The aim of the present paper was to check for the presence of cerebrovascular dystroglycan in vertebrates,because dystroglycan,which is localized in the vascular astroglial end-feet,has a pivotal function in glio-vascular *** mammalian brains,the immunoreactivity ofβ-dystroglycan subunit delineates the *** results of the present study demonstrate similar patterns in other vertebrates,except for anurans and the teleost groups Ostariophysi and *** this study,we investigated 1 or 2 representative species of the main groups of Chondrichthyes,teleost and non-teleost ray-finned fishes,urodeles,anurans,and *** also investigated 5 mammalian and 3 bird *** were obtained from breeders or *** presence ofβ-dystroglycan was investigated immunohistochemically in free-floating ***-embedding electron microscopical immunohistochemistry on Heterodontus japonicus shark brains demonstrated that in Elasmobranchii,β-dystroglycan is also localized in the perivascular glial end-feet despite the different construction of their blood-brain *** results indicated that the cerebrovascularβ-dystroglycan immunoreactivity disappeared separately in anurans,and in teleosts,in the latter group before its division to Ostariophysi and ***munohistochemistry in muscles and western blots from brain homogenates,however,detected the presence ofβ-dystroglycan,even in anurans and all teleosts.A possible explanation is that in the glial end-feet,β-dystroglycan is masked in these animals,or disappeared during adaptation to the freshwater habitat.

Assessing Quality of Reclaimed Urban Wastewater from Algarrobo Municipality to Be Used for Irrigation

In the European Mediterranean area, the lack of available water resources had led to consider the reclaimed urban waters as an integral part of water resources. Reclaimed urban waters could mitigate water shortage, support agriculture sector and protect rivers and groundwater resources. This work is focused on the evaluation of reclaimed urban water (RW) from Algarrobo municipality, which is located in the Málaga province in the Mediterranean coast at South-East of Spain. Wastewater in the municipal waste water treatment plant, was treated by a membrane bioreactor (MBR) as a tertiary treatment. The main goal of this work was to determine the composition of RW to be compared with fresh local water (LW) from the Algarrobo River, in order to evaluate its suitability for irrigation. Electrical conductivity (EC), pH, sodium absorption ratio (SAR), and mineral content were weekly measured from September 2017 until June 2018. Assessment of RW quality was carried out considering the limit values for each parameter as stated by the current Spanish legislation RD 1620/2007 for reuse of reclaimed water and European Directive for water reuse (EU 91/271/EEC, Annex I). The MBR facility was also focused on preserving essential minerals for plant nutrition to use them for crop fertilization instead of commercial fertilizers. The average content of nitrogen, phosphorous and potassium were, respectively, 72%, 65% and 46% of the amount needed for the Hoagland 1/4 strength solution that was used in this study for irrigation of tomato, avocado and mango crops. From this study it appears that it is possible using membrane technology, to treat urban wastewater in order to supply a good quality of water for irrigation. Water analysis has also proved MBR to be efficient for removal of microbiological pollutants, inorganic compounds, some trace elements and heavy metals. Exposure assessment is a key component of any risk-benefit assessment, yet it is clear that there is a lack of reliable methodology in this area for assessing consumer exposures to both food constituents and nonfood products. The International Life Sciences Institute (ILSI) Europe Food Intake Methodology Task Force has in the past explored methods to assess the intake of nutrients/additives and exposure to contaminants/residues from food. In December 2008, a workshop was held to discuss the differences between different types of exposure assessments as well as the difficulties involved in the practical application of the methods available. It was noted that although no two assessments are the same in terms of data required and its availability, or the assumptions made, there is also wide and perhaps unnecessary variation between the approaches taken by different assessors. As a result, the ILSI Europe Food Intake Methodology Task Force initiated an activity aimed at producing a practical guide for conducting intake/exposure assessments in the form of an interactive web-based application. During the course of this work it became clear that the best form in which to present the guide would be a web-based MediaWiki-type system. A website was developed and launched in October 2012, at which time global experts and practitioners in dietary exposure assessment were invited to register to use the site and help keep the knowledge contained within it, relevant and up-to-date. It is hoped that the guide will be an important reference source for a wide group of stakeholders, providing concise guidance on the planning, conduct, reporting and interpretation of exposure assessments and contributing to greater harmonisation of the dietary intake/exposure methodologies used.

Background: The endocannabinoid (EC) system is well characterized in the central nervous system but scarcely studied in peripheral organs. In this paper, we newly identify the effect of the EC anandamide (AEA) upon renal proximal tubule cells. Methods: Measurement of lactate dehydrogenase (LDH) release after treatment of primary renal proximal tubule cells (RPTEC) and renal carcinoma cell line (Caki-1) with AEA, arachidonic acid (AA), ethanolamide (EtAm), EC receptor CB1 antagonist (AM251), CB2 receptor antagonist (SR144528), TRPV1 receptor antagonist (capsazepine), degradation enzyme fatty acid amide hydrolase (FAAH) antagonist (URB597), antioxidants GSH-EE;Trolox, GSH depletor BSO, membrane cholesterol depletor (MCD), apoptosis inhibitor zVAD, necroptosis inhibitor Nec-1 or ferroptosis inhibitor Fer-1. Western blot and qRT-PCR analysis plus determination of reactive oxygen species (ROS) via H2-DCFDA were performed. Histology for EC enzymes, N-acetylphosphatidylethanolamine-hydrolyzing phospholipase D (NAPE-PLD) and FAAH, as well as the determination of physiological levels of ECs in human and rat renal tissue via liquid chromatography were conducted. Results: AEA both dose- and time-dependently induces cell death in RPTEC and Caki-1 within hours, characterized by cell blebbing, not influenced by blocking the described EC receptors by AM251, SR144528, capsazepine or FAAH by URB597 or MCD. Cell death is mediated via ROS. There is no difference found in the histology of the enzymes FAAH and NAPE-PLD in human renal tissue with interstitial nephritis. Blocking of apoptotic, necroptotic or ferroptotic cell death does not lead to a reduction in LDH release in vitro. Conclusion: The endocannabinoid anandamide induces cell death in renal proximal tubule cell in a time- and dose-dependent manner. This pathway is mediated via ROS and is independent of cannabinoid receptors, membrane cholesterol or FAAH activity.
