Mechanisms involved in selecting and maintaining neuroblastoma cancer stem cell populations, and perspectives for therapeutic targeting

Pediatric neuroblastomas(NBs)are heterogeneous,aggressive,therapy-resistant embryonal tumours that originate from cells of neural crest(NC)origin and in particular neuroblasts committed to the sympathoadrenal progenitor cell *** resistance,post-therapeutic relapse and subsequent metastatic NB progression are driven primarily by cancer stem cell(CSC)-like subpopulations,which through their self-renewing capacity,intermittent and slow cell cycles,drug-resistant and reversibly adaptive plastic phenotypes,represent the most important obstacle to improving therapeutic outcomes in unfavourable *** this review,dedicated to NB CSCs and the prospects for their therapeutic eradication,we initiate with brief descriptions of the unique transient vertebrate embryonic NC structure and salient molecular protagonists involved NC induction,specification,epithelial to mesenchymal transition and migratory behaviour,in order to familiarise the reader with the embryonic cellular and molecular origins and background to *** follow this by introducing NB and the potential NC-derived stem/progenitor cell origins of NBs,before providing a comprehensive review of the salient molecules,signalling pathways,mechanisms,tumour microenvironmental and therapeutic conditions involved in promoting,selecting and maintaining NB CSC subpopulations,and that underpin their therapy-resistant,self-renewing metastatic ***,we review potential therapeutic strategies and future prospects for targeting and eradication of these bastions of NB therapeutic resistance,post-therapeutic relapse and metastatic progression.

Mobile ad-hoc communication is a demonstrated solution to mitigate the impact of infrastructure failures during large-scale disasters. A very complex issue in this domain is the design validation of software applications that support decision-making and communication during natural disasters. Such disasters are irreproducible, highly unpredictable, and impossible to scale down, and thus extensive assessments cannot be led in situ. In this context, simulation constitutes the best approach towards the testing of software solutions for natural disaster responses. The present survey reviews mobility models, ad-hoc network architectures, routing protocols and network simulators. Our aim is to provide guidelines for software developers with regards to the performance evaluation of their applications by means of simulation.

Methodological choices of HR research conducted in Asia

A twenty-year study of the Human Resource （HR） practices- outcome relationship has found that more rigorous methodologies have been adopted over time. However, several problematic features such as cross-sectionaL single-informant, and single-level designs continue to be adopted （Bainbridge et al., Human Resource Management, 2016）. Responding to calls for increased contextualization of research by investigating the relationship between the location of data collection and the methodological choices of researchers, this study answers the question ＂How unique are the methodological choices of HR research conducted in Asia？＂ Applying content analysis to 241 published articles, we compare internal, external, construct and statistical conclusion validity of studies collected in North America （n^66）, Europe （n=95） and Asia （n=80, including 57 studies from China）. Results show that despite similarities in cross-sectional, single-informant and single-level designs across regions, research conducted in Asia is mainly undertaken via field studies, using subjective outcome measures at the organizational level, following a post-predictive design. In addition, studies from Asia are more recent, and show a shorter time gap between data collection and publication. Theoretical and practical implications embedded in the dynamic context of Asia in general, and China more specifically are discussed.

Comparative study of energetic potential from anaerobic digestion and direct combustion of date palm wastes

This work aims to evaluate the technical and environmental feasibility of converting date palm waste from a Moroccan oasis into *** this study,two different scenarios were *** first proposed scenario is the valorization of the date palm substrates using direct combustion(thermochemical conversion)to produce *** second proposed scenario is the valorization of the substrates using combined anaerobic digestion of the substrate and direct combustion of the *** this study,two different date palm wastes are:empty fruit bunch(EFB)and palm leaflet(PL).The experimental lower heating values of the digestates studied are 12.93 and 12.71 MJ/kg,*** results show that using Scenario 1,the estimated output energies for the EFB and PL waste were 1.13 and 1.17 MWh,*** Scenario 2,the EFB and PL output energies were 1.27 and 1.39 MWh,*** study revealed that the combined process is possible as an alternative way to produce ***,combining anaerobic digestion and direct combustion of lignocellulosic waste can contribute to reducing greenhouse gas emissions.

