Mechanistic links between gut microbial community dynamics, microbial functions and metabolic health

Gut microbes comprise a high density, biologically active community that lies at the interface of an animal with its nutritional environment. Consequently their activity profoundly influences many aspects of the physiology and metabolism of the host animal. A range of microbial structural components and metabolites directly interact with host intestinal cells and tissues to influence nutrient uptake and epithelial health. Endocrine, neuronal and lymphoid cells in the gut also integrate signals from these microbial factors to influence systemic responses. Dysregulation of these host-microbe interactions is now recognised as a major risk factor in the development of metabolic dysfunction. This is a two-way process and understanding the factors that tip host-microbiome homeostasis over to dysbiosis requires greater appreciation of the host feedbacks that contribute to regulation of microbial community composition. To date, numerous studies have employed taxonomic profiling approaches to explore the links between microbial composition and host outcomes(especially obesity and its comorbidities), but inconsistent host-microbe associations have been reported. Available data indicates multiple factors have contributed to discrepancies between studies. These include the high level of functional redundancy in host-microbiome interactions combined with individual variation in microbiome composition; differences in study design, diet composition and host system between studies; and inherent limitations to the resolution of r RNA-based community profiling. Accounting for these factors allows for recognition of the common microbial and host factors driving community composition and development of dysbiosis on high fat diets. New therapeutic intervention options are now emerging.

Dynamic modeling of a three-dimensional braided composite thin plate considering braiding directions

Currently, there are a limited number of dynamic models available for braided composite plates with large overall motions, despite the incorporation of three-dimensional(3D) braided composites into rotating blade components. In this paper, a dynamic model of 3D 4-directional braided composite thin plates considering braiding directions is established. Based on Kirchhoff's plate assumptions, the displacement variables of the plate are expressed. By incorporating the braiding directions into the constitutive equation of the braided composites, the dynamic model of the plate considering braiding directions is obtained. The effects of the speeds, braiding directions, and braided angles on the responses of the plate with fixed-axis rotation and translational motion, respectively, are investigated. This paper presents a dynamic theory for calculating the deformation of 3D braided composite structures undergoing both translational and rotational motions. It also provides a simulation method for investigating the dynamic behavior of non-isotropic material plates in various applications.

Answering Reachability Queries on Incrementally Updated Graphs by Hierarchical Labeling Schema

we proposed a novel solution schema called the Hierarchical Labeling Schema （HLS） to answer reachability queries in directed graphs. Different from many existing approaches that focus on static directed acyclic graphs （DAGs）, our schema focuses on directed cyclic graphs （DCGs） where vertices or arcs could be added to a graph incrementally. Unlike many of the traditional approaches, HLS does not require the graph to be acyclic in constructing its index. Therefore, it could, in fact, be applied to both DAGs and DCGs. when vertices or arcs are added to a graph, the HLS is capable of updating the index incrementally instead of re-computing the index from the scratch each time, making it more efficient than many other approaches in the practice. The basic idea of HLS is to create a tree for each vertex in a graph and link the trees together so that whenever two vertices are given, we can immediately know whether there is a path between them by referring to the appropriate trees. we conducted extensive experiments on both real-world datasets and synthesized datasets. we compared the performance of HLS, in terms of index construction time, query processing time and space consumption, with two state-of-the-art methodologies, the path-tree method and the 3-hop method. we also conducted simulations to model the situation when a graph is updated incrementally. The performance comparison of different algorithms against HLS on static graphs has also been studied. Our results show that HLS is highly competitive in the practice and is particularly useful in the cases where the graphs are updated frequently.

Energy Transfer Mechanism and Probability Analysis of Submarine Pipe Laterally Impacted by Dropped Objects

Energy transfer ratio is the basic-factor affecting the level of pipe damage during the impact between dropped object and submarine pipe. For the purpose of studying energy transfer and damage mechanism of submarine pipe impacted by dropped objects, series of experiments are designed and carried out. The effective yield strength is deduced to make the quasi-static analysis more reliable, and the normal distribution of energy transfer ratio caused by lateral impact on pipes is presented by statistic analysis of experimental results based on the effective yield strength, which provides experimental and theoretical basis for the risk analysis of submarine pipe system impacted by dropped objects. Failure strains of pipe material are confirmed by comparing experimental results with finite element simulation. In addition, impact contact area and impact time are proved to be the major influence factors of energy transfer by sensitivity analysis of the finite element simulation.

