Artemisia vulgaris Induces Tumor-Selective Ferroptosis and Necroptosis via Lysosomal Ca^(2+) Signaling

Objective To evaluate the chemical composition and effects of Artemisia vulgaris(AV)hydroalcoholic extract(HEAV)on breast cancer cells(MCF-7 and SKBR-3),chronic myeloid leukemia(K562)and NIH/3T3 *** Phytochemical analysis of HEAV was done by high-performance liquid chromatography-mass(HPLC)*** and cell death studies were performed using trypan blue and Annexin/FITC-7AAD,***-1(Fer-1)and necrostatin-1(Nec-1)were used to assess the mode of HEAV-induced cell death and acetoxymethylester(BAPTA-AM)was used to verify the involvement of cytosolic calcium in this *** calcium measurements were made using *** HEAV decreased the viability of MCF-7,SKBR-3 and K562 cells(P<0.05).The viability of HEAV-treated K562 cells was reduced compared to HEAV-exposed fibroblasts(P<0.05).Treatment of K562 cells with HEAV induced cell death primarily by late apoptosis and necrosis in assays using annexin V-FITC/7-AAD(P<0.05).The use of Nec-1 and Fer-1 increased the viability of K562 cells treated with HEAV relative to cells exposed to HEAV alone(P<0.01).HEAV-induced Ca^(2+)release mainly from lysosomes in K562 cells(P<0.01).Furthermore,BAPTA-AM,an intracellular Ca^(2+)chelator,decreased the number of non-viable cells treated with HEAV(P<0.05).Conclusions HEAV is cytotoxic and activates several modalities of cell death,which are partially dependent on lysosomal release of Ca^(2+).These effects may be related to artemisinin and caffeoylquinic acids,the main compounds identified in HEAV.

DNA vaccines are the third generation vaccines based on purified plasmid preparations containing transgenes that encode antigenic/therapeutic proteins or peptides capable of triggering an immune response against a wide range of diseases. This vaccine platform presents several attributes that confer distinct advantages over other vaccine technologies in terms of safety, ease of fabrication and stability. Many aspects, such as antigen expression and especially vector design, are under study because of their great influence on immunogenicity and efficacy of DNA vaccines. In this regard, with the attempt of improving the efficiency of DNA vaccines, co-expression of stimulatory sequences and diverse vector delivery systems are being optimized. With this in mind, this review aims to giving a conceptual approach of DNA vaccines, explaining their mechanisms of action and listing the already licensed veterinary DNA vaccines presented in the market.

Nutrient content affecting Spodoptera frugiperda and Dalbulus maidis occurrence in corn

This work aims to study the effect of the nutrient contents in the leaves of corn on the incidence of Spodopterafrugiperda （Smith）（Lepidoptera： Noctuidae） and Dalbulus maidis （Delong ＆ Wolcott）（Homoptera： Cicadellidae）. The treatments were represented by unfertilized corn plants and corn plants fertilized with 250 kg of the formulation 4-14-8 （N- P-K）＋ 100 kg of ammonium sulfate （AS）/ha and 500 kg of the formulation 4-14-8 （N-P-K）＋200 kg of AS/ha, arranged in randomized blocks with four replicates. Attacks by S. frugiperda and D. maidis were higher as the nitrogen, sulfur, calcium and copper content in the leaves of corn increased. The effects of calcium and copper found in this study were not consistent with that of lower susceptibility associated with higher levels of these nutrients mentioned in the literature. We believe that there was a cumulative effect of the nutrients altogether, building up a nutritiously better vegetable which in turn had less reserve for defense, or that the content of both nutrients in the leaves were not high enough to induce such effects.