Green Method, Optical and Structural Characterization of ZnO Nanoparticles Synthesized Using Leaves Extract of M. oleifera

ZnO nanoparticles(ZnO-NP)present innovative optical,electrical,and magnetic properties that depend on spe-cific characteristics,e.g.,size,distribution,and ***,these properties are essential to address various applications in areas such as electronics,medicine,energy,and *** addition,the performance of this ZnO-NP depends of their preparation which can be done by chemical,physical,and biological ***,nowadays,the main interest in developing ZnO-NP synthesis through biological methods bases on the decrease of use of toxic chemicals or energy applied to the procedures,making the process more cost-effective and environ-mentally ***,the large-scale production of nanoparticles by green synthesis remains a big challenge due to the complexity of the biological extracts used in chemical *** being the case,the preparation of ZnO-NP using Moringa oleifera extract as an alternative biological agent for capping and reduction in synthesis was evaluated in this ***,the results based on the analysis of the optical and structural characterization of the ZnO-NP obtained by employing UV-Vis,DLS,zeta potential,XRD,ATR-FTIR,and FE-SEM indicate mostly the presence of spherical nanosized material with a mean hydrodynamic diameter of 47.2 nm measured by DLS and a mean size diameter of 25 nm observed with FE-SEM ***,in FE-SEM images a homo-geneous dispersion and distribution is observed in the absence of agglutination,agglomeration,or generation of significant lumps of the *** XRD analysis showed that heat annealing induced the crystallite size favor-ing their *** obtained data confirm the synthesis of ZnO-NP and the absence of impurities associated with organic compounds in the annealed ***,those results and low-cost production pre-sent to the synthesized ZnO-NP by this biological method as a useful material in several applications.

Hydrogen breath test for the diagnosis of lactose intolerance,is the routine sugar load the best one?

AIM: To evaluate the prevalence of lactose intolerance (LI) following a load of 12.5 g in patients diagnosed as high-grade malabsorbers using the hydrogen breath test (HBT)-***: Ninety patients showing high-grade malabsorption at HBT-25 were submitted to a second HBT with a lactose load of 12.5 *** hydrogen production,area under the curve of hydrogen excretion and occurrence of symptoms were ***: Only 16 patients (17.77%) with positive HBT-25 proved positive at *** production was lower as compared to HBT-25 (peak value 21.55 parts per million (ppm) ± 29.54 SD vs 99.43 ppm ± 40.01 SD; P < 0.001).Symptoms were present in only 13 *** absence of symptoms during the high-dose test has a high negative predictive value (0.84) for a negative low-dose *** presence of symptoms during the first test was not useful for predicting a positive low-dose test (positive predictive value 0.06-0.31).CONCLUSION: Most patients with a positive HBT-25 normally absorb a lower dose of lactose and a strict lactose restriction on the basis of a "standard" HBT is,in most instances,***,the 25 g lactose tolerance test should probably be substituted by the 12.5 g test in the diagnosis of LI,and in providing dietary guidelines to patients with suspected lactose malabsorption/intolerance.

A framework for the modeling of gut blood flow regulation and postprandial hyperaemia

After a meal the activity of the gut increases markedly as digestion takes place. Associated with this increase in activity is an increase in blood flow, which has been shown to be dependent on factors such as caloric content and constitution of the meal. Much qualitative work has been carried out regarding mechanisms for the presence of food in a section of gut producing increased blood flow to that section, but there are still many aspects of this process that are not fully understood. In this paper we briefly review current knowledge on several relevant areas relating to gut blood flow, focusing on quantitative data where available and highlighting areas where further research is needed. We then present new data on the effect of feeding on flow in the superior mesenteric artery. Finally, we describe a framework for combining this data to produce a single model describing the mechanisms involved in postprandial hyperaemia. For a section of the model, where appropriate data are available, preliminary results are presented.

