Dopaminergic effects on in vitro osteogenesis

Multiple growth factors（e.g., BMP2, TGF-b1, FGF2） and isolated genes have been shown to improve osteoblastic proliferation and mineralization, advancing bone tissue engineering. Among these factors, both polydopamine（PDA） and dopamine（DA） monomer have recently been reported to increase osteoblast proliferation and mineralization in vitro. Although a well-characterized neurotransmitter, DA＇s role in the bone is unknown. We hypothesize that DA can directly act on osteoblasts, and examined whether osteoblasts express DA receptors that respond to exogenous DA. m RNAs and protein cell lysates were obtained from MC3T3-E1 cells during osteogenic differentiation phase. Reverse transcription polymerase chain reaction and western blot analysis were used to examine the expression of DA receptors, D1–D5. Dose-response effect and time course of DA treatment on cell proliferation, mineralization, and osteogenic differentiation were investigated at pre-determined days. Real-time PCR was performed to investigate whether DA affects osteogenic gene expression（ALP, BSP, OC, OSX, RUNX2, and Collagen1a2） with or without receptor antagonists（SCH233390 and GR103691）. Two-way ANOVA was used for statistical analysis. All five DA receptors（D1, D2, D3, D4, and D5） m RNAs and proteins were expressed in MC3T3-E1 cells. DA treatment increased cell proliferation for up to 7 days（P, 0.05）. Osteogenic mineralization was significantly greater in the DA-treated group than control group（P, 0.05）. Finally, expression of all the osteogenic genes was inhibited by DA receptor antagonists for D1, D3, and D5. Our findings suggest that MC3T3-E1 osteoblasts express functional DA receptors that enhance proliferation and mineralization. PDA is not biologically inert and has important implications in orthopedic applications. Furthermore, osteoblast differentiation might be regulated by the nervous system, presumably during bone development, remodeling, or repair.

Towards precision medicine: from quantitative imaging to radiomics

Radiology（imaging） and imaging-guided interventions, which provide multi-parametric morphologic and functional information, are playing an increasingly significant role in precision medicine. Radiologists are trained to understand the imaging phenotypes, transcribe those observations（phenotypes） to correlate with underlying diseases and to characterize the images. However, in order to understand and characterize the molecular phenotype（to obtain genomic information） of solid heterogeneous tumours, the advanced sequencing of those tissues using biopsy is required. Thus, radiologists image the tissues from various views and angles in order to have the complete image phenotypes, thereby acquiring a huge amount of data. Deriving meaningful details from all these radiological data becomes challenging and raises the big data issues. Therefore, interest in the application of radiomics has been growing in recent years as it has the potential to provide significant interpretive and predictive information for decision support. Radiomics is a combination of conventional computer-aided diagnosis, deep learning methods, and human skills, and thus can be used for quantitative characterization of tumour phenotypes. This paper discusses the overview of radiomics workflow, the results of various radiomics-based studies conducted using various radiological images such as computed tomography（CT）, magnetic resonance imaging（MRI）, and positron-emission tomography（PET）, the challenges we are facing, and the potential contribution of radiomics towards precision medicine.

Endolymphatic sac tumors(ELSTs) are rare, papillary adenomatous tumors that arise from the endothelium of the endolymphatic sac. We demonstrate a difficult case of endolymphatic sac tumor and how it is managed via transcanal endoscopic assisted technique, with discussion of feasibility of transcanal approach to lateral skull base tumor.

Industrial-scale Fixed-bed Coal Gasification: Modeling, Simulation and Thermodynamic Analysis

We have developed a process model to simulate the behavior of an industrial-scale pressurized Lurgi fixed-bed coal gasifier using Aspen Plus and General Algebraic Modeling System(GAMS). Reaction characteristics in the fixed-bed gasifier comprising four sequential reaction zones—drying, pyrolysis, combustion and gasification are respectively modeled. A non-linear programming(NLP) model is developed for the pyrolysis zone to estimate the products composition which includes char, coal gases and distillable liquids. A four-stage model with restricted equilibrium temperature is used to study the thermodynamic equilibrium characteristics and calculate the composition of syngas in the combustion and gasification zones. The thermodynamic analysis shows that the exergetic efficiency of the fixed-bed gasifier is mainly determined by the oxygen/coal ratio. The exergetic efficiency of the process will reach an optimum value of 78.3% when the oxygen/coal and steam/coal mass ratios are 0.14 and 0.80, respectively.

Lycium barbarum, a traditional Chinese anti-aging herb, has been shown to protect retinal ganglion cells （RGCs） in a rat chronic ocular hypertension （COH） model. Here, we investigated the expression of endothelin-1 （ET-1）, a strong vasoconstrictor, and its receptors, ETA and ETB, in the COH model and assessed the effects of Lycium barbarum on the ET-1 axis. Elevated intraocular pressure （IOP） was induced in the right eye of SD rats using argon laser photocoagulation. （1） The expression of ET-1, ETA and ETB in normal and COH retinas was studied. （2） Some COH rats were fed daily with Lycium barbarum Polysaccharides （LBP） using 1 mg/kg or phosphate-buffered saline （PBS） for 3 weeks （started 1 week before photocoagulation）. The effects of LBP on the expression of ET-1 and its receptors, ETA and ETB, in COH retina were evaluated. A semi-quantitative analysis of staining intensity was used to evaluate the expression levels of ET-1, ETA and ETB in retinal vasculature. We found that （1） Under COH condition, the immunoreactivity of ET-1 was increased in retina associated with an increase of ETB receptor immunoreactivity and a decrease of ETA receptor immunoreactivity. （2） After feeding COH rats with LBP, the expression of ET-1 was decreased with an increase of ETA expression and a decrease of ETB expression in the retina, especially in RGCs. （3） By comparing the staining intensity in the vasculature of COH retina in LBP-fed group with PBS-fed group, there was a decrease in the expression of ET-1 and ETA and an increase in ETB. In summary, ET-1 expression was up-regulated in the retina in COH model. LBP could decrease the expression of ET-1 and modulate the expression of its receptors, ETA and ETB, under the condition of COH. The neuroprotective effect of LBP on RGCs might be related to its ability to regulate the ET-1-mediated biological effects on RGCs and retinal vasculature.