Design and mathematical analysis of activating transcriptional amplifiers that enable modular temporal control in synthetic juxtacrine circuits

The ability to control mammalian cells such that they self-organize or enact therapeutic effects as desired has incredible *** only would it further our understanding of native processes such as development and the immune response,but it would also have powerful applications in medical fields such as regenerative medicine and *** control is typically obtained by synthetic circuits that use synthetic receptors,but control remains *** synthetic juxtacrine receptors(SJRs)are widely used as they are fully modular and enable spatial control,but they have limited gene expression amplification and temporal *** these are integral facets to cell control,I therefore designed transcription factor based amplifiers that amplify gene expression and enable unidirectional temporal control by prolonging duration of target gene *** a validated in silico framework for SJR signaling,I combined these amplifiers with SJRs and show that these SJR amplifier circuits can direct spatiotemporal patterning and improve the quality of self-organization.I then show that these circuits can improve chimeric antigen receptor(CAR)T cell tumor killing against various heterogenous antigen expression *** amplifiers are flexible tools that improve control over SJR based circuits with both basic and therapeutic applications.

In present research, a novel extractant system (D2EHPA + naphthenic acid + pyridine- ester) was used to purify cobalt anolyte and a simulated industrial production were carried out. This novel extraction system can extract Cu and/or Ni against Co from chloride medium solutions at pH range of 2.5-4.5. About 2g/l nickel and 0.2g/l copper were removed from the cobalt chloride anolyte containing about 100g/l cobalt and 200g/l chloride ions respectively, the raffinate contains nickel and copper less than 0.03g/l and 0.0003g/l respectively and can be used to electrolyze high-purity cobalt. About 5.5t cobalt anolyte was purified in the simulation industrial experiment and kilogram quantities of cobalt of 99.98% purity and about 95% recovery have been produced.

Fast compression of computer-generated holographic images based on a GPU-accelerated skip-dimension vector quantization method

A method for fast and low bit-rate compression of digital holograms based on a new vector quantization （VQ） method known as the skip-dimension VQ （SDVQ） is proposed. Briefly, a complex hologram is converted into a real off-axis hologram, and partitioned into a set of image vectors. The image vectors are passed into a graphic processing unit （GPU）, and compressed through SDVQ into a set of code indices considerably smaller in data size than the source hologram. Experimental evaluation reveals that our scheme is capable of compressing a digital hologram to a compression ratio of over 500 times, in approximately 20-22 ms.

Effect of terrain and building structures on the airflow in an airport

The aim of this study was to perform computational fluid dynamics (CFD) simulations on the airflows at the Hong Kong International Airport (HKIA). In particular, the effects of hangar buildings and terrain were studied to explore the effects of turbulence on flying aircraft, especially during landing. The CFD simulation showed that significant differences in wind speeds may occur between the north and the south runways on the western part of the HKIA under typhoon conditions with a strong north to northwesterly wind. Simulation also showed that the hanger buildings between the two runways on the western side and the nearby terrain could be causing the observed difference in the wind speeds. The results also indicated that these obstacles could cause significant wind speed variations at the western end of the south runway. This may affect the operation of landing aircraft. The CFD results for a typical typhoon case were analyzed and found to match the wind data recorded by an aircraft landing that day.

Fast and robust phase retrieval for masked coherent diffractive imaging

Conventional phase retrieval algorithms for coherent diffractive imaging(CDI)require many iterations to deliver reasonable results,even using a known mask as a strong constraint in the imaging setup,an approach known as masked *** paper proposes a fast and robust phase retrieval method for masked CDI based on the alternating direction method of multipliers(ADMM).we propose a plug-and-play ADMM to incorporate the prior knowledge of the mask,but note that commonly used denoisers are not suitable as regularizers for complex-valued latent images ***,we develop a regularizer based on the structure tensor and Harris corner *** with conventional phase retrieval methods,our technique can achieve comparable reconstruction results with less time for the masked ***,validation experiments on real in situ CDI data for both intensity and phase objects show that our approach is more than 100 times faster than the baseline method to reconstruct one complex-valued image,making it possible to be used in challenging situations,such as imaging dynamic ***,phase retrieval results for single diffraction patterns show the robustness of the proposed ADMM.
