Low Prevalence of Campylobacteriosis in the Northern Region of India

Campylobacter is one of the most common bacterial enteropathogens of food borne origin in industrialized countries with C. jejuni being the most common species followed by C. coli. The prevalence of Campylobacters in and around Chandigarh, India was studied by phenotypic and genotypic methods. Fecal samples from 1145 diarrheal patients and 102 healthy subjects from hospital and community were cultured on Campylobacter media and identified by Gram stain, biochemical investigations and serotyping. Molecular identification of Campylobacter isolates was done using specific primers to unique regions of 16S rRNA, Campylobacter jejuni (hipO), Campylobacter coli (aspK), Campylobacter lari (glyA) and Campylobacter upsaliensis (lpxA) genes. Identification of specific genes to look for resistance to nalidixic acid, ciprofloxacin, tetracyclin and streptomycin was also done. Campylobacters were isolated from 2.6% of patients with diarrhea. Campylobacteriosis was more prevalent in children ≤5 years old and during summer season. The most frequent serotypes were S:27, B:2, Z5:52 and V:32. All the Campylobacters isolated by culture were confirmed genotypically by identification of 16S rRNA, hipO and aspK genes. Of the 30 isolates, 27 were C. jejuni and 3 were C. coli. No C. lari or C. upsaliensis were detected. Antibiotic resistance was 40% for nalidixic acid, 23.3% for ciprofloxacin, 50% for tetracyclin and 20% for streptomycin. Campylobacter prevalence is low in the region with C. jejuni being the most common species. A high degree of resistance was found for nalidixic acid and tetracyclin but moderate for ciprofloxacin and streptomycin.

Managing cardiovascular disease risk in South Asian kidney transplant recipients

South Asians(SA)are at higher cardiovascular risk than other ethnic groups,and SA kidney transplant recipients(SA KTR)are no *** KTR experience increased major adverse cardiovascular events both early and late *** risk management should therefore begin well before *** candidates may require aggressive screening for pretransplant cardiovascular disease(CVD)due to their ethnicity and *** SA ethnicity during the pre-transplant evaluation may enable programs to better assess cardiovascular risk,thus allowing for earlier targeted periand post-transplant intervention to improve cardiovascular *** remains the most prominent post-transplant cardiovascular risk factor in SA *** also clusters with other metabolic syndrome components including lower high-density lipoprotein cholesterol,higher triglycerides,hypertension,and central obesity in this ***,metabolic syndrome,and obesity are all significant CVD risk factors in SA KTR,and contribute to increased insulin *** biomarkers such as adiponectin,apolipoprotein B,and lipoprotein(a)may be especially important to study in SA *** interventions to improve health behaviors involving diet and exercise may especially benefit SA ***,there are few interventional clinical trials specific to the SA population,and none are specific to SA *** all cases,understanding the nuances of managing SA KTR as a distinct post-transplant group,while still screening for and managing each CVD risk factor individually in all patients may help improve the long-term success of all kidney transplant programs catering to multi-ethnic populations.

Geochemical analysis and quality assessment of geothermal water in Gujarat,India

Geothermal water can have a wide range of direct applications,including drinking,domestic use,irrigation and industrial *** study analyses geothermal water from Dholera,Diu,Gandhar,Kutch,Porbandar,Tulsishyam,and Unai regions located in Gujarat,India,to determine the hydrochemistry for establishing the water's suitability for drinking and irrigation *** each region,three different water samples were *** drinking purpose,total eleven water quality parameters such as pH,electrical conductivity,total dissolved solids,calcium,magnesium,sodium,potassium,chloride,sulphate,carbonate and bicarbonate were *** and Durov plots were used to understand the ionic composition and evolution of *** for irrigation,the parameters such as total dissolved solids,sodium absorption ration,electrical conductivity,and potential salinity were used for water quality *** Plot was plotted to understand the sodium and salinity hazard of *** quality index and irrigation water quality index were used to assess the water quality for making the analytical procedure *** findings indicate that the water samples from the Dholera and Diu areas had extremely high values for both the water quality index and total dissolved solids *** in these places may be appropriate for making *** the other samples show adequacy for utilization in irrigation purpose.

Mining the 3′UTR of Autism-implicated Genes for SNPs Perturbing MicroRNA Regulation

Autism spectrum disorder（ASD） refers to a group of childhood neurodevelopmental disorders with polygenic etiology. The expression of many genes implicated in ASD is tightly regulated by various factors including microRNAs（miRNAs）, a class of noncoding RNAs 22 nucleotides in length that function to suppress translation by pairing with ‘miRNA recognition elements’（MREs） present in the 30untranslated region（30UTR） of target mRNAs. This emphasizes the role played by miRNAs in regulating neurogenesis, brain development and differentiation and hence any perturbations in this regulatory mechanism might affect these processes as well. Recently, single nucleotide polymorphisms（SNPs） present within 30UTRs of mRNAs have been shown to modulate existing MREs or even create new MREs. Therefore, we hypothesized that SNPs perturbing miRNA-mediated gene regulation might lead to aberrant expression of autism-implicated genes, thus resulting in disease predisposition or pathogenesis in at least a subpopulation of ASD individuals. We developed a systematic computational pipeline that integrates data from well-established databases. By following a stringent selection criterion, we identified 9 MRE-modulating SNPs and another 12 MRE-creating SNPs in the 30UTR of autism-implicated genes. These high-confidence candidate SNPs may play roles in ASD and hence would be valuable for further functional validation.

