In view of the demographic changes and projected increase of arthroplasty procedures worldwide,the number of prosthetic joint infection cases will naturally ***,in order to counteract this trend more rigid rules and a stricter implementation of effective preventive strategies is of highest *** the absence of a"miracle weapon"priorities should lie in evidence-based measures including preoperative optimization of patients at higher infection risks,the fulfilment of strict hygiene rules in the operating theatre and an effective antibiotic prophylaxis *** of a"one size fits all"philosophy,it has been proposed to adjust the antibiotic prophylaxis protocol to major infection risks taking into account important patient-and procedure-related risk factors.A stronger focus on the local application mode via use of high dose dual antibioticloaded bone cement in such risk situations may have its advantages and is easy to apply in the *** more potent antimicrobial growth inhibition in vitro and the strong reduction of the prosthetic joint infection rate in risk for infection patients with aid of dual antibiotic-loaded bone cement in clinical studies align with this hypothesis.

Cell-type specific regulation of MARCH1 E3 ubiquitin ligase by the anti-inflammatory cytokine IL-10

Membrane-associated RING-CH-1 (MARCH1) is an E3 ubiquitin ligase which targets MHC-II, CD86 and various other molecules for degradation. It is one of the most efficient post-translational regulators of antigen presentation. MARCH1 is expressed in resting immature dendritic cells and B cells but can be induced in other cell types. While activation of most immune cells results in a reduction in MARCH1 gene expression, its anti-inflammatory properties are highlighted by its induction in response to IL-10. Here, we review what is known about the regulation of MARCH1 gene expression in response to IL-10 by various immune cells. We speculate on the role of MARCH1 ininfection, its differential expression pattern and the promise that this E3 ubiquitin ligase holds to influence immune responses and mitigate immune pathologies.

Marine reserves are an important management tool for conserving local biodiversity and protecting fragile ecosystems such as seagrass that provide significant ecological functions and services to people and the marine *** humans placing ever-growing pressure on seagrass habitats,marine reserves also provide an important reference from which changes to seagrass and their ecological assemblages may be *** eight years of protection of seagrass beds(Posidonia australis)in no-take marine reserves(Sanctuary Zones)within the Jervis Bay Marine Park(New South Wales,Australia;zoned in 2002),we aimed to assess what changes may have occurred and assess continuing change through time in fish assemblages within these seagrass *** baited remote underwater videos(BRUVs),we sampled seagrass fish assemblages at three locations in no-take zones and five locations in fished zones three times from 2010 to ***,we observed a total of 2615 individuals from 40 fish species drawn from 24 *** detected no differences in total fish abundance,diversity,or assemblage composition between management zones,although we observed a significant increase in Haletta semifasciata,a locally targeted fish species,in no-take marine reserves compared with fished *** assemblages in seagrass varied greatly amongst times and *** species varied in relative abundance greatly over months and years,whilst others had consistently greater relative abundances at specific *** discuss the potential utility of marine reserves covering seagrass habitats and the value of baseline data from which future changes to seagrass fish populations may be measured.

This study presents a comprehensive impact analysis of the rotor angle stability of a proposed international connection between the Philippines and Sabah,Malaysia,as part of the Association of Southeast Asian Nations(ASEAN)Power *** study focuses on modeling and evaluating the dynamic performance of the interconnected system,considering the high penetration of renewable *** flow,small signal stability,and transient stability analyses were conducted to assess the ability of the proposed linked power system models to withstand small and large disturbances,utilizing the Power Systems Analysis Toolbox(PSAT)software in *** components used in the model are documented in the PSAT ***,there is a lack of publicly available studies regarding the implementation of this specific ***,the study investigates the behavior of a system with a high penetration of renewable energy *** on the findings,this study concludes that a system is generally stable when interconnection is realized,given its appropriate location and dynamic component ***,the critical eigenvalues of the system also exhibited improvement as the renewable energy sources were augmented.

