To study the molecular mechanisms involved in the hepatoprotective effects of naringenin (NAR) on carbon tetrachloride (CCl4)-induced liver fibrosis. METHoDSThirty-two male Wistar rats (120-150 g) were randomly divided into four groups: (1) a control group (n = 8) that received 0.7% carboxy methyl-cellulose (NAR vehicle) 1 mL/daily p.o.; (2) a CCl4 group (n = 8) that received 400 mg of CCl4/kg body weight i.p. 3 times a week for 8 wk; (3) a CCl4 + NAR (n = 8) group that received 400 mg of CCl4/kg body weight i.p. 3 times a week for 8 wk and 100 mg of NAR/kg body weight daily for 8 wk p.o.; and (4) an NAR group (n = 8) that received 100 mg of NAR/kg body weight daily for 8 wk p.o. After the experimental period, animals were sacrificed under ketamine and xylazine anesthesia. Liver damage markers such as alanine aminotransferase (ALT), alkaline phosphatase (AP), γ-glutamyl transpeptidase (γ-GTP), reduced glutathione (GSH), glycogen content, lipid peroxidation (LPo) and collagen content were measured. The enzymatic activity of glutathione peroxidase (GPx) was assessed. Liver histopathology was performed utilizing Masson’s trichrome and hematoxylin-eosin stains. Zymography assays for MMP-9 and MMP-2 were carried out. Hepatic TGF-β, α-SMA, CTGF, Col-I, MMP-13, NF-κB, IL-1, IL-10, Smad7, Smad3, pSmad3 and pJNK proteins were detected via western blot. RESULTSNAR administration prevented increases in ALT, AP, γ-GTP, and GPx enzymatic activity; depletion of GSH and glycogen; and increases in LPo and collagen produced by chronic CCl4 intoxication (P ver histopathology showed a decrease in collagen deposition when rats received NAR in addition to CCl4. Security Regulations in Mexican Renewable Energies: Case of Geothermal Projects

A review of natural resources existing in México is done. The description of the renewable energies for electricity generation operating at date along the country, includes hydro, wind, solar, biomass and geothermal, among others. The installed capacity (to 2012) in México for electric generation from renewable energies is equivalent to 22% of total generation capacity. México has geothermal resources, which can be classified as high and low enthalpy, and of hot dry rock. To date, the exploitation has focused mainly on high enthalpy geothermal fields. Geothermal power plants do not burn fuel, preventing gas emissions helping to reduce global warming and greenhouse effect. Security risks in México geothermal fields, as a part of renewable energies linked to Smart Grids, are described emphasizing their geographical locations to facilitate the exposure to dangerous events. The results about research on Mexican official Norms protecting environment related with geothermal operation projects are shown. The Mexican geothermal projects have developed under rules that provide security to workers and people, avoiding impacts on the environment. However, it was found that it necessarily emphasized previsions to damages and remedial actions for grids due to risks by natural contingencies (cyclones, winds, earthquakes) and by artificial causes such as vandalism (grids breaking, fire, explosions, etc.). Unfortunately, there are no preventive norms against natural risks. After all the analyses carried out, security must be considered by nature a dynamic and ever-changing process.

C3G is a GEF(guanine nucleotide exchange factor)for Rap GTPases,among which the isoform Rap1b is an essential protein in platelet *** transgenic mouse models with platelet-specific overexpression of C3G or mutant C3GΔCat,we have unveiled a new function of C3G in regulating the hemostatic function of platelets through its participation in the thrombin-PKCRap1b pathway.C3G also plays important roles in angiogenesis,tumor growth,and metastasis through its regulation of the platelet *** addition,C3G contributes to megakaryopoiesis and ***,we used a platelet-specific C3G-Ko mouse model to further support the role of C3G in hemostasis.C3G-Ko platelets showed a significant delay in platelet activation and aggregation as a consequence of the defective activation of Rap1,which resulted in decreased thrombus formation in ***,we explored the contribution of C3G-Rap1b to platelet signaling pathways triggered by thrombin,PMA or ADP,in the referenced transgenic mouse model,through the use of a battery of specific *** found that platelet C3G is phosphorylated at Tyr504 by a mechanism involving *** phosphorylation was shown to be positively regulated by ERKs through their inhibition of the tyrosine phosphatase ***,C3G participates in the ADP-P2Y12-PI3K-Rap1b pathway and is a mediator of thrombin-TXA2 ***,it inhibits the synthesis of TXA2 through cPLA2 *** together,our data reveal the critical role of C3G in the main pathways leading to platelet activation and aggregation through the regulation of Rap1b.

