Nonstationarities and At-site Probabilistic Forecasts of Seasonal Precipitation in the East River Basin, China

Seasonal precipitation changes under the influence of large-scale climate oscillations in the East River basin were studied using daily precipitation data at 29 rain stations during 1959–2010. Seasonal and global models were developed and evaluated for probabilistic precipitation forecasting. Generalized additive model for location,scale, and shape was used for at-site precipitation forecasting. The results indicate that:(1) winter and spring precipitation processes at most stations are nonstationary,while summer and autumn precipitation processes at few of the stations are stationary. In this sense, nonstationary precipitation processes are dominant across the studyregion;(2) the magnitude of precipitation is influenced mainly by the Arctic Oscillation, the North Pacific Oscillation, and the Pacific Decadal Oscillation(PDO). The El Nin? o/Southern Oscillation(ENSO) also has a considerable effect on the variability of precipitation regimes across the East River basin;(3) taking the seasonal precipitation changes of the entire study period as a whole, the climate oscillations influence precipitation magnitude, and this is particularly clear for the PDO and the ENSO. The latter also impacts the dispersion of precipitation changes; and(4) the seasonal model is appropriate for modeling spring precipitation, but the global model performs better for summer, autumn, and winter precipitation.

A small focused library of eighteen new 1,2,3-triazole tethered acetophenones has been efficiently prepared via click chemistry approach and evaluated for their antifungal and antioxidant *** antifungal activity was evaluated against five human pathogenic fungal strains：Candida albicans,Fusarium oxysporum,Aspergillus flavus,Aspergillus niger,and Cryptococcus *** the synthesized compounds,9c,9i,and 9p found to be more potent antifungal agents that the reference *** 1,2,3-triazole based derivatives were also evaluated for antioxidant activity,and compound 9h was found to be the most potent antioxidant as compared to the standard ***,molecular docking study of the newly synthesized compounds was performed and results showed good binding mode in the active site of fungal *** enzyme P450 cytochrome ***,the synthesized compounds were also analyzed for ADmE properties and showed potential as good oral drug candidates.

AIm To analyze how various implants placement variables affect sacroiliac(SI) joint range of motion. mETHODS An experimentally validated finite element model of the lumbar spine and pelvis was used to simulate a fusion of the SI joint using various placement configurations of triangular implants(iF use Implant System~?). Placement configurations were varied by changing implant orientation, superior implant length, and number of implants. The range of motion of the SI joint was calculated using a constant moment of 10 N-m with a follower load of 400 N. The changes in motion were compared between the treatment groups to assess how the different variables affected the overall motion of the SI joint. RESULTS Transarticular placement of 3 implants with superior implants that end in the middle of the sacrum resulted in the greatest reduction in range of motion(flexion/extension = 73%, lateral bending = 42%, axial rotation = 72%). The range of motions of the SI joints were reduced with use of transarticular orientation(9%-18%) when compared with an inline orientation. The use of a superior implant that ended mid-sacrum resulted in median reductions of(8%-14%) when compared with a superior implant that ended in the middle of the ala. Reducing the number of implants, resulted in increased SI joint range of motions for the 1 and 2 implant models of 29%-133% and 2%-39%, respectively,when compared with the 3 implant *** Using a validated finite element model we demonstrated that placement of 3 implants across the SI joint using a transarticular orientation with superior implant reaching the sacral midline resulted in the most stable construct. Additional clinical studies may be required to confirm these results.

Docosanol is the only US Food and Drug Administration(FDA)approved over-the-counter topical product for treating recurrent oral-facial herpes simplex *** analytical methods for docosanol are required to demonstrate the bioequivalence of docosanol topical products.A gas chromatography/selected ion monitoring mode mass spectrometry(GC/SIm-mS)method was developed and validated for docosanol determination in biological *** and isopropyl palmitate(internal standard)were separated on a high-polarity GC capillary column with(88%cyanopropy)aryl-polysiloxane employed as the stationary *** ions of m/z 83 and 256 were selected to monitor docosanol and isopropyl palmitate,respectively;the total run time was 20 *** GC/SIm-mS method was validated in accordance with US FDA guidelines,and the results met the US FDA acceptance *** docosanol calibration standards were linear in the 100-10000 ng/mL concentration range(R^(2)>0.994).The recoveries for docosanol from the receptor fluid and skin homogenates were>93.2%and>95.8%,*** validated method was successfully applied to analyze ex vivo human cadaver skin permeation *** applying Abreva®cream tube and Abreva®cream pump,the amount of docosanol that penetrated human cadaver skin at 48 h was 21.5±7.01 and 24.0±6.95 ng/mg,***,we concluded that the validated GC/SIm-mS was sensitive,specific,and suitable for quantifying docosanol as a quality control *** method can be used for routine analysis as a costeffective alternative to other techniques.

