Donor preoperative oxygen delivery and post-extubation hypoxia impact donation after circulatory death hypoxic cholangiopathy

AIM: To evaluate donation after circulatory death (DCD) orthotopic liver transplant outcomes [hypoxic cholangiopathy (HC) and patient/graft survival] and donor ***: From 2003-2013, 45 DCD donor transplants were performed. Predonation physiologic data from UNOS DonorNet included preoperative systolic and diastolic blood pressure, heart rate, pH, SpO2, PaO2, FiO2, and hemoglobin. Mean arterial blood pressure was computed from the systolic and diastolic blood pressures. Donor preoperative arterial O2 content was computed as [hemoglobin (gm/dl) × 1.37 (ml O2/gm) × SpO2%) + (0.003 × PaO2)]. The amount of preoperative donor red blood cell transfusions given and vasopressor use during the intensive care unit stay were documented. Donors who were transfused ≥ 1 unit of red-cells or received ≥ 2 vasopressors in the preoperative period were categorized as the red-cell/multi-pressor group. Following withdrawal of life support, donor ischemia time was computed as the number-of-minutes from onset of diastolic blood pressure l aortic cross clamping. Donor hypoxemia time was the number-of-minutes from onset of pulse oximetry l clamping. Donor hypoxia score was (ischemia time + hypoxemia time) ÷ donor preoperative ***: The 1, 3, and 5 year graft and patient survival rates were 83%, 77%, 60%; and 92%, 84%, and 72%, respectively. HC occurred in 49% with 16% requiring retransplant. HC occurred in donors with increased age (33.0 ± 10.6 years vs 25.6 ± 8.4 years, P = 0.014), less preoperative multiple vasopressors or red-cell transfusion (9.5% vs 54.6%, P = 0.002), lower preoperative hemoglobin (10.7 ± 2.2 gm/dl vs 12.3 ± 2.1 gm/dl, P = 0.017), lower preoperative arterial oxygen content (14.8 ± 2.8 ml O2/100 ml blood vs 16.8 ± 3.3 m

Using proteomics to compare the molecular structures of sulfide and permeate-depilated sheepskins

An environmentally friendly method using real or artificial bovine milk permeate to both depilate and preserve sheepskins has been reported which completely and cleanly removed the wool from the hair follicle and had no detrimental effects on the skin.A proteomic analysis,assessing the relative abundance of proteins in matched permeate-depilated and chemically depilated(sulfide)sheepskins,showed variations in the levels of specific collagen types in the skin's basement membrane and other proteins associated with the *** findings were corroborated by biochemical analyses of matched permeate depilated and raw skin samples,and provide clues to the mechanism of non-invasiveand complete *** also support the observation that permeate-depilated skins were smoother than their sulfide-depilated counterparts and resulted in leather with a superior surface.

Monitoring the effects of land transformation on African clawless otters(Aonyx capensis)using fecal glucocorticoid metabolite concentrations as a measure of stress

In a time of increasing environmental change caused by anthropogenic disturbance,there is a greater need to understand animal adaptations to manmade *** this regard,the measurement of stress-related endocrine markers provides a useful tool to examine the impact of environmental challenges and the physiological consequences for wildlife occupying such *** aims of the present study were to validate fecal glucocorticoid metabolite(fGCM)concentrations as a measure of stress using samples from a male African clawless otter(Aonyx capensis;n=1)and to compare fGCM concentrations of otters occurring in a transformed and in 2 natural areas in South *** the 5 different enzyme-immunoassays(EIA)tested,a cortisol and oxoetiocholanolone(measuring 11,17 dioxoandrostanes)EIA revealed the highest response(74%and 48%increase,respectively)30 and 24 hours after a stress event(translocation of a captive individual as part of its rehabilitation prior to release),respec*** both EIAs,fGCM concentrations were comparable for samples collected up to 3 h *** the cortisol EIA for subsequent analyses,fGCM concentrations of animals from the transformed area(n=20;mean[±SD]:0.468[±0.539]µg/g dry weight[DW])were significantly higher(P=0.013)than those from otters in the natural areas(n=17;0.242[±0.226]µg/g DW).These preliminary results suggest that African clawless otters may have increased adrenocortical activity that could be due to conditions linked to living in a transformed environment.