Ballistic parameter plays a major role in determining the re-entry *** ballistic coefficient offers an optimal re-entry,wherein the vehicle decelerates higher up in the atmosphere thereby decreasing the imposed aerothermal *** current computational study proposes an add-on,to the existing Orion-based re-entry vehicle:a duct circumventing the capsule from the shoulder to the base,to improve the aerocapture ability of the re-entry *** design cases are categorised based on a non-dimensional parameter termed the Annular Area Ratio(AAR).Dragand ballistic coefficient of the Ducted Re-entry Vehicles(DRVs)at various Mach numbers are evaluated and compared with those of the baseline *** results show that the proposed design increases the drag for all the AARs considered in the subsonic *** the supersonic regime,ducted models of higher AAR are more promising with the increase in Mach *** also exhibit lower ballistic coefficients than their baseline counterparts.

SMS 2.0: An Updated Database to Study the Structural Plasticity of Short Peptide Fragments in Non-redundant Proteins

The function of a protein molecule is greatly influenced by its three-dimensional （3D） structure and therefore structure prediction will help identify its biological function. We have updated Sequence, Motif and Structure （SMS）, the database of structurally rigid peptide fragments, by combining amino acid sequences and the corre- sponding 3D atomic coordinates of non-redundant （25%） and redundant （90%） protein chains available in the Protein Data Bank （PDB）. SMS 2.0 provides information pertaining to the peptide fragments of length 5-14 resi- dues. The entire dataset is divided into three categories, namely, same sequence motifs having similar, intermedi- ate or dissimilar 3D structures. Further, options are provided to facilitate structural superposition using the pro- gram structural alignment of multiple proteins （STAMP） and the popular JAVA plug-in （Jmol） is deployed for visualization. In addition, functionalities are provided to search for the occurrences of the sequence motifs in other structural and sequence databases like PDB, Genome Database （GDB）, Protein Information Resource （PIR） and Swiss-Prot. The updated database along with the search engine is available over the World Wide Web through the following URL http：//***/sms/.

Homepeptide Repeats:Implications for Protein Structure,Function and Evolution

Analysis of protein sequences from Mycobacterium tuberculosis H37Rv （Mtb H37Rv） was performed to identify homopeptide repeat- containing proteins （HRCPs）. Functional annotation of the HRCPs showed that they are preferentially involved in cellular metabolism. Furthermore, these homopeptide repeats might play some specific roles in protein-protein interaction. Repeat length differences among Bacteria, Archaea and Eukaryotes were calculated in order to identify the conservation of the repeats in these divergent kingdoms. From the results, it was evident that these repeats have a higher degree of conservation in Bacteria and Archaea than in Eukaryotes. In addi- tion, there seems to be a direct correlation between the repeat length difference and the degree of divergence between the species. Our study supports the hypothesis that the presence of homopeptide repeats influences the rate of evolution of the protein sequences in which they are embedded. Thus, homopeptide repeat may have structural, functional and evolutionary implications on proteins.

Automatic Evaluation of Test Strips for Anti-Ganglioside Antibodies in Patients with Guillain-BarréSyndrome Using EUROLineScan Software

Campylobacter jejuni infection has been implicated in the pathogenesis of Guillain-Barré syndrome (GBS) due to production of humoral immune response against neural antigens. A case-control study was performed in a tertiary care teaching hospital for the estimation of anti-ganglioside antibodies in GBS patients and their controls. Blood samples were collected from 59 GBS cases, 58 neurological controls (NC) and 60 non-neurological control (NNC) patients for automatic estimation of IgG and IgM antibodies to seven gangliosides using EUROLineScan software. Antibodies of IgG class for GM1 were highly significant in GBS (p = 0.000) and NC (p = 0.031) compared to NNC. However GBS group was not significant (p = 0.413) compared to NC. For GM2 ganglioside, GBS and NC groups were significant (p = 0.000) compared to NNC, but GBS group was not significant (p = 0.999) compared to NC. For GM3 ganglioside, GBS and NC groups were significant (p = 0.000) compared to NNC;but GBS group was insignificant (p = 0.858) compared to NC with similar trend for all other ganglioside antibodies. When IgM class of antibodies was evaluated for GM1, GBS group was not significant (p = 0.604) whereas NC group was significant (p = 0.000) compared to NNC. GBS group was not significant compared to NC (p = 0.011). The trend was the same for GM2 antibodies. For GM3, GBS group was significant (p = 0.010) and NC was near significant (p = 0.055) compared to NNC. However GBS group was not significant (p = 0.808) compared to NC. No groups were significant (p > 0.05) in relation to the remaining gangliosides except for GQ1b where GBS group (p = 0.001) and NC group were significant (p = 0.000) compared to NNC. GBS group was also significant (p = 0.001) compared to NC and NNC. Anti-gangliosides antibodies were present in highly significant levels in the GBS group, though they were also present in the non-paralytic neurological control patients compared to the non-neurological control group.
