Since the first publication regarding the existence of stem cells in cancer[cancer stem cells(CSCs)]in 1994,many studies have been published providing in-depth information about their biology and *** research has paved the way in terms of appreciating the role of CSCs in tumour aggressiveness,progression,recurrence and resistance to cancer *** CSCs for cancer therapy has still not progressed to a sufficient degree,particularly in terms of exploring the mechanism of dynamic interconversion between CSCs and *** the CSC scenario,the problem of cancer dissemination has been analyzed indepth with the identification and isolation of microRNAs(miRs),which are now considered to be compelling molecular markers in the diagnosis and prognosis of tumours in general and specifically in patients with non-small cell lung *** release of miRs via“exosomes”(small membrane vesicles(30-100 nm),the derivation of which lies in the luminal membranes of multi-vesicular bodies)released by fusion with the cell membrane is gaining *** exosomes play a significant role in maintaining a dynamic equilibrium state between CSCs and non-CSCs and their mechanism of activity is as yet *** studies on CSC-related exosomes will provide new perspectives for precision-targeted treatment strategies.

Eco-physiological studies on desert plants: germination of Halothamnus iraqensis Botsch. seeds under different conditions

With the aim to investigate if the halophyte Halothamnus iraqensis Botsch. can be suitable for re-vegetation and remediation of salt-affected lands, this study evaluated(1) the effects of photoperiod, thermoperiod, storage period and wings' presence on its seed germination, and(2) the ability of its seeds to have successful germination recovery after salt stress. Germination tests in different photoperiods(12 h light/12 h darkness and total darkness) and thermoperiods(15℃/20℃ and 20℃/25℃) were conducted for seeds collected in 2012, 2013, 2014, 2015 and 2016. The seeds collected in 2016 were sown under different salinity levels(0, 100, 200, 400 and 600 m M Na Cl) to assess the salinity tolerance during the germination. Wings' presence highly inhibited seed germination of this species in both photoperiods and thermoperiods under all salinity level treatments. In addition, the germination recovery occurred well when seeds were deprived of their wings. The photoperiod of 12 h light/12 h darkness and the thermoperiod of 15℃/20℃ were the best conditions for seed germination. Germination percentages of H. iraqensis seeds decreased with the increasing storage duration, especially after three years of the collection. In addition, H. iraqensis seeds were able to germinate under different salinity levels, and their germination percentages decreased with increasing salinity levels. H. iraqensis seeds have the ability to recover their germination after alleviating the salt stress, irrespective of photoperiod, highlighting the halophilous character of this species.

To explore the role of a novel Obg-like ATPase 1 （OLA1） in cancer metastasis, small interference RNA （siRNA） was used to knockdown the protein, and the cells were subjected to in vitro cell migration and invasion assays. Knockdown of OLA 1 significantly inhibited cell migration and invasion in breast cancer cell line MDA-MB-231. The knockdown caused no changes in cell growth but affected ROS production. In wound-healing assays, decreased ROS in OLA1-knockdown cells were in situ asso- ciated with the cells＇ decreased motile morphology. Further, treatment ofN-acetylcysteine, a general ROS scavenger, blunted the motility and invasiveness of MDA-MB-231 cells, similar to the effect of OLAl-knockdown. These results suggest that knock- down of OLA1 inhibits breast cancer cell migration and invasion through a mechanism that involves the modulation of intracellular ROS levels.

The endoplasmic reticulum(ER)is the site of entry of all proteins that function in the secretory pathway including the extracellular *** it controls the folding of newly synthesized secretory proteins,the ER is indispensable for the maintenance of proteostasis in the secretory *** the ER and,in part,in post-ER compartments,the quality control of protein folding is under the regulation of the unfolded protein response(UPR)*** UPR strategy is to enhance protein folding,increase the ER degradation pathway of misfolded proteins,and allow the exit from the ER of only correctly folded *** latter is controlled by the multimeric complex COPII,which also provides some of the components for ER-phagy the only route for the disposal of protein *** this overview,we wish to contribute to the introduction of new perspectives in the study of the mechanisms underlying the control of proteostasis within the secretory pathway.