Research on two-dimensional(2D) materials is one of the most active fields in materials science and nanotechnology. Among the members of the 2D family, layered hydroxides(LHs) represent an exceptional case of study due to their unparalleled chemical versatility which allows the modulation of their physicochemical properties at will. Nowadays, LHs based on earth-abundant metals are key materials in the areas of energy storage and conversion, hybrid materials or magnetism. α-Co hydroxides(Simonkolleite-like structures) are promising phases with tuneable electronic and magnetic properties by ligand modification. However, even in the simple case of α-Co^(Ⅱ) hydroxychlorides, the preparation of well-defined large 2D crystals is not straightforward, hindering the development of fundamental studies. Herein, we present the synthesis of 2D hexagonal crystals with outstanding sizethickness relationship(diameter > 5 μm and thickness of 20 ± 7 nm) by a simple homogeneous synthesis taking place at room temperature. In structural terms, no differences are observed between our layered materials and those obtained hydrothermally. However, dynamic susceptibility measurements alert about different arrangements of the magnetic sublattices, which have been rationalized with structural DFT calculations. This work provides an extremely easy bottom-up method to obtain high-quality 2D crystals based on α-CoIIhydroxides,paving the way for the development of fundamental studies and applications.

E-Cadherin Dysfunction and Cancer

E-cadherin molecules are cell-cell molecular connectors, but also act attaching cell surface to the cytoskeleton through catenin’s and additional partner proteins. Its main function is to regulate cellular adhesion and motility, and therefore acts as an invasion suppressor system. Its role is crucial in the induction and maintenance of cell polarity and differentiation, and in the organization and maintenance of tissue architecture. Downregulation or loss of its function is associated with an invasive and aggressive phenotype in many types of human cancers. In alterations of animal development, E-cadherin dysfunction influence in premature lethality or epidermal barrier and immunity defects. However, new hypothesis over its promoter role in tissue invasion is on focus. In addition to its tumor suppressor role, E-cadherin is a guiding molecule in collective cell migration increasing the metastasis risk during in vivo tumorigenesis. This E-cadherin function explains the retention of the functional E-cadherin expression, and that the epithelial-mesenchymal transition (EMT) is not required for the metastasis occurrence. However, like a switch, in some tumoral environments E-cadherin post-translational modifications may cause cell cluster migration. Like in development, in certain in vivo tumoral contexts, E-cadherin apparently involves signaling rather than cell contact formation. In this report, we explore the possibility of a novel role of thyroid hormones (THs) in the signal via E-cadherin-catenins, this not only should be involved in development and homeostasis, but also in cancer susceptibility of gastrointestinal tract.