Objective: To investigate new scolicidal agent from natural resources to cope with the side effects associated with synthetic drugs in Echinococcosis. methods: The scolicidal potential of methanolic fruit powder extract (10 and 20 mg/mL) of mallotus philippinensis ( m. philippinensis ) was investigated. Viability of protoscoleces was confirmed by trypan blue exclusion method, where mortality was observed at concentration of 10 and 20 mg/mL in 60 min treatment against Echinococcus granulosus ( E. granulosus ), under in-vitro conditions with reference to the known standard drug Praziquantel . Results: At concentration 10 and 20 mg/mL, the mortality rate was observed 97% and 99% respectively for 60 min treatment; while up to 93% mortality was observed with 20 mg/mL for only 10 min treatment. The concentration above 20 mg/mL for above 2 h showed 100% mortality, irrespective of further incubation. Conclusions: As compared with the standard anti-parasitic drug Praziquantel our extract has significant scolicidal activity with almost no associated side effects.

Determination of cilostazol and its active metabolite 3,4-dehydro cilostazol from small plasma volume by UPLC-mS/mS

A simple,rapid and sensitive ultra performance liquid chromatography-tandem mass spectrometry（UPLC-mS/mS） method has been developed for the simultaneous determination of cilostazol and its pharmacologically active metabolite 3,4-dehydro cilostazol in human plasma using deuterated analogs as internal standards（ISs）.Plasma samples were prepared using solid phase extraction and chromatographic separation was performed on UPLC BEH C18（50 mm × 2.1 mm.1.7 μm） *** method was established over a concentration range of 0.5-1000 ng/mL for cilostazol and 0.5-500 ng/mL for 3.4-dehydro ***- and inter-batch precision（%CV） and accuracy for the analytes were found within 0.93-1.88 and 98.8-101.7% for cilostazol and 0.91-2.79 and 98.0-102.7% for the metabolite *** assay recovery was within 95-97% for both the analytes and internal *** method was successfully applied to support a bioequivalence study of 100 mg cilostazol in30 healthy subjects.

创面处理的原则,包括清创、创面准备和使用涉及改变创面生理以促进愈合的新技术,在试图治愈慢性糖尿病相关的足溃疡时是至关重要的。此外,糖尿病相关的足溃疡的发病率和治疗成本不断上升,这就需要结合作为金标准的多学科治疗已有共识,运用高质量的疗效和成本效益证据来支持和加强慢性糖尿病相关的足溃疡创面愈合的干预措施。本指南是2023年国际糖尿病足工作组(IWGDF)针对促进糖尿病患者足溃疡愈合的干预措施的循证指南。它是对2019年IWGDF指南的更新。我们遵循推荐、评估、发展、评价分级(GRADE)方法,以患者-干预-对照-结果(PICO)形式设计临床问题从而得出重要结果,进行系统综述,制定评判表汇总,并为每个问题撰写推荐和理由。每条推荐的形式都是基于系统综述中发现的证据,并使用GRADE评判汇总项目,包括理想和不理想的效果、证据的可信度、对患者的价值、所需的资源、成本效益、公平性、可行性和可接受性;我们制定的推荐由作者同意并由独立专家和利益相关者审查。根据系统综述和从证据到决策过程的结果,我们提出29条推荐。我们对使用干预措施改善糖尿病患者足溃疡的愈合提出了一些有条件的支持性推荐。这些推荐包括蔗糖八硫酸盐敷料的使用,对手术后的创面使用负压疗法,使用胎盘衍生产品,使用自体白细胞、血小板、纤维蛋白贴剂,使用局部氧疗,以及使用高压氧,但我们强调在任何情况下这些推荐应在单独应用最佳标准治疗无法治愈伤口且有资源可用于干预的情况下使用。这些关于创面愈合的推荐应该有助于提高糖尿病伴足溃疡患者的疗效,我们希望这些推荐能得到广泛的实施。然而,尽管作为推荐基础的许多证据的准确性正在提高,但总体上仍然很差,我们鼓励在这一领域进行更多、质量更好的试验,包括那些有关卫生经济学分析的试验。

A series of novel flavonols were synthesized by cyclizing corresponding hydoxy chalcones using Algar flynn Oyamada reaction (1)The synthesized compounds were characterized by various spectrochemical methods including IR, mASS, NmR spectroscopy. Out of 13 compounds screened for antibacterial activity [2], compound VmF 12 (a bromo derivative) showed activity with an IC50value at 0.16μm against S. aureus.

Development and Identification of Novel Rice Blast Resistant Sources and Their Characterization Using molecular markers

To develop and characterize introgression lines for leaf and neck blast resistance, 326 introgression lines were developed using various accessions of six different AA genome wild species in the genetic background of elite Indian varieties like PRl14 and Pusa 44 and were screened for blast resistance. Stringent phenotyping coupled with genotyping using gene based markers led to the identification of four resistant introgression lines, which showed promising resistance and do not possess any of the tested genes. Furthermore, multi-location screening confirmed the field resistance of the four introgression lines to both leaf and neck blast. molecular characterization of these introgression lines using genome-wide simple sequence repeat markers revealed the presence of small percentage of wild Oryza genome introgrssion. So these lines can be used for mapping and identification of novel leaf and neck blast resistance genes. Thus, these four introgression lines can be considered as new genetic resources for blast resistance.