Sequential treatment with bortezomib plus dexamethasone followed by autologous hematopoietic stem cell transplantation in patients with multiple myeloma

Background Whether the sequential treatment with bortezomib plus dexamethasone （BD） followed by autologous hematopoietic stem cell transplantation （ASCT） could extend the overall survival period in multiple myeloma patients is still not clear. Few large case studies about this therapeutics in multiple myeloma were reported in China. Our purpose was to assess the efficacy and adverse effects of sequential treatment with BD chemotherapy and ASCT in patients with multiple myeloma. Methods Fifty-three patients with newly diagnosed or relapsed/refractory multiple myeloma received BD as induction therapy before ASCT. Stem-cell mobilization was undertaken with cyclophosphamide 3-5 g/m2 plus granulocyte colony-stimulating factor 300 IJg/d. Target yield was 2.0x106 CD34＋ cells/kg. Conditioning for ASCT consisted of melphalan 200 mg/m2. Thalidomide and/or Q-interferon was used as post-transplantation maintenance treatment. Results The BD chemotherapy before transplantation was effective in 86.7% of the 53 patients, including 22.6% with complete remission （CR）, 39.6% with near complete remission （nCR）, and 24.5% with partial remission （PR）. The best effect was achieved after two treatment courses. Most bortezomib-related adverse effects were classes 1-2. All patients were successfully mobilized after BD for autologous peripheral blood stem cell transplantation. The ASCT was effective in 96.3% of patients, including 49.1% with CR, 32.1% with nCR, and 15.1% with PR. The CR rate was significantly increased （49.1% vs. 22.6%, P 〈0.05） by sequential ASCT. Within 27 （range, 6-53） months of follow-up, the efficacy of ASCT was maintained in 29 patients and further enhanced by post-transplantation maintenance treatment in four patients. Eleven patients died after transplantation. Among the patients undergoing BD/ASCT treatment, overall survival （OS） was significantly better in newly diagnosed patients in comparison to relapsed/refractory patients （P=0.046）. 旅游是一种文化体验的方式,无论旅者在前往一地旅游的目地为何,在旅游的过程中,与旅游接待地文化上的接触、影响、交融,是必然的。旅者就像一个载体,为两地的文化带来碰撞和影响。因此,旅游本身就具有文化传播的能力。马来西亚旅者对于北京本身的印象,主要来自各个层面的传播媒介,透过这些管道获取的信息,自然就会在人们心中建立起可能是正面,也可能是负面的印象。相较其他传播方式,旅游本身有着独特的优势,除了能够通过旅游传播文化符合旅游者旅游的目的外,游客旅途中愉悦的心理对于异域文化的接受,是偏向正面的。本文从跨文化旅游的内涵、旅游跨文化传播的特点以及旅游跨文化的重要性,探讨了苹果旅游行程设计、新旧媒体对北京形象的建构。透过描述性统计分析了旅游如何影响人们对中国北京形象的认知和偏见等内容,并透过问卷调查,阐明跨文化旅游能顺利改写旅客对北京的固有印象及重新建构旅客对北京的印象,并且带动了何种话语权。除此之外,跨文化旅游也可以让马来西亚旅客打破了 "身份认同"之下对中国北京的"向往"。而从发出的问卷数据上确实发现,旅客虽到北京的目的各有不同,但进行的内容全都是重要的文化交流元素。过程中,还包括了人际交流传播形式,人与人之间的沟通与交流。问卷调查发现,透过旅游这种跨文化传播途径,确实是能够将直接左右旅者对旅游目的地的印象。而笔者也祈望通过研究得知跨文化传播的反思和建议,以期让马来西亚摒弃对中国北京的负面印象,达到两国之间的旅游业能够蓬勃发展。

With the increasing incidence of heavy rainfall events,particularly over the monsoon regions,the highly dense populations are more vulnerable[1].Research initiatives on observation,modeling,and prediction of monsoon heavy rainfall have been promoted actively by World Weather Research Programme's(WWRP)Working Group on Tropical Meteorology Research(WGTMR)of the World Meteorological Organization(WMO)since *** of monsoon-heavy-rainfall workshops were held in Beijing(2011),Petaling Jaya(2012),and New Delhi(2015)to benefit scientists worldwide and forecasters from the National Meteorological and Hydrological *** international Research and Development Project,namely,the Southern China Monsoon Rainfall Experiment(SCMREX)[2]was established in 2013 to coordinate field campaign experiments and to conduct scientific research on presummer(April-June)heavy rainfall processes in southern China.