Operationalizing physical literacy through sport education

Physical literacy （PL）, as embodied within physical education （PE）, has been vaunted as having increasing importance as a disposition for students of all abilities to establish lifelong adherence to physical activity. The purpose of this paper was to provide a discussion of how the pedagogical features of a contemporary pedagogical model, Sport Education （SE）, may be used to operationalize PL in PE and what empirical evidence currently exists to validate this claim. Substantial empirical evidence exists that the attributes associated with the development of PL （Whitehead, 2010） can be operationalized in PE with the effective implementation of the model. SE has distinct pedagogical features which positively contribute to many of the dimensions of PL and can further an individuals＇ journey towards greater PL and having an embodied self within PE. That stated, there remains concern that the context for this embodiment remains too narrow to be viewed as a panacea for the development of lifelong physical activity. SE must be developed as a connective specialism if these PL attributes are to transform the motivation and confidence for individuals to capitalize on their innate physical potential and make a more significant contribution to the quality of life.

Cell signalling pathways underlying induced pluripotent stem cell reprogramming

Induced pluripotent stem(i PS) cells, somatic cells reprogrammed to the pluripotent state by forced expression of defined factors, represent a uniquely valuable resource for research and regenerative medicine. However, this methodology remains inefficient due to incomplete mechanistic understanding of the reprogramming process. In recent years, various groups have endeavoured to interrogate the cell signalling that governs the reprogramming process, including LIF/STAT3, BMP, PI3 K, FGF2, Wnt, TGFβ and MAPK pathways, with the aim of increasing our understanding and identifying new mechanisms of improving safety, reproducibility and efficiency. This has led to a unified model of reprogramming that consists of 3 stages: initiation, maturation and stabilisation. Initiation of reprogramming occurs in almost all cells that receive the reprogramming transgenes; most commonly Oct4, Sox2, Klf4 and c Myc, and involves a phenotypic mesenchymal-to-epithelial transition. The initiation stage is also characterised by increased proliferation and a metabolic switch from oxidative phosphorylation to glycolysis. The maturation stage is considered the major bottleneck within the process, resulting in very few "stabilisation competent" cells progressing to the final stabilisation phase. To reach this stage in both mouse and human cells, pre-i PS cells must activate endogenous expression of the core circuitry of pluripotency, comprising Oct4, Sox2, and Nanog, and thus reach a state of transgene independence. By the stabilisation stage, i PS cells generally use the same signalling networks that govern pluripotency in embryonic stem cells. These pathways differ between mouse and human cells although recent work has demonstrated that this is context dependent. As i PS cell generation technologies move forward, tools are being developed to interrogate the process in more detail, thus allowing a greater understanding of this intriguing biological phenomenon.

Clinical significance of prostate 18F-labelled fluorodeoxyglucose uptake on positron emission tomography/computed tomography:A five-year review

AIM To determine the significance and need for investigation of incidental prostatic uptake in men undergoing ^(18)F-labelled fluorodeoxyglucose(^(18)F-FDG) positron emission tomography/computed tomography(PET/CT) for other *** Hospital databases were searched over a 5-year period for patients undergoing both PET/CT and prostate magnetic resonance imaging(MRI). For the initial analysis, the prostate was divided into six sectors and suspicious or malignant sectors were identified using MRI and histopathology reports respectively. Maximum and mean ^(18)F-FDG standardised uptake values were measured in each sector by an investigator blinded to the MRI and histopathology findings. Two agematched controls were selected per case. Results were analysed using a paired t-test and one-way ANOVA. For the second analysis, PET/CT reports were searched for prostatic uptake reported incidentally and these patients were followed up. RESULTS Over a 5-year period, 15 patients underwent both PET/CT and MRI and had biopsy-proven prostate *** prostatic sectors had a trend to higher ^(18)F-FDG uptake than benign sectors, however this was neither clinically nor statistically significant(3.13 ±0.58 vs 2.86 ± 0.68, P > 0.05). ^(18)F-FDG uptake showed no correlation with the presence or histopathological grade of tumour. ^(18)F-FDG uptake in cases with prostate cancer was comparable to that from age-matched controls. Forty-six(1.6%) of 2846 PET/CTs over a 5-year period reported incidental prostatic uptake. Of these, 18(0.6%) were investigated by PSA, 9(0.3%)were referred to urology, with 3(0.1%) undergoing MRI and/or biopsy. No cases of prostate cancer were diagnosed in patients with incidental ^(18)F-FDG uptake in our institute over a 5-year *** ^(18)F-FDG uptake overlaps significantly between malignant and benign prostatic conditions.
