The role of systemic therapy in borderline resectable and locally advanced pancreatic ductal adenocarcinoma

Pancreatic ductal adenocarcinoma(PDAC)remains a deadly disease,even in patients whose cancer is localized and *** resection provides the only option for cure,but long-term survival rates remain *** patients with borderline resectable(BR)disease who undergo upfront resection,many patients are either too unwell for subsequent adjuvant systemic therapy,develop recurrence soon after,or are found to have unresectable disease *** is increasing evidence for a neoadjuvant approach,using more conventional multi-agent chemotherapy regimens,which have demonstrated higher activity in the metastatic setting compared to single *** patients with locally advanced(LA)disease,which is unresectable by current definitions,there is mounting evidence that effective neoadjuvant systemic therapy is able to convert some patients’disease to a resectable state,offering the potential for long-term survival and *** we present a review of key trials focusing on prospective,randomized studies to provide high-level evidence supporting a neoadjuvant approach to both BR and LA ***,many knowledge gaps exist,such as the optimal neoadjuvant multi-agent chemotherapy regimen,the role of radiotherapy,and the safety and efficacy of adding immunotherapy to chemo/radiation *** challenges in determining the optimal approach to patients with BR or LA PDAC include not only overcoming the inherent difficulties in conducting complex,multidisciplinary,multicentre randomized trials in patients with a high-morbidity and mortality disease,but also trying to standardize disease definitions,treatment regimens,and outcome measures.

Proteome changes of porcine follicular fluid during follicle development

Background:Ovarian follicular fluid influences follicle and oocyte growth,but the fluctuation of its protein content during folliculogenesis has not been comprehensively *** we used a shotgun approach and bioinformatics analyses to investigate and compare the proteomes of porcine follicular fluid(pFF)obtained from small(mm),medium(4–6 mm)and large(>6–12 mm)***:Follicular fluid samples containing highest estrogen levels were selected as non-atretic from small(SNA:26.1±15 ng/mL),medium(mNA:162±54 ng/mL),and large(LNA:290±37 ng/mL)follicles for proteomic *** detected 1627,1699,and 1756 proteins in SNA,mNA,and LNA samples,*** 60–63%of total proteins were specific to each sample,11–13%were shared in pairwise comparisons,and 247 proteins were shared among all *** categorization indicated comparable gene ontology(GO)terms distribution per cellular component,molecular function,and biological process categories across samples;however,the ranking of highly significantly enriched GO terms per category revealed differences between *** patterns of proteinto-protein interactions varied throughout follicle development,and proteins such as serine protease inhibitor,clade E(SERPINE);plasminogen activator,urokinase(PLAU);and plasminogen activator,urokinase receptor(PLAUR)appeared stage-specific to SNA,mNA,and LNA,***“complement and coagulation cascades”was the common major ***,properdin and fibulin-1 were abundant proteins that appeared absent in LNA ***:This study provides extensive and functional analyses of the pFF proteome changes during folliculogenesis and offers the potential for novel biomarker discovery in pFF for oocyte quality assessment.

Strain-Rate Dependency of a Unidirectional Filament Wound Composite under Compression

This article presents the results of experimental studies concerning the dynamic deformation and failure of a unidirectional carbon fiber reinforced plastic(T700/LY113)under *** test samples were manufactured through the filament winding of flat *** establish the strain rate dependencies of the strength and elastic modulus of the material,dynamic tests were carried out using a drop tower,the Split Hopkinson Pressure Bar method,and standard static *** samples were loaded both along and perpendicular to the direction of the reinforcing *** applicability of the obtained samples for static and dynamic tests was confirmed through finite elementmodeling and the high-speed imaging of the deformation and failure of samples during *** a result of the conducted experimental studies,static and dynamic stress-strain curves,time dependencies of deformation and the stress and strain rates of the samples during compression were *** on these results,the strain rate dependencies of the strength and elasticity modulus in the strain rate range of 0.001-6001/s are *** is shown that the strain rate significantly affects the strength and deformation characteristics of the unidirectional carbon fiber composites under *** increase in the strain rate by 5 orders of magnitude increased the strength and elastic modulus along the fiber direction by 42%and 50%,*** loading resulted in a strength and elastic modulus increase by 58%and 50%,*** average strength along the fibers at the largest studied strain rate was about *** obtained results can be used to design structural elements made of polymer composite materials operating under dynamic shock loads,as well as to build models of mechanical behavior and failure criteria of such materials,taking into account the strain rate effects.

The outage probability of a composite microscopic and macroscopic diversity system is evaluated over correlated shadowed fading *** correlations on both a microlevel and macrolevel are taken into account for the *** expression of the desired outage probability is explicitly presented,and two evaluation approaches,i.e.a compact Gaussian-Hermite quadrature method and an effective iterative algorithm,are *** accuracy and efficiency of the proposed approaches are analysed,and a guideline is provided for their *** employing the proposed evaluation approaches,results and demonstrations are presented,which display the implied effects of the corresponding parameters on the system outage performance,and reveal the potential to facilitate the design and analysis of such composite diversity systems.

