Synergistic Effects of Glutamine Deprivation and Metformin in Acute Myeloid Leukemia

Objective The metabolic reprogramming of acute myeloid leukemia(AML)cells is a compensatory adaptation to meet energy requirements for rapid *** study aimed to examine the synergistic effects of glutamine deprivation and metformin exposure on AML *** SKM-1 cells(an AML cell line)were subjected to glutamine deprivation and/or treatment with metformin or bis-2-(5-phenylacetamido-1,2,4-thiadiazol-2-yl)ethyl sulfide(BPTES,a glutaminase inhibitor)or *** viability was detected by Cell Counting Kit-8(CCK-8)assay,and cell apoptosis and reactive oxygen species(ROS)by flow *** blotting was conducted to examine the levels of apoptotic proteins,including cleaved caspase-3 and poly(ADP-ribose)polymerase(PARP).Moreover,the human long noncoding RNA(lncRNA)microarray was used to analyze gene expression after glutamine deprivation,and results were confirmed with quantitative RT-PCR(qRT-PCR).The expression of metallothionein 2A(MT2A)was suppressed using *** growth and apoptosis were further detected by CCK-8 assay and flow cytometry,respectively,in cells with MT2A *** Glutamine deprivation or treatment with BPTES inhibited cell growth and induced apoptosis in SKM-1 *** lncRNA microarray result showed that the expression of MT family genes was significantly upregulated after glutamine ***2A knockdown increased apoptosis,while proliferation was not affected in SKM-1 *** addition,metformin inhibited cell growth and induced apoptosis in SKM-1 *** glutamine deprivation and metformin enhanced the sensitivity of SKM-1 cells to ***,the combination of glutamine deprivation with metformin exhibited synergistic antileukemia effects on SKM-1 *** Targeting glutamine metabolism in combination with metformin is a promising new therapeutic strategy for AML.

Dear Editor,Tracking control in networked environment is a very challenging problem due to the contradiction of rapid response to the time-varying signal and the inevitable delay introduced by networks. This letter has proposed several fuzzy-inverse-model-based network tracking control frameworks which are helpful in handling the system with nonlinear dynamics and uncertainties.

Rotational Doppler effect of composite vortex beams with tailored OAM spectra

There recently has been increasing interest in the research and application of the rotational Doppler effect(RDE),which paves a promising way to detect rotating objects *** order to obtain more information about the rotating object from the rotational Doppler signal,composite vortex beams by coaxial superposition of orbital angular momentum(OAM)modes are often used as the probe ***,to the best of our knowledge,the RDE of composite vortex beams with arbitrary OAM spectra has not yet been comprehensively *** this paper,the correspondence between the OAM spectrum of a probe beam and the frequency spectrum of a rotational Doppler signal is theoretically *** is explicitly revealed that the RDE frequency spectrum of scattered light is related to the product of two autocorrelation functions:one from the OAM spectrum of probe beam and the other from the spiral spectrum of rotating *** the basis of this relation,one can regulate the RDE frequency spectrum on demand via tailoring the OAM spectrum of the probe *** a proof of concept we design a special composite vortex beam to eliminate the broadening of the RDE spectrum induced by *** findings are of practical value in applications such as remote sensing and optical metrology.

Highly Red Emissive Conjugated Homopolymers Based on Double B←N Bridged Bipyridine Unit

Conjugated homopolymers based on six-member rings,e.g.,polyfluorene,always exhibit blue emission and conjugated homopolymers based on five-member rings,e.g.,polythiophene,can give red emission with low *** this work,we report a series of new conjugated homopolymers based on six-member rings with high-efficiency deep-red *** repeating units of the red light emitting homopolymers are double B←N bridged bipyridine(BNBP)with the boron atoms functionalized with diphenyl,borafluorene,and 2,7-di-tert-butyl-borafluorene groups,*** relationship between the chemical structures and the opto-electronic properties of the monomers and the homopolymers has been systematically *** three polymers emit pure red light(λ_(max)=656 nm)or deep red light(λ_(max)=693 nm)with fluorescence quantum efficiency in solution higher than 60%.The polymers can be used as the emitters in solution-processed organic light-emitting diodes with red emission and decent device *** work indicates a new strategy to design highly efficient light emitting conjugated polymers.