The photocatalytic properties of CeO2-Nb2O5 photocatalysts in heterogeneous photocatalysis(under ultraviolet and visible radiation)and in Fenton-like process were *** blue dye(MB)and phenol(Ph)were used as models of pollutant molecules for these reactions,and the photocatalysts were characterized by X-ray diffraction(XRD),diffuse reflectance spectroscopy(DRS),scanning electron microscopy(SEM),energy-dispersive X-ray spectroscopy(EDX)and thermally stimulated luminescence(TL).The results indicated that the addition of CeO2(0.3 wt%,1.0 wt%and 2.0 wt%)to Nb2O5 sensitized the resultant materials,increasing light absorption in the visible ***,there is a suitable formulation of CeO2-Nb2O5 photocatalysts to improve each photocatalytic *** heterogeneous photocatalysis,the addition of small CeO2 quantities to Nb2O5 was enough to improve the photocatalytic activity of CeO2-Nb2O5 photocatalysts(The best composition reported was CeO20.3 wt%.).The effectiveness of the catalyst was explained by the decrease in the number of trapping and luminescence centers in the conduction band of the material after the addition of CeO2 to Nb2O5,but a large amount of CeO2 decreased the number of trapping,luminescent and active centers to a large ***,in a Fenton-like process,the addition of CeO2 to Nb2O5 was favorable in all the proportions studied.(The best composition was 2.0 wt%CeO2.)In this case,the effectiveness was explained by the influence of the adsorption process(adsorption-triggered process),and the interactions between H2 O2 and Ce3+of the CeO2 in each photocatalyst thus formed surface peroxide species O22-,which induced the removal of the organic molecules under visible light.

Anthropogenic Disturbances and the Natural Vegetation Regeneration: A Case Study of a Forest Fragment Located in a Cuesta Relief Area, State of São Paulo, Brazil

The present study has been conducted in one of the five natural vegetation fragments at the Experimental Farm Edgardia, in Botucatu, São Paulo State, Brazil. In the past the forest fragment suffered disturbances due to the selective exploitation of tree species and the use of part of the surface as pasture. The objectives of this research were to evaluate the structure and the dynamics of natural vegetation. Three phytosociological surveys (2006, 2010 and 2014) were carried out in a sampled area of one hectare. The most abundant species were mapped in 2014, in a sampling unit with two hectares. Individuals of 37 families and 110 species have been recorded. The oscillation of the basal area observed through the forest inventories was due to the instability caused by disturbances. Only two out of the seven most abundant species mapped showed exponential diameter distribution. The unimodal distributions shown by the other five species have suggested that the vegetation has been occupying the most open spaces through regeneration in cycles. There was direct association between the spatial distribution and the dispersion syndrome for five out of the seven most abundant species. Three species have zoochoric dispersion, showing that wild animals populations have accomplished their function in the dispersion of seeds/fruit. The spatial distributions of four out of the seven most abundant species have provided evidence of how the regeneration of natural vegetation is being processed in this area.

Various post-harvest processes of rice are commonly employed,especially during the off-season,to ensure its consumption feasibility,which often affect the grain *** forms of drying,storage and processing of rice are evaluated to identify their effects on grain *** drying has emerged as an alternative to the widely-used intermittent-drying and fixed-bed-dryer methods of drying paddy *** of drying-air temperatures(between 40℃ and 60℃)according to the rice variety can improve quality,especially for exotic *** stored grain in hygroscopic balance,with water content between 11%to 15%,at temperatures between 16℃ and 20℃ and with intergranular relative humidity near 60%,allows 12 months of storage in a controlled environment without significant *** innovations,notably the application of artificial refrigeration to grain stored in bulk in vertical cylindrical silos and the use of impermeable packaging for storage,ensure the conservation of grain *** different stages and equipments used to obtain polished,brown and parboiled rice result in significant changes in the nutritional value of rice because of the removal of the outermost layers of the *** reduces the nutritional value and physical homogeneity of *** rice retains more bioactive compounds and nutrients because it does not lose the outer layer of the grains in the polishing *** rice,although less nutritious than brown rice,has better grain integrity and milling yield and less loss of nutrients than white rice.

Parkin protein expression and its impact on survival of patients with advanced colorectal cancer