Effects of cellulose nanocrystals on the acid resistance of cementitious composites

Acid mine drainage presents an important threat to cementitious *** study is aimed at investigating the effect of cellulose nanocrystals(CNCs)on the acid resistance of cementitious *** were added to mortar mixtures as additives at cement volume ratios of 0.2vol%,0.4vol%,1.0vol%,and 1.5vol%.After 28 d of standard curing,the samples were immersed in a sulfuric acid with a pH value of 2 for 75 *** unconfined compressive strength(UCS)test,the density,water absorption,void volume test,and thermogravimetric analysis were carried out to investigate the properties of CNC mixtures before sulfuric acid *** was found that the addition of CNC reduced the volume of permeable voids and increased the hydration degree and mechanical strength of the *** in mass and length were monitored during immersion to evaluate the acid resistance of *** mixture with 0.4vol%CNC showed a reduced mass change and length change indicating its improved acid resistance.

Understanding surface charge effects in *** 2:Hydrogen peroxide reactions at platinum

Electrocatalytic activity is influenced by the surface charge on the solid ***,our attention has been focused on how the surface charge shapes the electric potential and concentration of ionic reactant(s)in the local reaction *** H_(2)O_(2)redox reactions at Pt(111)as a model system,we reveal a peculiar surface charge effect using ab initio molecular dynamics simulations of electrified Pt(111)-water *** this scenario,the negative surface charge on Pt(111)repels the O-O bond of the reactant(H_(2)O_(2))farther away from the electrode *** leads to a higher activation barrier for breaking the O-O *** this microscopic mechanism into a microkinetic-double-layer model,we are able to semi-quantitatively interpret the pH-dependent activity of H_(2)O_(2)redox reactions at Pt(111),especially the anomalously suppressed activity of H_(2)O_(2)reduction with decreasing electrode *** relevance of the present surface charge effect is also examined in wider scenarios with different electrolyte cations,solution pHs,crystal facets of the catalyst,and model *** contrast with previous mechanisms focusing on how surface charge influences the local reaction condition at a fixed reaction plane,the present work gives an example in which the location of the reaction plane is adjusted by the surface charge.

mass Spectrometric Analyses Reveal a Central Role for Ubiquitylation in Remodeling the Arabidopsis Proteome during Photomorphogenesis

The switch from skotomorphogenesis to photomorphogenesis is a key developmental transition in the life of seed plants. While much of the underpinning proteome remodeling is driven by light-induced changes in gene expression, the proteolytic removal of specific proteins by the ubiquitin-26S proteasome system is also likely paramount. Through mass spectrometric analysis of ubiquitylated proteins affinity-purified from etiolated Arabidopsis seedlings before and after red-light irradiation, we identified a number of influential proteins whose ubiquitylation status is modified during this switch. We observed a substantial enrichment for proteins involved in auxin, abscisic acid, ethylene, and brassinosteroid signaling, peroxisome function, disease resistance, protein phosphorylation and light perception, including the phytochrome （Phy） A and phototropin photoreceptors. Soon after red-light treatment, PhyA becomes the dominant ubiquitylated species, with ubiquitin attachment sites mapped to six lysines. A PhyA mutant protected from ubiquitin addition at these sites is substantially more stable in planta upon photoconversion to Pfr and is hyperactive in driving photomorphogenesis. However, light still stimulates ubiquitylation and degradation of this mutant, implying that other attachment sites and/or proteolytic pathways exist. Collectively, we expand the catalog of ubiquitylation targets in Arabidopsis and show that this post- translational modification is central to the rewiring of plants for photoautotrophic growth.

AIm: To study ocular axial lengths in pediatric subjects without intraocular pathology. mETHODS: An Institutional Review Board-approved consecutive retrospective chart review of axial lengths measured in pediatric subjects who underwent examination under anesthesia due to positive family history of retinoblastoma or other inherited ocular disease. Only subjects without any intraocular pathology in either eye were included. Subjects were stratified into age groups. An axial length model using a logarithmic regression algorithm was ***: Data from 330 eyes of 165 subjects were included in the study. The mean age at the time of examination was 30.62(SD 18.04)mo. The steepest increase in axial length was present during the first 10 mo of life. After 36 mo, there was no statistically significant axial length growth. CONCLUSION: This study presents the biggest series of pediatric axial lengths in healthy eyes. The axial length model developed with these data may assist in the diagnosis and management of a wide variety of pediatric ophthalmic diseases.

A wide range of events observed at the giant pulses （high energy density, observed localization of giant pulses GPs relative to the average pulse, fine structure of GPs with duration up to some nanoseconds, observed circular polarization of GPs, correlation between the GP phase and the phase of the hard pulsar emission X-ray and gamma） can be explained from the viewpoint that the internal polar gap is a cavity-resonator stimulated by discharges and radiating through the breaks in the magnetosphere. The new results in this field [the electromagnetic （era） waves generation in the gap in the process of longitudinal acceleration in the electric field vanishing on the star surface, high frequency break in the spectrum as a result of switching off this generation, formation in this process a power-low spectrum with a high frequency （hf） break, the possibility determination of pulsar magnetic field by the hf break position, the difference between main pulse and inter pulse mechanism generation, quantization of em tornado rotation in the gap and appearance of the bands in the inter pulse spectrum, influence the high energy density in the gap on pair generation and position of the dead line in pulsars] are added in the intermediate epilogue.