Upconversion luminescence and temperature sensing performance of Er^(3+) ions doped self-activated KYb(MoO_(4))_(2) phosphors

In this work,a series of self-activated KYb(MoO_(4))_(2) phosphors with various x at% Er^(3+) doping concentrations(x=0.5,1,3,5,8,10,15) was synthesized by the solid-state reaction *** phase structure of the as-prepared samples was analyzed by X-ray diffraction(XRD),XRD Rietveld refinement and Fourier transform infrared(FT-IR) *** as-prepared samples retain the orthorhombic structure with space group of Pbcn even Er^(3+) doping concentration up to 15 at%.High-purity upconversion(UC) green emission with green to red intensity ratio of 55 is observed from the as-prepared samples upon the excitation of 980 nm semiconductor laser and the optimum doping concentration of Er^(3+) ions in the self-activated KYb(MoO_(4))_(2) host is revealed as 3 at%.The strong green UC emission is confirmed as a two-photon process based on the power-dependent UC *** addition,the fluorescence intensity ratios(FIRs) of the two thermally-coupled energy levels,namely ^(2)H_(11/2) and ^(4)S_(3/2).of Er^(3+) ions were investigated in the temperature region 300-570 K to evaluate the optical temperature sensor behavior of the *** maximum relative sensitivity(S_(R)) is determined to be 0.0069 K^(-1) at300 K and the absolute sensitivity(S_(A)) is determined to be 0.0126 K^(-1) at 300 *** S_(A) of self-activated KYb(MoO_(4))2:Er^(3+)is almost twice that of traditional KY(MoO_(4))2:Er^(3+)/Yb^(3+)codoping *** results demonstrate that Er^(3+) ions doped self-activated KYb(MoO_(4))2 phosphor has promising application in visible display,trademark security and optical temperature sensors.

The Shanghai high-repetition-rate X-ray free-electron laser and extreme light facility(SHINE)operates at a maximum repetition rate of 1 *** magnets are key components that distribute electron bunches into three different undulator lines in a bunch-by-bunch *** kicker field width must be less than the time interval between bunches.A lumpedinductance kicker prototype was developed using a vacuum chamber with a single-turn *** full magnetic field strength was 0.005 *** paper presents the requirements,design considerations,design parameters,magnetic field calculations,and measurements of the kicker *** relevant experimental results are also *** pulse width of the magnetic field was approximately 600 ns,and the maximum operation repetition rate was 1 *** developed kicker satisfies the requirements for the SHINE ***,numerous recommendations for the future optimization of kicker magnets are provided.

The recurrence of head and neck squamous cell carcinoma(HNSCC)after surgical resection continues to pose a major challenge to cancer *** HNSCC exhibits a low response rate to immune checkpoint blockade(ICB),while photothermal therapy(PTT)can increase the infiltration of immune cells to make tumors more susceptible to cancer *** this regard,we designed and constructed a novel multifunctional nanocomposite comprised of oxidized bacterial cellulose(OBC),thrombin(TB),and gold nanocages(AuNCs)containing anti-programmed death 1(PD-1)antibody(αPD-1@AuNCs),which allows the combination of therapies with remarkable postoperative antitumor immunity to control local tumor ***αPD-1@AuNCs displayed high light-to-heat conversion efficiency and induced pyroptosis under near infrared(NIR)irradiation,which activated a potent antitumor immune *** importantly,the therapeutic system could induce tumor pyroptosis and enhance antitumor immune response by increasing T-cell infiltration and reducing the immune suppressive cells,when combined with local ICB therapy,which effectively avoided the tumor recurrence in a HNSCC postoperative mice ***,the newly developed multifunctional nanocomposites could be a promising candidate for the treatment of postoperative HNSCC.

Lipid and sugar homeostasis is critical for insect development and *** this study,we characterized an acetyl coenzyme A carboxylase gene in Blattella germanica(BgACC)that is involved in both lipogenesis and sugar *** found that BgACC was dominantly expressed in the fat body and integument,and was significantly upregulated after *** of BgACC in 5th-instar nymphs did not affect their normal molting to the next nymphal stage,but it caused a lethal phenotype during adult ***-RNA interference(RNAi)significantly downregulated total free fatty acid(FFA)and triacylglycerol(TAG)levels,and also caused a significant decrease of cuticular hydrocarbons(CHCs).Repression of BgACC in adult females affected the development of oocytes and resulted in sterile females,but BgACC-RNAi did not affect the reproductive ability of ***,knockdown of BgACC also changed the expression of insulin-like peptide genes(BglLPs),which mimicked a physiological state of high sugar *** addition,BgACC was upregulated when *** were fed on a high sucrose diet,and repression of BgACC upregulated the expression of the glycogen synthase gene(BgGlyS).Moreover,BgACC-RNAi increased the circulating sugar levels and glycogen storage,and a longevity assay suggested that BgACC was important for the survival of *** under conditions of high sucrose *** results confirm that BgACC is involved in multiple lipid biogenesis and sugar homeostasis processes,which further modulates insect reproduction and sugar *** study benefits our understanding of the crosstalk between lipid and sugar metabolism.