Superlubricity of epitaxial monolayer WS2 on graphene

We report the superlubric sliding of monolayer tungsten disulfide (WS2) on epitaxial graphene (EG) grown on silicon carbide (SiC). Single-crystalline WS2 flakes with lateral size of hundreds of nanometers are obtained via chemical vapor deposition (CVD) on EG. Microscopic and diffraction analyses indicate that the WS2/EG stack is predominantly aligned with zero azimuthal rotation. The present experiments show that, when perturbed by a scanning probe microscopy (SPM) tip, the WS2 flakes are prone to slide over the graphene surfaces at room temperature. Atomistic force field-based molecular dynamics simulations indicate that, through local physical deformation of the WS2 flake, the scanning tip releases enough energy to the flake to overcome the motion activation barrier and trigger an ultralow-friction rototranslational displacement, that is superlubric. Experimental observations show that, after sliding, the WS2 flakes come to rest with a rotation of nπ/3 with respect to graphene. Moreover, atomically resolved measurements show that the interface is atomically sharp and the WS2 lattice is strain-free. These results help to shed light on nanotribological phenomena in van der Waals (vdW) heterostacks, and suggest that the applicative potential of the WS2/graphene heterostructure can be extended by novel mechanical prospects.

Colorectal cancer (CRC) remains one of the most common malignancies in the world. Although surgical resection combined with adjuvant therapy is effective at the early stages of the disease, resistance to conventional therapies is frequently observed in advanced stages, where treatments become ineffective. Resistance to cisplatin, irinotecan and 5-fluorouracil chemotherapy has been shown to involve mitogen-activated protein kinase (MAPK) signaling and recent studies identified p38α MAPK as a mediator of resistance to various agents in CRC patients. Studies published in the last decade showed a dual role for the p38α pathway in mammals. Its role as a negative regulator of proliferation has been reported in both normal (including cardiomyocytes, hepatocytes, fibroblasts, hematopoietic and lung cells) and cancer cells (colon, prostate, breast, lung tumor cells). This function is mediated by the negative regulation of cell cycle progression and the transduction of some apoptotic stimuli. However, despite its anti-proliferative and tumor suppressor activity in some tissues, the p38α pathway may also acquire an oncogenic role involving cancer related-processes such as cell metabolism, invasion, inflammation and angiogenesis. In this review, we summarize current knowledge about the predominant role of the p38α MAPK pathway in CRC development and chemoresistance. In our view, this might help establish the therapeutic potential of the targeted manipulation of this pathway in clinical settings.

In the animal kingdom there are countless strategies via which males optimize their reproductivesuccess when faced with male-male competition. These male strategies typically fall into two maincategories： pre- and post-copulatory competition. Within these 2 categories, a set of behaviors,referred to as reproductive suppression, is known to cause inhibition of reproductive physiologyand/or reproductive behavior in an otherwise fertile individual. What becomes evident when con-sidering examples of reproductive suppression is that these strategies conventionally encompassreproductive interference strategies that occur between members of a hierarchical social ***, mechanisms aimed at impairing a competitor＇s reproductive output are also present innon-social animals. Yet, current thinking emphasizes the importance of sociality as the primarydriving force of reproductive suppression. Therefore, the question arises as to whether there is anactual difference between reproductive suppression strategies in social animals and equivalentpre-copulatory competition strategies in non-social animals. In this perspective paper we explore abroad taxonomic range of species whose individuals do not repeatedly interact with the same indi-viduals in networks and yet, depress the fitness of rivals. Examples like alteration of male repro-ductive physiology, female mimicry, rival spermatophore destruction, and cementing the rival＇sgenital region in non-social animals, highlight that male pre-copulatory reproductive suppressionand male pre-copulatory competition overlap. Finally, we highlight that a distinction between malereproductive interference in animals with and without a social hierarchy might obscure importantsimilarities and does not help to elucidate why different proximate mechanisms evolved. We there-fore emphasize that male reproductive suppression need not be restricted to social animals.
