Expression significance of biomarker MORC4 in colorectal cancer patients and its relationship with pathological features and prognosis

BACKGROUND Colorectal cancer(CRC)is one of the most common malignant gastrointestinal tumors worldwide,with high incidence and mortality *** To investigate the expression significance of the chromatin-remodeling protein MORC family CW-type zinc finger 4(MORC4)as a biomarker in CRC patients,and to explore its relationship with pathological features and *** A total of 143 CRC specimens and 57 adjacent tissue specimens,surgically removed from our hospital between January 2020 and January 2021,were ***4 protein expression was assessed using immunohistochemistry after paraffin *** relationship between MORC4 protein expression and clinicopathological characteristics of patients was ***-Meier survival curves were plotted to analyze the relationship between MORC4 protein expression and prognosis in CRC *** Compared with adjacent tissues,the expression rate of MORC4 protein in CRC tissues was significantly higher(P0.05).However,significant differences were found in the high expression rate of MORC4 protein in patients with different degrees of differentiation,lymph node metastasis,distant metastasis,tumor-lymph node-metastasis stage,and serum carcinoembryonic antigen levels(P<0.05).Compared with patients with low MORC4 expression,patients with high MORC4 expression had a worse prognosis(P<0.05).CONCLUSION The upregulation of MORC4 expression in CRC patients is closely related to disease severity and prognosis,suggesting its potential as an evaluation biomarker,which warrants further investigation.

Deformation and failure characteristics of gas drainage drilling-reaming coal mass in non-uniform stress field

Clarifying the internal relationship between pore structure and methane adsorption and diffusion in coal is the basis for quantitative evaluation of methane desorption. The multi-scale pores in coal were quantitatively characterized by the low-pressure nitrogen and carbon dioxide adsorption methods, and the occurrence sites and migration channels of adsorbed and free methane were determined as filling pores with size of 0.38～1.5 nm and diffusion pores with size of 1.5～100 nm, respectively. A dual-pore series diffusion model of adsorbed methane in filling pores and free methane in diffusion pores was established. The methane diffusion coefficient and pore structure parameters were obtained by fitting the methane desorption data of coal particles. The results indicate that the calculated methane limiting adsorption based on multi-scale pore methane adsorption model matches the measured value well. More than 90% of adsorbed methane is stored in filled pores in the form of micropore filling, and the methane adsorption ability in coal depends on the filled pores. During methane desorption, the adsorbed methane first diffuses in filled pores, and then flows out of coal in free phase through the diffusion pores. The adsorbed and free methane diffusion coefficients are in the order of magnitude of 10−13 m2/s and 10−6 m2/s, the equivalent pore lengths of filling and diffusion pores are 0.109～2.855 μm and 0.525～3.106 mm, and the corresponding equivalent pore numbers per unit mass of coal are 1016～1017 g and 1010～1012 g, respectively. Due to more filled pores with shorter pore lengths and higher adsorbed methane concentration, the mass transfer rate of adsorbed methane in coal is much faster than that of free methane in diffusion pores, and the methane desorption in coal is mainly controlled by the free methane diffusion process in diffusion pores. © 2025 China Coal Society. Random integration of massive distributed photovoltaic(PV)generation poses serious challenges to distribution *** violations,line overloads,increased peak-valley differences,and power-flow reversals can occur at different locations,times,and *** planning methods such as energy storage(ES)allocation and upgrading of lines may result in poor economics and low equipment *** study proposes a distribution-network planning strategy that coordinates three planning mechanisms:ES allocation to substations and to feeders,and line *** proposed strategy achieved the lowest annual cost,considering the distribution network problems at different locations,times,and severities.A net load curve description method was first proposed considering economic planning and the uncertain distribution of loads and ***,a multistage optimization model for ES allocation and line upgrading was established based on the net load ***,an improved IEEE33 node system was used in the case *** planning results show that the proposed coordinated planning strategy can address the problems caused by the random integration of massive distributed PVs,improve equipment utilization,and obtain the lowest total annual planning cost.

目的:探讨改良无管化经皮肾镜碎石取石术(percutaneous nephrolithotomy,PCNL)治疗2~3 cm肾下盏结石后肾下盏解剖结构变化及对术后清石率的影响。方法:选取2020年9月至2023年9月西南医科大学附属医院泌尿外科收治的300例2~3 cm肾下盏结石患者作为研究对象,经改良无管化PCNL治疗术后疗效将患者分为清除组(n=253)和残余组(n=47),对2组患者一般资料进行单因素分析,采用限制性三次样条回归法分析肾下盏解剖结构参数变化与术后结石残余的关系;采用逐步logistic回归分析进行影响因素分析,并基于此构建列线图预测模型,绘制校准曲线、受试者工作特征(receiver operating characteristic,ROC)曲线对模型的诊断效能进行评估。结果:2组患者在肾功能不全史(P=0.017)、肾下盏长度(infundibular length,IL)(P<0.001)、结石数量(P=0.025)、结石CT值(P=0.001)、肾下盏宽度(infundibular width,IW)(P<0.001)、手术时间(P=0.002)、肾盂漏斗部夹角(infundibulopelvic angle,IPA)(P<0.001)、肾盂肾下盏高度(pelvic caliceal height,PCH)(P<0.001)、血尿酸(uric acid,UA)(P<0.001)、尿素氮(blood urea nitrogen,BUN)(P=0.014)方面差异有统计学意义(P<0.05);logistic回归显示调整后IL、PCH、IW、IPA仍是术后结石残余的危险因素;逐步logistic回归分析结果显示结石数量、结石CT值、IL、PCH是影响患者经改良无管化PCNL治疗肾下盏结石残余的独立危险因素,IW、IPA是保护因素(均P<0.05);利用以上影响因素进行列线图预测模型构建,得到回归方程为Logit(P)=-2.567+0.1109×X_(1)+1.170×X_(2)-0.226×X3+0.023×X4-0.442×X5+0.714×X6,其一致性指数0.893(95%CI=0.878~0.901),ROC曲线分析显示,模型AUC为0.888(95%CI=0.869~0.897),灵敏度为0.827,特异度为0.795,区分度较好;校准曲线提示该模型准确度较高。结论:改良无管化PCNL治疗2~3 cm肾下盏结石是一种安全有效的方法,肾下盏结石患者可通过充分考虑解剖结构方面影响因素进行合理制定治疗方案,从而进行针对性预防,提高患者术后结石清除率。