Objective:Features of colorectal cancer such as natural history,molecular,chromosomal,and epigenetic alterations have been well ***,there is still a lack of accurate prognostic markers,which is evident by the lower overall survival rates of patients with advanced *** alterations in parkin protein expression have been described in colorectal cancer,the functional significance of this protein remains *** present study aimed to investigate the involvement of parkin expression in colorectal adenocarcinoma development and progression by evaluating the association between its expression,clinicopathological parameters,and expression of known proteins involved in colorectal ***:Tissue microarrays consisting of 73 tumor and 64 normal tissue samples were generated to examine parkin expression and localization by ***:A positive correlation of parkin and APC expression was observed in the superficial,intermediate,and profound regions of all cases(ρ=0.37;P=0.001).Parkin expression was also significantly associated with tumors in men(P=0.049),those of the mucinous subtype(P=0.028),and of advanced stage(III+IV,P=0.041).In addition,increased parkin expression was observed in the invasive front tumor region(P=0.013).More importantly,a positive correlation was found between parkin expression and the overall survival of patients with advanced colorectal cancer(P=0.019).Multivariate analysis showed that parkin expression was independent of any of the clinicopathological parameters evaluated in relation to patient ***:These results suggest that parkin expression status can be used as a potential independent prognostic marker of survival in advanced colorectal cancer.

Convergence Criterium of Numerical Chaotic Solutions Based on Statistical Measures

Solutions of most nonlinear differential equations have to be obtained numerically. The time series obtained by numerical integration will be a solution of the differential equation only if it is independent of the integration step h. A numerical solution is considered to have converged, when the difference between the time series for steps h and h/2 becomes smaller as h decreases. Unfortunately, this convergence criterium is unsuitable in the case of a chaotic solution, due to the extreme sensitivity to initial conditions that is characteristic of this kind of solution. We present here a criterium of convergence that involves the comparison of the attractors associated to the time series for integration time steps h and h/2. We show that the probability that the chaotic attractors associated to these time series are the same increases monotonically as the integration step h is decreased. The comparison of attractors is made using 1) the method of correlation integral, and 2) the method of statistical distance of probability distributions.

Fruiting increases total content of flavonoids and antiproliferative effects of Cereus jamacaru D.C. cladodes in sarcoma 180 cells in vitro

Objective: To evaluate the influence of fruiting phenological stage on total flavonoid content, antioxidant activity, and antiproliferative effects of Cereus jamacaru(C. jamacaru)(mandacaru) cladodes and fruit. Methods: Fruit and cladodes at vegetative and fruiting stage of C. jamacaru were collected. The fruit was dissected and bark, pulp, and seeds were separated. Vegetative and fruiting cladodes, together with bark, pulp, and seeds were used to obtain five hydroalcoholic extracts. The extracts were investigated for total flavonoid content, using AlCl3 colorimetric method, antioxidant activity by 2,2-diphenyl-1-picrylhydrazyl and 2,2′-azino-bis(3-ethylbenzthiazoline-6-sulfonic acid) radical scavenging capacity and Fe^(2+) ion chelating activity, and in vitro antiproliferative effects(sarcoma 180 cells) by 3-(4,5-dimethyl-2-thiazolyl)-2,5-diphenyl-2 H-tetrazolium bromide assay. Results: The extract of C. jamacaru cladodes at the fruiting stage showed higher flavonoid content compared to the other extracts. Seed extracts showed the highest antioxidant activity in 2,2-diphenyl-1-picrylhydrazyl and 2,2′-azino-bis(3-ethylbenzthiazoline-6-sulfonic acid) assays, and the extract of cladodes at vegetative stage showed better antioxidant activity in Fe^(2+) ion chelating activity. The extract of fruiting cladodes promoted higher antiproliferative effects compared to the other extracts. Conclusions: These findings suggest that fruiting increases the content of flavonoids and antiproliferative effects of C. jamacaru cladodes. Data reinforce the potential use of C. jamacaru cladodes and fruits as natural antioxidants and potent anticancer agent.

Study of Succinic Acid Production by Actinobacillus Succinogenes

Succinic acid has recently emerged as an important chemical （commodity） because it can be used for the manufacturing of synthetic resins and biodegradable polymers and as an intermediate for chemical synthesis. Till date, succinic acid is mainly produced by chemical processes, however, due to the environmental concerns and the concepts of sustainability, researches are directed towards the production of succinic acid by microbial fermentation. The fact that carbon dioxide （CO2） is needed by the microorganisms for succinic acid production is another interesting feature. The fermentation was carried out with Actinobacillus succinogenes using a two-level fractional factorial design 2sl. The variables analyzed and their levels were： concentration of glucose, yeast extract, temperature, pH and agitation. The results show that the variables that more influenced on succinic acid production were pH, temperature and yeast extract.