Plant glycosylphosphatidylinositol anchored proteins at the plasma membrane-cell wall nexus

Approximately 1% of plant proteins are predicted to be post-translationally modified with a glycosylphospha- tidylinositol （GPI） anchor that tethers the polypeptide to the outer leaflet of the plasma membrane. Whereas the synthesis and structure of GPI anchors is largely conserved across eukaryotes, the repertoire of functional domains present in the GPl-anchored proteome has diverged substantially. In plants, this includes a large fraction of the GPl-anchored proteome being further modified with plant-specific arabinogalactan （AG） O-glycans. The impor- tance of the GPl-anchored proteome to plant development is underscored by the fact that GPI biosynthetic null mutants exhibit embryo lethality. Mutations in genes encoding specific GPl-anchored proteins （GAPs） further supports their contribution to diverse biological processes, occurring at the interface of the plasma membrane and cell wall, including signaling, cell wall metabolism, cell wall polymer cross-linking, and plasmodesmatal transport. Here, we review the literature concerning plant GPl-anchored proteins, in the context of their potential to act as molecular hubs that mediate interactions between the plasma membrane and the cell wall, and their potential to transduce the signal into the protoplast and, thereby, activate signal transduction pathways.

The problems associated with the subjective assessment of human sperm morphology have been well aired in another Asian Journal of Andrology Special Issue1 that marked the publication of the 5th edition of the WHO Semen analysis manual, and contrary views have subsequently been presented） However, the vagaries of the eye-brain system in assessing whether a sperm head is large or small can be eliminated by objective assessment where definitive structures are defined by their dimensions. These can then be classified automatically into as many categories as the data permit, conventionally on the basis of preset upper and lower limits, but also by more comprehensive analysis as discussed here.

Maturation of sperm volume regulation in the rat epididymis

Sperm maturation in the epididymis may involve differences between mature and immature spermatozoa in their volume regulatory osmolyte response. Spermatozoa obtained from the rat caput and cauda epididymidis were examined for their ability to regulate volume after transfer from in situ epididymal osmolality （measured to be 343 ± 13 and 365 ± 19 mmol kg^-1, respectively） to that of the female tract in single- and multiple-step protocols. Cells withstood the single-step treatment better than the multistep protocol. Sperm volume estimates by flow cytometric measure- ments of forward scatter of cells with intact head membranes was more sensitive than those by assessing cell coiling microscopically. At osmolalites below 210 mmol kg l both caput and cauda cells ruptured, limiting the use of flow cytometry. Above this critical value, the use of quinine showed that both caput and cauda cells could regulate volume, but cauda cells were the more effective. Of several organic osmolytes studied, myo-inositol, glutamate and KCl caused only temporary and slight swelling of spermatozoa cells in hypotonic medium. Spermatozoa of both maturities seemed to use potassium as the preferred osmolyte for regulating volume.

Cloning,Sequencing and Characterization of 3-Hydroxybutyrate Dehydrogenase Encoding Gene(bdh A)in Bradyrhizobium japonicum USDA110 Strain

The current study describes the molecular characterization of a clone which can restore the ability of bdh A mutant strains NGRPA2 and Rm11107 to utilize 3-hydroxybutyrate as a sole carbon source (Hbu+ ). This clone was screened out by complementation experiment from Bradyrhizobium japonicum US-DAI 10 genomic library, and the presence of bdhA gene in the clone was verified by Bdh assay and Southern blot analysis. Furthermore, the entire sequence of bdhA. gene was sequenced and the sequence was deposited in GenBank database under the accession number AY077581. bdhA gene comprises 789 base pairs and encodes Bdh with 262 amino acid of MW 27.59 kDa. Interposon JlKm was inserted into the bdhA ORF at EcoR I site and the bdhA mutant was constructed in B .japonicum by homologous recombination. Plant assay result did not show obvious effects of mutation of bdhA gene on nodulation and nitrogen-fixation.

Search of inhibitors of aldose reductase for treatment of diabetic cataracts using machine learning

Purpose:Patients with diabetes mellitus have an elevated chance of developing cataracts,a degenerative visionimpairing condition often needing *** process of the reduction of glucose to sorbitol in the lens of the human eye that causes cataracts is managed by the Aldose Reductase Enzyme(AR),and it is been found that AR inhibitors may mitigate the onset of diabetic *** exists a large pool of natural and synthetic AR inhibitors that can prevent diabetic complications,and the development of a machine-learning(Ml)prediction model may bring new AR inhibitors with better characteristics into clinical ***:Using known AR inhibitors and their chemical-physical descriptors we created the Ml model for prediction of new AR *** predicted inhibitors were tested by computational docking to the binding site of ***:Using cross-validation in order to find the most accurate Ml model,we ended with final cross-validation accuracy of 90%.Computational docking testing of the predicted inhibitors gave a high level of correlation between the Ml prediction score and binding free ***:Currently known AR inhibitors are not used yet for patients for several *** think that new predicted AR inhibitors have the potential to possess more favorable characteristics to be successfully implemented after clinical *** new inhibitors can improve patient well-being and lower surgical complications all while decreasing long-term medical expenses.
