Protein and non-protein sulfhydryls and disulfides in gastric mucosa and liver after gastrotoxic chemicals and sucralfate: Possible new targets of pharmacologic agents

AIM： To investigate the role of major non-protein and protein sulfhydryls and disulfides in chemically induced gastric hemorrhagic mucosal lesions （HML） and the mechanism of gastroprotective effect of sucralfate.METHODS： Rats were given 1 mL of 75% ethanol, 25%NaCl, 0.6 mol/L HCI, 0.2 mol/L NaOH or 1% ammonia solutions intragastrically （i.g.） and sacrificed 1, 3, 6 or 12 min later. Total （reduced and oxidized） glutathione （GSH ＋ GSSG）, glutathione disulfide （GSSG）, protein free sulfhydryls （PSH）, protein-glutathione mixed disulfides （PSSG） and protein cystine disulfides （PSSP） were measured in gastric mucosa and liver.RESULTS： Reduced glutathione （GSH） was depleted in the gastric mucosa after ethanol, HCI or NaCl exposure,while oxidized glutathione （GSSG） concentrations increased, except by HCI and NaOH exposure. Decreased levels of PSH after exposure to ethanol were observed,NaCl or NaOH while the total protein disulfides were increased. Ratios of reduced to oxidized glutathione or sulfhydrils to disulfides were decreased by all *** changes in thiol homeostasis were detected in the liver after i.g. abbreviation should be spelled out the first time here administration of ethanol. Sucralfate increased the concentrations of GSH and PSH and prevented the ethanol-induced changes in gastric mucosal thiol concentrations.CONCLUSION： Our modified methods are now suitable for direct measurements of major protein and nonprotein thiols/disulfides in the gastric mucosa or liver.A common element in the pathogenesis of chemically induced HML and in the mechanism of gastroprotective drugs seems to be the decreased ratios of reduced and oxidized glutathione as well as protein sulfhydryls and disulfides.

NUMERICAL SIMULATIONS FOR A VARIABLE ORDER FRACTIONAL CABLE EQUATION

In this article, Crank-Nicolson method is used to study the variable order fractional cable equation. The variable order fractional derivatives are described in the Riemann- Liouville and the Griinwald-Letnikov sense. The stability analysis of the proposed technique is discussed. Numerical results are provided and compared with exact solutions to show the accuracy of the proposed technique.

Pointwise Convergence of Cone-like Restricted Two-dimensional Fejér Means of Walsh-Fourier Series

For the two-dimensional Walsh system, Gat and Weisz proved the a.e. convergence of Fejer means σnf of integrable functions, where the set of indices is inside a positive cone around the identical function, that is, β^-1≤n1/n2 ≤β is provided with some fixed parameter ~ 〉 1. In this paper we generalize the result of Gat and Weisz. We not only generalize this theorem, but give a necessary and sufficient condition for cone-like sets in order to preserve this convergence property.

Alcoholic liver disease(ALD)and nonalcoholic fatty liver disease(NAFLD)are the two major types of chronic liver disease *** processes play key roles in the pathogeneses of fatty liver diseases,and continuous inflammation promotes the progression of alcoholic steatohepatitis(ASH)and nonalcoholic steatohepatitis(NASH).Although both ALD and NAFLD are closely related to inflammation,their respective developmental mechanisms differ to some ***,we review the roles of multiple immunological mechanisms and therapeutic targets related to the inflammation associated with fatty liver diseases and the differences in the progression of ASH and *** cell types in the liver,including macrophages,neutrophils,other immune cell types and hepatocytes,are involved in fatty liver disease *** addition,microRNAs(miRNAs),extracellular vesicles(EVs),and complement also contribute to the inflammatory process,as does intertissue crosstalk between the liver and the intestine,adipose tissue,and the nervous *** point out that inflammation also plays important roles in promoting liver repair and controlling bacterial *** the complex regulatory process of disrupted homeostasis during the development of fatty liver diseases may lead to the development of improved targeted therapeutic intervention strategies.

Direct and Simultaneous Determination of Several Impurity Elements in Manganese Tetroxide using ICP-AES

A rapid method for the determination of impurity elements, such as Zn, Ni, Co, Cr, Cu, Cd and Pb in manganese tetroxide was developed, using inductively coupled plasma atomic emission spectrometry （ICP-AES）. The critical instrumental parameters such as sample flow rate and radio frequency incident were thoroughly optimized. The effect of matrix was also examined. The sensitivity was investigated using calibration curves obtained in presence of the matrix. The obtained recoveries for Ni, Co, Cr, Cu, and Cd at the μg· g^ -1 level were satisfactory and practically independent of the matrix used for the calibration standards. The recoveries of Pb and Zn were less suffwient. The method can be applied for routine analysis and quality control purposes at μg· g^-1 level of concentration.

