Neuroprotective effect of a dietary supplement against glutamate-induced excitotoxicity in retina

AIM: To evaluate the neuroprotective effect of a dietary supplement(ClearVision EX~?;CV) against glutamateinduced excitotoxicity in ***: We evaluated the protective effects CV on glutamate-induced cell toxicity of an immortalized mouse hippocampal cell line(HT-22) in vitro and N-methyl-Daspartate(NMDA) induced retinal injury in vivo. Once-daily oral administration of CV or vehicle(5% Arabic gum) was started the day before the NMDA injection and continued until the end of the study. Electroretinograms(ERGs) were recorded to evaluate the retinal function at 2 d after NMDA injection. Furthermore, a histological evaluation, Western blot analysis, and immunohistochemistry were performed for assessing the signal transduction pathway. RESULTS: HT-22 cell death was induced by the addition of glutamate and co-incubation with CV protected against it. Oral administration of CV inhibited the decrease in scotopic threshold response amplitudes induced by the intravitreal injection of NMDA and those of the thickness of the inner retinal layer in the histological evaluation. The increased phosphorylated levels of extracellular signalregulated kinase(ERK) but not cAMP response element binding protein(CREB) or Akt were observed 1 h after NMDA injection in both the vehicle-and CV-treated rats;however, p ERK activation was no more upregulated at 3 h after NMDA injection. pERK upregulation was observed in Müller ***: CV shows a protective effect against both glutamate-induced HT-22 cell death and NMDAinduced retinal damage. pERK upregulation in the Müller cells plays a key role in the protective effect of CV against glutamate-induced retinal toxicity.

Simple Detection Method of Biogenic Amines in Decomposed Fish by Intramolecular Excimer Fluorescence

Biogenic amines are known to have various biological functions such as not only neurotransmitter and cell proliferation but also food poisoning. Bacterially-decomposed amines such as histamine, agmatine, putrescine, cadaverine, spermidine and spermine cause allergic symptoms. We developed simple method for measurement of polyamine as indicator of food decomposition with a fluorometer by using 4-(1-Pyrene)butyric acid N-hydroxysuccinimide ester (PSE). PSE reacts with primary and secondary amino moieties of polyamines and produces the intramolecular excimer fluorescence. Excimer fluorescence with broad peak at around 470 nm was clearly detected in linear type biogenic amines such as putrescine, cadaverine, spermidine and spermine at 10 mM. However neither histamine nor trimethylamine altered the fluorescence. Decomposed sardine and mackerel by improper storage showed stronger intensity than fresh ones. Comparing with OPA-method, PSE method was useful for screening biogenic amines present in food, esp. fish since the analysis was simple after one-step purification procedure. An inexpensive system which can rapidly detect biogenic amines from food is necessary in a medium and small-sized food business. The technique using excimer has potential to realize the high through-put screening system for evaluation of food freshness and is expected to bring the public interests such as food security and safety of consumer.

Assessment of the Water Quality in the Ariake Sea Using Principal Component Analysis

The aim of this study is to investigate the condition of the Ariake Sea, Japan, which has been suffering from severe environmental issues for the past few decades. Water quality data have been generated from several points in this area for over 30 years by the Fukuoka, Saga, Kumamoto, and Nagasaki prefectures. In order to understand the characteristics of this sea, principal component analysis (PCA) was utilized using 11 water quality parameters;transparency, temperature, salinity, dissolved oxygen (DO), chemical oxygen demand (COD), dissolved inorganic nitrogen (DIN), ammonium-nitrogen (NH4+-N), nitrate-nitrogen (NO3--N), nitrite-nitrogen (NO2--N), phosphate-phosphorus, (PO43--P) and silica. PCA conveyed the amount of nutrients originating from the river, the organic pollution level, and seasonal changes. Subsequently, principal component scores were calculated for each point. It was concluded that the Ariake Sea environment has been affected by two main factors, which are the nutrients from the Chikugo River and anticlockwise tidal residual flow. These two factors must be considered for the environmental restoration of the Ariake Sea.