目的：为了解中国和赞比亚消费者在多大程度上理解和使用营养标示栏,并评价选择食物时使用营养标示栏(NFP)的影响因素水平。方法：分别从南京和利文斯敦的两个国际都市募集498名中国消费者和325名赞比亚消费者。使用购物中心现场采访调查技术,在中国的主要食品超市如麦德龙、沃尔玛、苏果,在赞比亚的斯帕尔、绍普莱特附近对消费者进行方便的抽样,并使用结构式问卷进行调查。选择18岁及以上的参与者。性别、年龄、教育程度、职业、收入、家庭人数和健康状况是自变量,消费者阅读和理解NFP是应变量。数据统计参用卡方检验法分析研究结果。结果：记录了中国(58.7%)和赞比亚(67.1%)消费者意识并阅读NFP信息的平均水平。此外,卡方检验揭示基于性别的统计学差异。在两个地区更多女性(中国65.3%,赞比亚74.4%)消费者都比男性消费者(中国51.1%,赞比亚60.9%)在使用NFP上意识性更强。使用NFP的消费者大多是中年人(31-45岁和46-50岁)。并且中等及以上教育水平的消费者对NFP有更高的意识和理解水平。在中国和赞比亚,当消费者属于医学领域、工程师、商人、教师和家庭主妇,则会更多地使用NFP。调查结果显示两地家庭对NFP知晓度产生的影响具有显著性差异。在中国的家庭比赞比亚的家庭更有意识使用NFP。至于收入,在中国那些高收入的消费者比低收入使用NFP更多,而在赞比亚所有收入水平使用度平均分布。健康状况代表个人或任何一名家庭成员当前的健康状况,在中国,健康状况这一变量在意识和阅读NFP上有积极影响,存在健康问题的消费者(72.6%)比那些没有健康问题的消费者会更多的使用NFP。由于结果没有统计学意义,这种情况在赞比亚并非如此。结论：NFP的知晓度和使用存在的多样性主要是由于个人特征的影响。这些个人特征可能促进消费者使用NFP(如受过更高教育的个体意识并使用NFP的可能性更高),或对消费者产生负面影响(较低或未受教育的消费者不太可能意识到使用NFP)。性别、教育和职业对中国和赞比亚的消费者都具有积极影响。收入水平和家庭人数对中国消费者具有积极的影响,但赞比亚的消费者无此效应。健康状况也积极影响着中国消费者使用NFP,而赞比亚消费者却不是这样。结果表明,需要发展项目旨在提高NFP信息的意识水平,以确保发展健康的消费习惯。特别是那些不知道并混淆NFP成分表,在意食品标签的其他组分而忽略了NFP的高比例的消费者。这也将确保消费者明智选择食物,避免饮食相关的疾病。

Spectrum and visual outcomes of Vogt-Koyanagi-Harada disease in Argentina

AIM： To review all cases of Vogt-Koyanagi-Harada （VKH） disease in an Inflammatory Eye Disease Service in Argentina and to describe the clinical profile and outcomes of ***： The records from patients with VKH disease between January 1980 and December 2008 were retrospectively analyzed for clinical profile, complications, and treatment. Patients were classified according to their initial treatment in group 1： high corticosteroid dose [≥1 mg/（kg·d）] within 2wk of illness onset; group 2： high corticosteroid dose, 2 to 4wk of onset and group 3： patients who received the high dose after 1mo of illness onset, patients who received lower oral doses than 1 mg/（kg·d） without regarding the time of beginning of the ***： A total of 210 eyes of 105 patients were included. The mean age at presentation was 32.6±13y （range： 10-74y）, and 86.7% were female. The mean duration of follow up was 144±96.6mo. Patients in the group 1 had significantly higher visual acuity than the other groups （P〈0.0001）, none had （loss of, or no） light perception at the end of follow up, whereas 24.7% patients in group 3 ended in light perception （P〈0.004）.CONCLUSION： Patients with early high dose corticosteroid treatment have better visual acuity and fewer complications. Proper timing in referral and treatment is critical for better visual outcome in VKH disease.

Localization and role of metabotropic glutamate receptors subtype 5 in the gastrointestinal tract

Metabotropic glutamate receptor subtype 5 (mGluR5) is a Group I mGlu subfamily of receptors coupled to the inositol trisphosphate/diacylglycerol pathway. Like other mGluR subtypes, mGluR5s contain a phylogenetically conserved, extracellular orthosteric binding site and a more variable allosteric binding site, located on the heptahelical transmembrane domain. The mGluR5 receptor has proved to be a key pharmacological target in conditions affecting the central nervous system (CNS) but its presence outside the CNS underscores its potential role in pathologies affecting peripheral organs such as the gastrointestinal (GI) tract and accessory digestive organs such as the tongue, liver and pancreas. Following identification of mGluR5s in the mouth, various studies have subsequently demonstrated its involvement in mechanical allodynia, inflammation, pain and oral cancer. mGluR5 expression has also been identified in gastroesophageal vagal pathways. Indeed, experimental and human studies have demonstrated that mGluR5 blockade reduces transient lower sphincter relaxation and reflux episodes. In the intestine, mGluR5s have been shown to be involved in the control of intestinal inﬂammation, visceral pain and the epithelial barrier function. In the liver, mGluR5s have a permissive role in the onset of ischemic injury in rat and mice hepatocytes. Conversely, livers from mice treated with selective negative allosteric modulators and mGluR5 knockout mice are protected against ischemic injury. Similar results have been observed in experimental models of free-radical injury and in vivo mouse models of acetaminophen intoxication. Finally, mGluR5s in the pancreas are associated with insulin secretion control. The picture is, however, far from complete as the review attempts to establish in particular as regards identifying specific targets and innovative therapeutic approaches for the treatment of GI disorders.