Sanger Sequencing Implementation in Clinically Ill Patients for Bacterial and Fungal Pathogens Identification

The diagnosis of bacterial or fungal infections requires the identification of the pathogen etiology in the shortest time possible. Although some biomarkers are used as indicators of bacterial infections, their specificity and sensitivity are highly variable, and there is no direct relationship between the level increase of these biomarkers for mycosis. It is common to obtain negative microbiological cultures in patients infected by non-culturable, intracellular bacteria or mycosis, even though there is a high clinical suspicion of infection. This study identifies the pathogen present in critically infected patients through 16S and 18S/eEF1 genes detection by polymerase chain reaction (PCR) coupled with Sanger sequencing. Thirty clinical samples were evaluated by PCR, of which 40% were positive for fungi, 23.33% for bacteria, 26.7% for fungi and bacteria, and 10% for no pathogen. The PCRs outcomes period for bacteria or fungi was one day compared to seven and up to 14 days (on average) of microbiological culture for bacteria and fungi. Then, we assessed the relationship with the most used biomarkers (procalcitonin, C-reactive protein, globular sedimentation velocity, and the neutrophil-lymphocyte index). This combination of molecular techniques has been shown as helpful in identifying intracellular bacteria and fungi that are difficult to culture by conventional methods. Screening with genomic markers 16S and 18S/eEF1 by PCR allowed us to optimize the time to obtain the result of the infection caused by bacteria or fungi. Also, identifying the specific etiological microorganism by Sanger sequencing was very helpful in avoiding the progression of the disease and setting targeted treatment with better clinical outcomes.

PUB16 gene expression under abiotic stress and their putative role as an ARM repeat protein in <i>Arabidopsis thaliana</i>self-pollination pathway

The armadillo repeat super-family proteins (ARM repeat super-family proteins) possess tandem armadillo repeats and have been postulated to play different roles in plant development, morphogenesis, defense, cell death, and signal transduction through hormone signalling. In The Arabidopsis Information Resource (TAIR), we found 113 loci closely related to ARM repeat family proteins. This extensive group of proteins was studied in flowers tissues by western blot using antibodies directed against the most conserved region of the ARM repeat family proteins. The amino acid residues sequences from TAIR were aligned and the resulting phylogenetic tree allows us to inferring their evolutionary relationships. The main finding was the high similarity between the gene product of PUB16 (At5g01830, A. thaliana) and ARC1 (Brassica napus). In order to search a possible role for PUB16 we carried out stress bioassays using hormonal and saline approaches. Gene expression using RT-PCR showed that some of the ARM repeat super-family proteins are expressed both under salt or hormonal stress conditions. Particularly these studies allowed to detect and semi-quantify PUB16 gene expression in normal or stress growth conditions. In this approach it was revealed that, only in presence of GA, the expression of mRNA-PUB16 became evident. To morphologically verify the increasing number of germinated pollen grain in gibberellins treated flowers, we used epi-fluorescence microscopy assay. These results suggest that PUB16 may participate in GA signaling pathway favoring self-pollination.

Helicobacter pylori (H. pylori) affects nearly half of the world’s population and, thus, is one of the most frequent and persistent bacterial infections worldwide. H. pylori is associated with peptic ulcer disease, gastric ulcers, mucosa-associated lymphoid tissue lymphoma, and gastric cancer. various diagnostic methods exist to detect infection, and the choice of one method or another depends on several factors, such as accessibility, advantages and disadvantages of each method, cost, and the age of patients. once H. pylori infection is diagnosed, the clinician decides whether treatment is necessity, according to the patient’s clinical condition. Typically, eradication of H. pylori is recommended for treatment and prevention of the infection. Cure rates with the standard triple therapy are acceptable, and effective quadruple therapies, sequential therapies, and concomitant therapies have been introduced as key alternatives to treat H. pylori infection. In this work, we review the main diagnostic methods used to identify H. pylori infection and to confirm eradication of infection. In addition, key factors related to treatment are reviewed.

Logarithm of a Function, a Well-Posed Inverse Problem

It poses the inverse problem that consists in finding the logarithm of a function. It shows that when the function is holomorphic in a simply connected domain , the solution at the inverse problem exists and is unique if a branch of the logarithm is fixed. In addition, it’s demonstrated that when the function is continuous in a domain , where is Hausdorff space and connected by paths. The solution of the problem exists and is unique if a branch of the logarithm is fixed and is stable;for what in this case, the inverse problem turns out to be well-posed.