A Study on Ecotourism Considered from Viewpoint of Landscape

＜正＞Recently, ecotourism has been acknowledged as one form of sightseeing. Ecotourism is an idea of sightseeing that aims at uniting the protection of regional resources such as nature, the history, and the culture, the sightseeing industry, and revitalization of local economy. Landscape architects have been playing a constant role to the sightseeing development and the regional plan in the area of conventional sightseeing. Then, what sort of relationship are ecotourism and ’Landscape’ building now? What does ecotourism have to do with ’Landscape’? Do actual ecotours as marketed tour packages make a point of ’Landscape’ ? In this study, we consider ecotourism from the aspect of ’Landscape’. Specifically, we examine the definition of ecotourism, the history of ecotourism and sightseeing, and actual ecotours. Firstly, definitions of ecotourism were examined by searching for terms related to ’Landscape’, and were examined how the terms were described. Secondly, the chronology was made in connection with ecotourism and ’Landscape’, and was analyzad. Thirdly, terms related ’Landscape’ were searched in the description of actual ecotours registered in ECOTOUR SOURAN, and the results were compared with that of the general sightseeing tours. As a result, we could not find remarkable sign that ’Landscape’ has special recognition in the definition, the history, and ecotour programs of ecotourism. In the definitions of ecotourism, only NACS-J use the term of ’nature （keikan）’ The landscape may be indirectly expressed as in ’nature’, history’, ’culture’, and an inclusive word ’environment’. But, we may say that the concept of ’Landscape’ is not clearly considered in ecotourism. In the history of ecotourism, landscape seems to have stayed as one of sightseeing objects while both objects and forms of the sightseeing are diversified. In the tours registered as ecotours, the number of tours for which the term related to the landscape was used limited to 22 out of 757. The occurrences

Growth of Gallium Oxide Nanowires by Pulsed Laser Deposition

We report on the synthesis of gallium oxide nanowires by pulsed laser deposition using a gold catalyst. In the vapor-liquid-solid process, gold thickness was the crucial parameter for deciding the morphology of nanowires. In the case of 1 nm thick gold, homogeneous nanowire growth was confirmed at temperatures of 700°C to 850°C. Transmission electron microscopy and selected area electron diffraction measurements showed that the nanowires were polycrystalline. In the cathode luminescence spectra, UV, blue, green and red emission peaks were observed, as reported in previous studies. As growth temperature was increased, the relative intensities of blue, green, and red emissions decreased. Thermal annealing treatments were effective in decreasing the blue, green and red emission peaks, suggesting that these emission peaks were associated with oxygen vacancies.

Differences between diffuse and focal autoimmune pancreatitis

AIM:To investigate differences in clinical features between diffuse-and focal-type autoimmune pancreatitis(AIP).METHODS:Based on radiological findings by computed tomography and/or magnetic resonance imaging,we divided 67 AIP patients into diffuse type(D type) and focal type(F type).We further divided F type into head type(H type) and body and/or tail type(B/T type) according to the location of ***,we classified the 67 AIP patients into three groups:D type,H type and B/T *** compared the three types of AIP in terms of clinical,laboratory,radiological,functional and histological findings and clinical ***:There were 34 patients with D-type,19 with H-type and 14 with B/T-type *** obstructive jaundice was frequently detected in D-typepatients(88%) and H-type patients(68%),no B/T-type patients showed jaundice as an initial symptom(P < 0.001).There were no differences in frequency of abdominal pain,but acute pancreatitis was associated more frequently in B/T-type patients(36%) than in D-type patients(3%)(P = 0.017).Serum immunoglobulin G(IgG)4 levels were significantly higher in D-type patients(median 309 mg/dL) than in B/T-type patients(133.5 mg/dL)(P = 0.042).Serum amylase levels in B/T-type patients(median:114 IU/L) were significantly greater than in H-type patients(72 IU/L)(P = 0.049).Lymphoplasmacytic sclerosing pancreatitis(LPSP) was histologically confirmed in 6 D-type,7 H-type and 4 B/T-type patients;idiopathic duct-centric pancreatitis was observed in no *** fibrosis and abundant infiltration of CD20-positive B lymphocytes with few IgG4-positive plasma cells were detected in 2 B/T-type *** therapy was effective in all 50 patients(31 D type,13 H type and 6 B/T type).Although AIP relapsed during tapering or after stopping steroids in 3 D-type and 3 H-type patients,no patients relapsed in B/T *** follow-up,radiological features of 6 B/T-type patients were not changed and 1 B/T-type patient impr

