NECAB family of neuronal calcium-binding proteins in health and disease

The N-terminal EF-hand calcium-binding proteins 1–3(NECAB1–3) constitute a family of predominantly neuronal proteins characterized by the presence of at least one EF-hand calcium-binding domain and a functionally less well characterized C-terminal antibiotic biosynthesis monooxygenase domain. All three family members were initially discovered due to their interactions with other proteins. NECAB1 associates with synaptotagmin-1, a critical neuronal protein involved in membrane trafficking and synaptic vesicle exocytosis. NECAB2 interacts with predominantly striatal G-protein-coupled receptors, while NECAB3 partners with amyloid-β A4 precursor protein-binding family A members 2 and 3, key regulators of amyloid-β production. This demonstrates the capacity of the family for interactions with various classes of proteins. NECAB proteins exhibit distinct subcellular localizations: NECAB1 is found in the nucleus and cytosol, NECAB2 resides in endosomes and the plasma membrane, and NECAB3 is present in the endoplasmic reticulum and Golgi apparatus. The antibiotic biosynthesis monooxygenase domain, an evolutionarily ancient component, is akin to atypical heme oxygenases in prokaryotes but is not wellcharacterized in vertebrates. Prokaryotic antibiotic biosynthesis monooxygenase domains typically form dimers, suggesting that calcium-mediated conformational changes in NECAB proteins may induce antibiotic biosynthesis monooxygenase domain dimerization, potentially activating some enzymatic properties. However, the substrate for this enzymatic activity remains uncertain. Alternatively, calcium-mediated conformational changes might influence protein interactions or the subcellular localization of NECAB proteins by controlling the availability of protein–protein interaction domains situated between the EF hands and the antibiotic biosynthesis monooxygenase domain. The relationship between the average penetration velocity,UˉUˉ,and the initial impact velocity, V0V0,in long-rod penetration has been studied recently. Experimental and simulation results all show the linear relationship between UˉUˉ and V0V0 over a wide range of V0V0 for different combinations of rod and target materials. However, the physical essence has not been fully *** this paper, the Uˉ?V0Uˉ?V0relationship is profoundly analyzed using hydrodynamic model and Alekseevskii-Tate model. Especially, the explicitUˉ?V0Uˉ?V0 relationships are derived fromapproximate solutions of Alekseevskii-Tate model. Besides, the decelerationin long-rod penetration is discussed. The decelerationdegree is quantified by adeceleration index,α=2μˉ/(KΦjp)≈Ypρ?1/2p(ρ?1/2p+ρ?1/2t)V?20α=2μˉ/(KΦjp)≈Ypρp?1/2(ρp?1/2+ρt?1/2)V0?2, which is mostly related to the impact velocity, rod strength and rod/target densities. Thus, the state of penetration process can be identified and designed in experiments.

The Alekseevskii–Tate model is the most successful semi-hydrodynamic model applied to long-rod penetration into semi-infinite targets. However, due to the nonlinear nature of the equations, the rod(tail) velocity, penetration velocity, rod length, and penetration depth were obtained implicitly as a function of time and solved numerically By employing a linear approximation to the logarithmic relative rod length, we obtain two sets of explicit approximate algebraic solutions based on the implicit theoretica solution deduced from primitive equations. It is very convenient in the theoretical prediction of the Alekseevskii–Tate model to apply these simple algebraic solutions. In particular, approximate solution 1 shows good agreement with the theoretical(exact) solution, and the first-order perturbation solution obtained by walters et al.(Int. j. Impac Eng. 33:837–846, 2006) can be deemed as a special form of approximate solution 1 in high-speed penetration. Meanwhile, with constant tail velocity and penetration velocity approximate solution 2 has very simple expressions, which is applicable for the qualitative analysis of long-rod penetration. Differences among these two approximate solutions and the theoretical(exact) solution and their respective scopes of application have been discussed, and the inferences with clear physical basis have been drawn. In addition, these two solutions and the first-order perturbation solution are applied to two cases with different initial impact velocity and different penetrator/target combinations to compare with the theoretical(exact) solution. Approximate solution 1 is much closer to the theoretical solution of the Alekseevskii–Tate model than the first-order perturbation solution in both cases, whilst approximate solution 2 brings us a more intuitive understanding of quasi-steady-state penetration.

The feature of reinforcing bars is introduced into dynamic cavity-expansion theory. Based on the elastic-plastic response penetration model of plain (i.e., unreinforced) concrete (Forrestal and Tzou, 1997), a dynamic spherical cavity-expansion penetration model for reinforced-concrete targets is developed with consideration of the circumferential restriction effect derived from reinforcing bars in the crushed region. The theoretical solution and simplified calculation formula for the cavity radial stress in incompressible and compressible reinforced concrete are obtained by introducing a reinforcement ratio as the volume fraction of rebars in the concrete target. A damping function is presented to describe the restriction effect of a single layer of reinforcing bars on the surrounding concrete, thus establishing a model to calculate the penetration resistance of multilayer reinforced-concrete targets. Compared with test data for the penetration depth, this model considering the circumferential restriction effect produces better results compared with the existing theory.

we review our recent efforts on developing HgCdSe infrared materials on Ga Sb substrates via molecular beam epitaxy(MBE) for fabricating next generation infrared detectors with features of lower production cost and larger focal plane array format size. In order to achieve high-quality HgCdSe epilayers, ZnTe buffer layers are grown before growing HgCdSe, and the study of misfit strain in ZnTe buffer layers shows that the thickness of ZnTe buffer layer needs to be below 300 nm in order to minimize the generation of misfit dislocations. The cut-off wavelength/alloy composition of HgCdSe materials can be varied in a wide range by varying the ratio of Se/Cd beam equivalent pressure during the HgCdSe *** temperature presents significant impact on the material quality of HgCdSe, and lower growth temperature leads to higher material quality for HgCdSe. Typically, long-wave infrared HgCdSe(x = 0.18, cut-off wavelength of 10.4 μm at 80 K) presents an electron mobility as high as 1.3×10~5cm^2·V^(-1)·s^(-1), a background electron concentration as low as 1.6×10^(16)cm^(-3), and a minority carrier lifetime as long as 2.2 μs. These values of electron mobility and minority carrier lifetime represent a significant improvement on previous studies of MBE-grown HgCdSe reported in the open literatures,and are comparable to those of counterpart HgCdTe materials grown on lattice-matched CdZnTe substrates. These results indicate that HgCdSe grown at the University of western Australia, especially long-wave infrared can meet the basic material quality requirements for making high performance infrared detectors although further effort is required to control the background electron concentration to below 10^(15)cm^(-3). More importantly, even higher quality HgCdSe materials on GaSb are expected by further optimizing the growth conditions, using higher purity Se source material, and implementing postgrowth thermal annealing and defect/impurity gettering/filtering.