Anti-inflammatory pathways and alcoholic liver disease: Role of an adiponectin/interleukin-10/heme oxygenase-1 pathway

The development of alcoholic liver disease (ALD) is a complex process involving both the parenchymal and non-parenchymal cells in the liver. Enhanced inflammation in the liver during ethanol exposure is an important contributor to injury. Kupffer cells, the resident macrophages in liver, are particularly critical to the onset of ethanol-induced liver injury. Chronic ethanol exposure sensitizes Kupffer cells to activation by lipopolysaccharide via Toll-like receptor 4. This sensitization enhances production of inflammatory mediators, such as tumor necrosis factor-α and reactive oxygen species, that contribute to hepatocyte dysfunction, necrosis, apoptosis, and fibrosis. Impaired resolution of the inflammatory process probably also contributes to ALD. The resolution of inflammation is an active, highly coordinated response that can potentially be manipulated via therapeutic interventions to treat chronic inflammatory diseases. Recent studies have identif ied an adiponectin/interleukin-10/heme oxygenase-1 (HO-1) pathway that is profoundly effective in dampening the enhanced activation of innate immune responses in primary cultures of Kupffer cells, as well as in an in vivo mouse model of chronic ethanol feeding. Importantly, induction of HO-1 also reduces ethanol-induced hepatocellular apoptosis in this in vivo model. Based on these data, we hypothesize that the development of therapeutic agents to regulate HO-1 and its downstream targets could be useful in enhancing the resolution of inflammation during ALD and preventing progression of early stages of liver injury.

Effects of a structurally related substance on the crystallization of paracetamol

Paracetamol （PCM） was crystallized from an isopropanol （IPA） solution containing various small amounts of metacetamol as an additive. The effect on the nucleation kinetics was studied by measuring the induction time to nucleation and the metastable zone width using focused beam reflectance measurements （FBRM） and attenuated total reflectance （ATR-UV/Vis） spectroscopy. Both the induction time and the metastable zone width were expressed as functions of the additive concentration. Small amounts ofmetacetamol （1-4 tool-%） were found to cause significant inhibition to the nucleation by extending both the induction time and the metastable zone width. A progressive change in the morphology of the paracetamol crystals from tabular to columnar habit was observed with increasing metacetamol concentration. The solvent also had a significant effect on the size of the paracetamol crystals as smaller crystals were obtained in IPA than in aqueous solution. The dissolution rate of paracetamol was improved by the incorporation of metacetamol with 4 tool-% having the most effect. A supersaturation control （SSC） approach was implemented for the PCM-IPA system with and without metacetamol in an attempt to control and obtain larger metacetamol-doped paraeetamol crystals.

Kinetic parameter estimation for cooling crystallization process based on cell average technique and automatic differentiation

In this paper,a cell average technique(CAT)based parameter estimation method is proposed for cooling crystallization involved with particle growth,aggregation and breakage,by establishing a more efficient and accurate solution in terms of the automatic differentiation(AD)*** overcome the deficiency of CAT that demands high computation cost for implementation,a set of ordinary differential equations(ODEs)entailed from CAT based discretized population balance equation(PBE)are solved by using the AD based high-order Taylor ***,an AD based trust-region reflective(TRR)algorithm and another interior-point(IP)algorithm are established for estimating the kinetic parameters associated with particle growth,aggregation and *** a result,the estimation accuracy can be further improved while the computation cost can be significantly reduced,compared to the existing *** examples from the literature are used to illustrate the accuracy and efficiency of the AD-based CAT,TRR and IP algorithms in comparison with the existing ***,seeded batch cooling crystallization experiments ofβform L-glutamic acid are performed to validate the proposed method.

Conditional Probability Approach for Fault Detection in Photovoltaic Energy Farms

Detection of electric faults in photovoltaic (PV) farms enhances a sustainable service continuity of farm energy generation. In this paper, a probabilisticfunction is introduced to detect the faults in the PV farms. The conditional probability functions are adopted to detect different fault conditions such as internalstring faults, string-to-string faults, and string-to-negative terminal faults. As thediodes are important to make the PV farms in-service safely during the faults,the distribution currents of these faults are evaluated with different concepts ofdiode consideration as well as without considering any diode installation. Thispart of the study enhances the diode utilization in the PV farms concerning theprotection point of view. The PV string currents are used as inputs to the conditional probability detection algorithms. However, the setting of the fault detectiontechnique is not portable for the other PV systems due to broad ranges of PV system ratings. To accordingly generalize the proposed fault detection algorithm, thePV string currents are first normalized to the total array current for universallyapplying the detection function at different PV string ratings. The limiting faultresistances are evaluated to show the sensitivity of the proposed fault *** results ensure the application of the proposed probabilistic detection functionfor PV farms.
