Keratinocyte growth factor-2 and autologous serum potentiate the regenerative effect of mesenchymal stem cells in cornea damage in rats

AIM:To investigate the healing process after severe corneal epithelial damage in rats treated with mesenchymal stem cells(MSCs)cultured with or without keratinocyte growth factor(KGF-2)and autologous serum(AS)on amniotic membrane(AM).Many patients are blind and devastated by severe ocular surface diseases due to limbal stem cell *** marrow-derived MSCs are potential sources for cellbased tissue engineering to repair or replace the corneal tissue,having the potential to differentiate to epithelial ***:The study included 5 groups each including 10 female'Sprague Dawley'rats in addition to20 male rats used as bone marrow *** I rats received AM+MSCs,Group II rats AM+MSCs cultured with KGF-2,Group III rats AM+MSCs cultured with KGF-2+AS,Group IV rats only AM and Group V rats,*** was derived from blood drawn from male rats and bone marrow was obtained from the femur and tibia bones of the same *** effect was evaluated with clinical,histopathological and immunohistochemical *** engraftment was demonstrated via detection of donor genotype(Y+)in the recipient tissue(X)with polymerase chain ***:Corneal healing was significantly better in Groups I-III rats treated with MSC transplantation compared to Group IV and Group V rats with supportive treatment *** best results were obtained in Group III rats with 90%transparency,70%lack of neovascularization,and 100%epithelium damage limited to less than 1/4 of ***:We suggest that culture of MSCs with KGF-2 and AS on AM is effective in corneal repair in case of irreversible damage to limbal stem cells.

Evaluating ultrasound signals of carbon steel fatigue testing using signal analysis approaches

The application of ultrasound techniques to monitor the condition of structures is becoming more prominent because these techniques can detect the early symptoms of defects such as cracks and other *** early detection of defects is of vital importance to avoid major failures with catastrophic *** assessment of an ultrasound technique was used to investigate fatigue damage *** tests were performed according to the ASTM E466-96 standard with the attachment of an ultrasound sensor to the test *** 1045 carbon steel was used due to its wide application in the automotive industry.A fatigue test was performed under constant loading stress at a sampling frequency of 8 *** sets of data acquisition systems were used to collect the fatigue strain signals and ultrasound *** of the signals were edited and analysed using a signal processing *** methods were used to evaluate the signals,the integrated Kurtosis-based algorithm for z-filter technique(I-kaz) and the short-time Fourier transform(STFT).The fatigue damage behaviour was observed from the initial stage until the last stage of the fatigue *** results of the I-kaz coefficient and the STFT spectrum were used to explain and describe the behaviour of the fatigue damage.I-kaz coefficients were ranged from 60 to 61 for strain signals and ranged from 5 to 76 for ultrasound signals.I-kaz values tend to be high at failure point due to high amplitude of respective *** spectrogram displays the colour intensity which represents the damage severity of the strain signals.I-kaz technique is found very useful and capable in assessing both stationary and non-stationary signals while STFT technique is suitable only for non-stationary signals by displaying its spectrogram.

A rare side effect of betamethasone applied subconjunctivally is described. A 51-year-old man who had undergone penetrating keratoplasty in his left eye 2 m onths previously developed graft rejection and was treated by subconjunctival in jection of beta methasone. Within 2 weeks, the patient complained of conjunctival hyperemia and irritation. Biomicroscopic examination revealed conjunctival and subconjunctival necrosis with white necrotic tissue at the subconjunctival corticosteroid injec tion site. Simple debridement and primarywound closure of the involved area resu lted in complete healing. Ophthalmologists should be aware that subconjunctival injection of betamethasone may cause conjunctival necrosis.

BACKGROUND AND OBJECTIVES: To evaluate the outcomes of vitreoretinal surgery f or the ocular complications of Behet’s disease. PATIENTS AND METHODS: This re trospective study included 20 eyes of 15 patients with Behet’s disease (9 men , 6 women; mean age at surgery, 31.0±1.8 years; range, 21-44 years) who had un dergone vitreoretinal surgery for Behet’s disease at Ondokuz Mayis University , Faculty of Medicine, Department of Ophthalmology, Samsun, Turkey, between Janu ary 2000 and September 2003. Postoperative visual acuity outcome, course of uvei tis, and complications were evaluated in all patients. RESULTS: Indications for surgery were vitreous opacities in 10 eyes (50%), cystoid macular edema in 5 ey es (25%), epiretinal membrane in 3 eyes (15%)-, and vitreous hemorrhage in 2 eyes (10%). At the last postoperative follow-up, visual acuity had increased 2 Snellen lines or more in 10 (50%) of 20 eyes. Cystoid macular edema completely improved in 3 (60%) of 5 eyes after vitrectomy. Uveitis attacks significantly decreased during follow-up (P=.015). Immunosuppressive treatment was stopped in 11 (73%) of 15 patients. Retinal detachment and phthisis were not observed in any eye. CONCLUSION: Vitreoretinal surgery may be useful in the treatment of pos terior segment complications of Behet’s disease.