The marine area of Japan, including territorial waters and the exclusive economic zone, is the sixth largest in the world at about 4,470,000 km2. Therefore, it is becoming necessary to establish appropriate means of transportation other than ships in order to utilize the area efficiently. In this respect, ultra-light seaplanes are attracting attention from the viewpoint of protecting the natural environment. Accordingly, JRPS （Japan Reinforced Plastics Society） is currently developing FRP （fiber-reinforced plastic） floats for such planes. In this study, we conducted simulations of seaplane behavior during alighting by using the smoothed particle hydrodynamics method, which is one of the functions in the PAM-CRASH solver, and we present the observed trend in the vertical acceleration of the floats as a first step toward deriving the impact force from analytical data.

Utility of pancreatography for diagnosing autoimmune pancreatitis

AIM: To identify pancreatographic findings that facilitate differentiating between autoimmune pancreatitis (AIP) and pancreatic cancer (PC) on endoscopic retrograde cholangiopancreatography (ERCP) and magnetic resonance cholangiopancreatography (MRCP). METHODS: ERCP findings of 48 AIP and 143 PC patients were compared. Diagnostic accuracies for AIP by ERCP and MRCP were compared in 30 AIP patients. RESULTS: The following ERCP findings suggested a diagnosis of AIP rather than PC. Obstruction of the main pancreatic duct (MPD) was more frequently detected in PC (P < 0.001). Skipped MPD lesions were detected only in AIP (P < 0.001). Side branch derivation from the narrowed MPD was more frequent in AIP (P < 0.001). The narrowed MPD was longer in AIP (P < 0.001), and a narrowed MPD longer than 3 cm was more frequent in AIP (P < 0.001). Maximal diameter of the upstream MPD was smaller in AIP (P < 0.001), and upstream dilatation of the MPD less than 5 mm was more frequent in AIP (P < 0.001). Stenosis of the lower bile duct was smooth in 87% of AIP and irregular in 65% of PC patients (P < 0.001). Stenosis of the intrahepatic or hilar bile duct was detected only in AIP (P = 0.001). On MRCP, diffuse narrowing of the MPD on ERCP was shown as a skipped non-visualized lesion in 50% and faint visualization in 19%, but segmental narrowing of the MPD was visualized faintly in only 14%. CONCLUSION: Several ERCP findings are useful for differentiating AIP from PC. Although MRCP cannot replace ERCP for the diagnostic evaluation of AIP, some MRCP findings support the diagnosis of AIP.

Comparison between Hypertonic Saline with Dextran and Mannitol on Vasodilatation of Encephalic Vessels Using a Magnetic Resonance Imaging in the Dogs

This study aimed to investigate whether a small volume of 7.2% hypertonic saline solution with 6% dextran 70 (HSD) is superior to mannitol in vasodilatation of encephalic vessels in the dogs using magnetic resonance imaging (MRI). Fifteen healthy 2.4 ± 0.9 year-old purpose-bred male Beagle dogs were assigned to receive 5 mL/kg of isotonic saline solution (ISS) as control, 20% mannitol or HSD infusion at a flow rate of 20 mL/kg/hours via right cephalic *** blood samples were collected immediately before fluid infusion (pre) and every 15 minutes until 120 minutes after the initiation of fluid infusion. Immediately after collection of each blood sample, T1 and T2-weighted magnetic resonance imaging recordings were undergone. Immediately after HSD infusion, the area of the cross-section of superior sagittal sinus was significantly greater than that of beagles in the other groups (p p < 0.001). Our results indicate that HSD induced a rapid and strong reduction in the area of the cross-section of CSF more than mannitol did. Therefore, it is suggested that 5 mL/kg of HSD might be superior to isovolume of mannitol in inducing vasodilatation in the dog.
